  • Utjecaj plinovitog ozona na redukciju Escherichia coli biofilma
    Kovač, Bruno ...
Escherichia coli is a commonly known bacterium that is part of the normal microbiota, but certain strains can cause urinary and intestinal infections and lead to very severe forms of the disease with ... a possible fatal outcome. It has the ability to create biofilms, which makes it more resistant to disinfectants and requires more than one method of disinfection for its eradication. Gaseous ozone proved to be an appropriate disinfectant because it possesses good antimicrobial activity and strong oxidative properties. The aim of this study was to investigate the effectiveness of ozone on reducing the number of bacteria and total biomass in E. coli biofilm on four clinical strains and one standard strain. The 24-hour-old biofilm E. coli created on ceramic tiles, with dimensions of 1 cm x 1 cm, is exposed to a concentration of 25 ppm of gaseous ozone for one hour in a closed box model. For each strain, before and after ozone treatment, the total number of bacte ria expressed as CFU/cm2 was determined, measured biological activity expressed as RLU (Relative Light Units) and conducted crystal violet staining to determine the total biomass. Ozone treatment achieved a reduction in the total biomass of E. coli by up to 61.40%, as well as a reduction in the total number of bacteria by up to 99.41%. Biomass reduction determined by the ATP bioluminescence method was up to 90.31%. In conclusion, ozone at a concentration of 25 ppm for an hour of exposure was not sufficient for the complete removal of the biofilm created, and it is definitely recommended to combine it with other mechanical and chemical methods of disinfection or observe its effectiveness through a prolonged time of action or at a concentration greater than 25 ppm.
