Uporaba nanodelcev za odstranjevanje biofilmov bakterij Listeria innocua = Use of nanoparticles for removal of Listeria innocua biofilms : Master's Study Programme Industrial PharmacyHajdarović, ElenaBakterije rodu Listeria tvorijo biofilm, ki jih varuje pred škodljivimi vplivi okolice, zato so znotraj biofilmov bakterije bolj odporne na zunanje vplive kot planktonske bakterije. Magistrska naloga ... je vsebinsko vezana na projekt Javne agencije za raziskovalno dejavnost Republike Slovenije (ARRS) "Baktericidna nanorezila: preizkus bimodalnega mehanokemijskega odstranjevanja trdovratnih biofilmov". Cilj naloge je bil testirati metodo za odstranjevanje biofilma bakterije Listeria innocua z uporabo treh različnih nanorezil po inkubaciji v magnetnem polju. Testirali smo delovanje nanorezil na trikrat spran biofilm bakterije Listeria innocua seva ŽM39, ki izraža fluorescenčni protein DsRedExpress. Analizirali smo fluorescenco in število živih bakterij v biofilmu po vsakem spiranju. Preverili smo uporabnost barvil za spremljanje vpliva nanorezil na biofilm divjega tipa bakterije Listeria innocua seva ŽM39. Po izbiri najbolj primernega barvila smo spremljali vpliv nanorezil na trikrat spran biofilm bakterije Listeria innocua ŽM39 ob dodatku tega barvila. Testirali smo tudi fluorescenco nanorezil pri različih valovnih dolžinah. Preverili smo rast bakterij Listeria innocua in Listeria monocytogenes ATCC 19115. Na koncu smo še analizirali vsa barvila na intaktnem in trikrat spranem biofilmu bakterij. Ugotovili smo, da nobeno izmed uporabljenih nanorezil ni bilo uspešno pri odstranjevanju biofilma baterije Listerie innocua seva ŽM39. Kot najbolj uporabno barvilo se je pokazalo barvilo 9-(dietilamino)-3-((1-(2-(metil(tetradeil)amino)etil)-1H-1,2,3-triazol-4-il)metoksi)-5H-benzo[a]fenoksazin-5-on (barvilo 10*), ker ima najmanjšo intenziteto fluorescence slepega vzorca. Pri spremljanju delovanja nanorezil na trikrat spran biofilm bakterije Listeria innocua seva ŽM39 ob dodatku barvila 10* smo dobili podobne vrednosti pri vseh uporabljenih nanorezilih, vendar so se kot najbolj učinkovita izkazala nanorezila Hex. Ugotovili smo da imajo nanorezila najmočnejšo fluorescencno pri valovni dolžini ekscitacije 563 nm in emisije 581 nm. Rezultati analize vseh barvil so pokazali, da ima barvilo (5-(4-(((9-(dietilamino)-5-okso-5H-benzo[a]fenoksazin-3-il)oksi)metil)-1H-1,2,3-triazol-1-il)pentil)trifenilfosfonium bromide (barvilo 7*) najbolj negativni vpliv na rast bakterij tako na intaktnem biofilmu kot na spranem. Pokazali smo, da se lahko za sledenje sevom listerij pri analizi vpliva nanorezil na biofilm bakterij uporabljajo fluorescenčna barvila ali reporterski proteini.Type of material - master's thesis ; adult, seriousPublication and manufacture - Ljubljana : [E.
