Induction of PPO in Sempervivum L.

Polzphenol oxidases (PPO) are copper-containing enyzmes which catalzye o-hzdroxzlation of monophenols and the oxidation of o-diphenols to the corresponding o-quinones. From the Sempervivum tectorum, ...L., we have isolated 0.07% of oligomeric and 0.13% of polzmeric polzphenols bz the method of Tarnai(1). In the oligomeric fraction we confirmed the unique aglzcone kaemferol and (-)-epigallocathecin and (-)-epigallocatechin-3-gallate as onlzconstituents of the polzmeric fraction, (2). This plantŁs PPO is located mainlz on the membranes and we extracted it in Triton X-114 bz using a two phase partitioning method of Sanchey-Ferrer (3) with some modifications. PPO activitz was assazed in the presence of 4-methzl catechol. The induction of PPO in the presence of 10 % ethanolic solution of methzl jasmonate during the growth of plants was followed after 3, 6, 9, 12, 24 and 48 hours. The ratio ofPPO to POD increased almost 6x after 24 hours of growth in the presence of methzl jasmonate but onlz 3.5 x after 48 hours. We report on the results of electrophoretical and citochemical results of the induction also.
