    Since the introduction of chemotherapy as first choice of treatment in small round cell tumors, which include rhabdomyosarcoma (RMS), fine needle aspiration biopsy (FNAB) aquired an important role in ... providing the morphologic diagnosis. Therefore, the aim of our study was to analise light-microscopic and image-cytometric characteristics from cytologic specimens of RMS in order to improve their recognition from FNAB smears. In the first part of our study we analysed morphological features and architectural patterns in FNAB specimens of 58 patients, including children and adults. Classification of RMS was possible from surgical biopsies of 48 patients. In the second part we analysed the results of immunocytochemical staining for desmin in 71 FNAB samples from 50 patients, using the ABC method.We compared the quality of staining on fresh, routinely prepared slidesvs archival material, the quality of staining on smears vs cytospins andthe ability of two antidesmin antibodies to recognise desmin. Furthermore, we investigated the percentage of desmin positive cells in individual FNAB smears. In the last part of our study we analysed nuclear features in Feulgen stained slides, prepared from archival tissue sections of RMS from 42 patients. We investigated the ability of individual nuclear features to discriminate between hystological subtypes of RMS. Furthermore, we tried to find a correlation between DNA ploidy and other prognostic factors of RMS, as well as a correlation between DNA ploidy and patient survival. On the basis ofmorphological features and architectural patterns of RMS we were able to differentiate between embrional and alveolar RMS from FNAB smears in approximately 80% of cases. The characteristic features in smears of embryonalRMS were large tissue fragments, containing cells and abundant eosinophilic material (Giemsa), accompanied by cells with oval muclei or rhabdomyoblasts in various stages of maturation.
