  • Učinek električnih impulzov in pregrevanja na krvni pretok in rast mišjega tumorja in vivo : [magistrska naloga]
    Karner, Katarina Barbara
Purpose: To determine whether application of electric pulses increases hyperthermic cytotoxicity electric pulses were combined with hyperthermia. Materials and methods: The effects of short intense ... electric pulses, hyperthermia and combination of the two modalities on the vascular perfusion and growth of a LPB fibrosarcoma syngenic to C557BI mouse were studied. Electric pulses (1300 Vžcm, 100 micro s, 1 Hz) by percutaneously placed electrodes and the local microwave hyperthermia (915 Mhz) were applied to the tumours. The antitumour effectiveness of heat treatment at 40.5, 42.5 and 43.5degree C. for 30 minutes, electric pulses as a single modality and the combination of electric pulses and heating was determined by tumour growth delay and tumour cures as well as extent of tumour necrosis. Tumour blood flowmodifying effect of therapy was evaluated by Patent blue staining technique. ResultsČ Heat treatments at 42.5 degree C and 43.5 degree C for 30 minutes caused a significant increase in tumour growth time when given individually, hyperthermia at 43.5 degree C additionally resulted in 6% tumourcure. Application of electric pulses resulted in minor tumour growth delay, but was followed by substantial reduction in tumour blood flow within 30 minutes returning to control level in next 24 hours. The application of electric pulses followed by immediate heating at 43.5 degree C for 30 minutes caused a marked tumour growth delay (14.5 +- 3.1 days) and high tumour controlrate (43%). It resulted in complete shut down of tumour blood flow lasting for at least 24 hours. The results on tumour blood flow reduction correlated well with the good antitumour effectiveness of the combination of electric pulses and hyperthermia following the electric pulses immediately andwith the extent of the necrosis in the tumours.
