Inhaled corticosteroids (ICS) are commonly used in asthma treatment ... since they suppress airway inflammation. However, the response to ICS treatment is highly variable. Vascular endothelial growth factor A (VEGFA) is a major regulator of angiogenesis and vascular permeability and has been shown to be elevated in asthma patients. Objectives. The aim of this study was to investigate the involvement of variants in the VEGFA gene in treatment response to ICS in adult asthmatics. Methods. A cohort of 208 adult asthmatic patients treated with ICS were genotyped for two variants, rs2146323 and rs833058, in the VEGFA gene. The response was analysed after shortterm treatment (change in % predicted FEV1) and long-term treatment (changes in % predicted FEV1, Asthma Control Test (ACT), Asthma Quality of Life Questionnaire (AQLQ), bronchial hyperresponsiveness (BHR)). Results. Patients with the CC+AC genotype in variant rs2146323 show higher improvement in % predicted FEV1 after three months and after at least three years of treatment, compared to the AA genotype. In variant rs833058, the TT+TC genotype shows better treatment response to ICS, compared to the CC genotype, according to ACT, AQLQ, and BHR. Conclusion. Variants rs2146323 and rs833058 in VEGFA are associated with treatment response to ICS after short-term and long-term treatment, assessed as changes in % predicted FEV1, ACT, AQLQ scores, and BHR.
