The pore-forming protein equinatoxin II (EqtII) from Actinia equina, and five single cysteine EqtII mutants (K20C, K77C, R126C, R144C, A179C), were evaluated for the preparation of specific ...immunotoxins (IT). They were linked to a monoclonal antibody (mAb) specific for tumor cells via the high affinity avidin-biotin interaction. Three human tumor cell lines (malignant melanoma, breast adenocarcinoma, and ovary adenocarcinoma) and one control line (chinesehamster ovary cells) were tested. The ITs displayed some specific activity when mAbs directed against tumor antigens were used. However, unspecific activity remained high, being still around 30% of the specific activity in the best case.
