  • Koncentracija serumskega trombopoetina med zdravljenjem bolnikov z akutno mieloblastno levkemijo s citostatiki = Serum thrombopoietin levels in patients with acute myeloid leukemia diring chemotherapy

    To understand the regulation of circulating platelet counts (PC) with thrombopoietin (TPO) serum TPO levels and PC before and after myelosuppressivechemotherapy and after the administration of ... platelet transfusions in 10 patients with acute myeloid leukaemia (AML) were studied. Serum TPO levels were measured by the quantitative sandwich enzyme-linked immunosorbent assay (Quantikine, RD Systems). The lower detection limit of the TPO assay was 30 pg/mL. The median serum TPO level in normal subjects was 55 pg/mL (range 31-88 pg/mL). At the start of induction chemotherapy, the patients had a median serum TPO level of 261 pg/mL (range 120-2150 pg/mL): 10 to 12 days after the end of chemotherapy, their TPO levels were substantially increased, the median value being 1900 pg/mL (range 1050-4194 pg/mL). As a result of chemotherapy, the patients developed aplasia: after the administration of platelet transfusions their median PC increased to 21 x 10(9)J1 (range 5-55 x 109/1), while the median TPO value decreased by 300 pgžmL (range 11-1125 pg/mL). The correlation between PC and TPO was statistically significant prior to chemotherapy (p = 0,04). After chemotherapy, in the period of regeneration, PC increased, while TPO levels decreased to normal levels.
