Sproščanje tkivnega aktivatorja plazminogena v pljučih : magistrsko delo

We studied the basal release of tissue type plasminogen activator (t-PA) and its fast acting inhibitor (PAI-1) in vascular compartment of lungs and the influence of pulmonary hypertension on the I-PA ...release. During the diagnosticheart catheterization 4 samples of blood were takenČ from cubital and femoral vein and simultaneously from right atrium and left ventricle. The following fibrinolytic parameters were determinedČ euglobulin clot lysis time (ECLT), t-PA antigen, t-PA activity, PAI-1 antigen and PAI-1 activity. During the blood flow through the lungs the concentration of t-PA antigen was slightly, though not significantly reduced, while the concentration of PAI-1 antigen was unchanged and the PAI-1 activity was reduced (p less th. 0,01). The concentrations of t-PA and PAI-1 antigens did not differ significantly between the cubital vein, femoral vein and right atrium. The influence of pulmonary hypertension on t-PA release we studied in the group of 12 patients with pulmonary hypertension (the systolic blood pressure in the pulmonary artery ranged from 31 to 130 mmHg) and in the control group of 9 patients withnormal blood pressures in the lung circulation. There was no differences in the concentration of t-PA antigen in right atrium and left ventricle in both groups. It was concluded that t-PA was not released in any appreciable quantity into the vascular compartment of the lung. The reduction of blood PAI-l activity during the flow through the lung can be speculatively attributed to PAI-1 antigen neutralization with other proteins such as the urokinase type plasminogen activator (u-PA). The pulmonary hypertension has noeffect on t-PA release in the lungs.
