The NASA Goddard Earth Observing System global model is evaluated through a cascade of simulations with increasing horizontal resolution. This model employs a non-hydrostatic dynamical core and ...includes a scale-aware, deep convection parameterization (DPCP). 40-daysimulations at six resolutions (100km to 3km) with unvarying model formulation were produced. At the highest resolution, extreme experiments were carried out; one with no DPCP, and one with its scale-awareness eliminated. Simulated precipitation, radiative balance, and atmospheric thermodynamic and dynamical variables are well reproduced with respect to both observational and reanalysis data. As model resolution increases, the convective precipitation smoothly transitions from being mostly produced by the convection parameterization to the cloud microphysics parameterization. However, contrary to current thought, these extreme cases argue for maintaining, to some extent, the scale-aware deep convection parameterization even at 3kmscale, as the run relying solely on explicit grid-scale production of rainfall performs more poorly at this resolution.

The Atmospheric Boundary Layer (ABL) height is a key parameter in air quality research as well as for numerical simulations and forecasts. The identification of thermally stable layers, often with ...radiosondes, has been a common approach for estimating ABL height, though with limited temporal coverage. Remote sensing techniques offer essentially continuous measurements. Nevertheless, ABL height retrievals from different methods can vary greatly when compared, which is particularly notable for topographically complex terrains, such as that surrounding Mexico City. This study, employing one year of data in Mexico City, reveals that the daytime convective boundary layer height (retrieved from Doppler lidar data) is typically lower than the aerosol layer height (retrieved from ceilometer data). Although both estimated heights evolved diurnally, the more elevated aerosol layer decays more slowly, suggesting that the mechanisms that elevate aerosols are not limited to convective motions. Additionally, both diurnal and seasonal variability are investigated, comparing remotely sensed-retrieved heights with thermally stable layers estimated from radiosonde data. Multiple stable layers often develop, those at higher levels have similar values to the ceilometer-retrieved heights, while stable layers at lower heights are similar to Doppler lidar height retrievals. The present research constitutes the first detailed analysis of ceilometer backscatter and Doppler lidar thresholding methods for estimating ABL height over Mexico City, and our results illustrate the complexity of mixing mechanisms on the ABL in this region of complex orography.

Improving T cell Ags by altering MHC anchor residues is a common strategy used to enhance peptide vaccines, but there has been little assessment of how such modifications affect TCR binding and T ...cell recognition. In this study, we use surface plasmon resonance and peptide-MHC tetramer binding at the cell surface to demonstrate that changes in primary peptide anchor residues can substantially and unpredictably alter TCR binding. We also demonstrate that the ability of TCRs to differentiate between natural and anchor-modified heteroclitic peptides distinguishes T cells that exhibit a strong preference for either type of Ag. Furthermore, we show that anchor-modified heteroclitic peptides prime T cells with different TCRs compared with those primed with natural Ag. Thus, vaccination with heteroclitic peptides may elicit T cells that exhibit suboptimal recognition of the intended natural Ag and, consequently, impaired functional attributes in vivo. Heteroclitic peptide-based immune interventions therefore require careful evaluation to ensure efficacy in the clinic.
Heat and drought affect plant chemical defenses and thereby plant susceptibility to pests and pathogens. Monoterpenes are of particular importance for conifers as they play critical roles in defense ...against bark beetles. To date, work seeking to understand the impacts of heat and drought on monoterpenes has primarily focused on young potted seedlings, leaving it unclear how older age classes that are more vulnerable to bark beetles might respond to stress. Furthermore, we lack a clear picture of what carbon resources might be prioritized to support monoterpene synthesis under drought stress. To address this, we measured needle and woody tissue monoterpene concentrations and physiological variables simultaneously from mature piñon pines (
Pinus edulis
) from a unique temperature and drought manipulation field experiment. While heat had no effect on total monoterpene concentrations, trees under combined heat and drought stress exhibited ~ 85% and 35% increases in needle and woody tissue, respectively, over multiple years. Plant physiological variables like maximum photosynthesis each explained less than 10% of the variation in total monoterpenes for both tissue types while starch and glucose + fructose measured 1-month prior explained ~ 45% and 60% of the variation in woody tissue total monoterpene concentrations. Although total monoterpenes increased under combined stress, some key monoterpenes with known roles in bark beetle ecology decreased. These shifts may make trees more favorable for bark beetle attack rather than well defended, which one might conclude if only considering total monoterpene concentrations. Our results point to cumulative and synergistic effects of heat and drought that may reprioritize carbon allocation of specific non-structural carbohydrates toward defense.

Readily available testing for SARS-CoV-2 is necessary to mitigate COVID-19 disease outbreaks. At-home collection kits, in which samples are self-collected without requiring a laboratory or clinic ...visit and sent to an external laboratory for testing, can provide convenient testing to those with barriers to access. They can prevent unnecessary exposure between patient and clinical staff, increase access for patients with disabilities or remote workers, and decrease burdens on health care resources, such as provider time and personal protective equipment. Exact Sciences developed an at-home collection kit for samples to be tested to detect SARS-CoV-2 that includes an Instructions for Use (IFU) document, which guides people without prior experience on collecting a nasal swab sample. Demonstrating successful sample collection and usability is critical to ensure that these samples meet the same high-quality sample collection standards as samples collected in clinics.
The aim of this study was to determine the usability of a SARS-CoV-2 at-home nasal swab sample collection kit.
A human factors usability study was conducted with 30 subjects without prior medical, laboratory, or health care training and without COVID-19 sample self-collection experience. Subjects were observed while they followed the IFU for the at-home sample collection portion of the SARS-CoV-2 test in a setting that simulated a home environment. IFU usability was further evaluated by requiring the subjects to complete a survey, answer comprehension questions, provide written feedback, and respond to questions from the observer about problems during use.
All 30 subjects successfully completed the sample collection process, and all 30 samples were determined by reverse transcription-polymerase chain reaction (RT-PCR) testing to meet quality standards for SARS-CoV-2 testing. The subjects' written feedback and comments revealed several recommendations to improve the IFU.
The study demonstrated the overall usability of an at-home SARS-CoV-2 collection kit. Various feedback mechanisms provided opportunities to improve the wording and graphics for some critical tasks, including placing the label correctly on the tube. A modified IFU was prepared based on study outcomes.
Background
Insufficient heat acclimatization is a risk factor for heat‐related illness (HRI) morbidity, particularly during periods of sudden temperature increase. We sought to characterize heat ...exposure on days before, and days of, occupational HRIs.
Methods
A total of 1241 Washington State workers' compensation State Fund HRI claims from 2006 to 2021 were linked with modeled parameter‐elevation regressions on independent slopes model (PRISM) meteorological data. We determined location‐specific maximum temperatures (Tmax,PRISM) on the day of illness (DOI) and prior days, and whether the Tmax,PRISM was ≥10.0°F (~5.6°C) higher than the average of past 5 days (“sudden increase”) for each HRI claim. Claims occurring on days with ≥10 HRI claims (“clusters”) were compared with “non‐cluster” claims using t tests and χ2 tests.
Results
Seventy‐six percent of analyzed HRI claims occurred on days with a Tmax,PRISM ≥ 80°F. Claims occurring on “cluster” days, compared to “non‐cluster” days, had both a significantly higher mean DOI Tmax,PRISM (99.3°F vs. 85.8°F 37.4°C vs. 29.9°C, t(148) = −18, p < 0.001) and a higher proportion of “sudden increase” claims (80.2% vs. 24.3%, χ21 = 132.9, p < 0.001). Compared to “cluster” days, HRI claims occurring during the 2021 Pacific Northwest “heat dome” had a similar increased trajectory of mean Tmax,PRISM on the days before the DOI, but with higher mean Tmax,PRISM.
Conclusions
Occupational HRI risk assessments should consider both current temperatures and changes in temperatures relative to prior days. Heat prevention programs should include provisions to address acclimatization and, when increases in temperature occur too quickly to allow for sufficient acclimatization, additional precautions.

In this review, we sought to assess from a study of the literature whether five in organic metals (viz., cadmium, copper, lead, nickel, and zinc) bio magnify in aquatic food webs. We also examined ...whether accumulated metals were toxic to consumers/predators and whether the essential metals (Cu and Zn and possibly Ni) behaved differently from non-essential ones (Cd and Pb). Biomagnification potential was indexed by the magnitude of single and multiple trophic transfers in food chains. In this analysis, we used three lines of evidence-laboratory empirical, biokinetic modeling, and field studies-to make assessments. Trophic transfer factors, calculatedfrom lab studies, field studies, and biokinetic modeling, were generally congruent.Results indicated that Cd, Cu, Pb, and Zn generally do not biomagnify in food chains consisting of primary producers, macro invertebrate consumers, and fish occupying TL 3 and higher. However, bio magnification of Zn (TTFs of 1-2) is possible for circumstances in which dietary Zn concentrations are below those required for metabolism. Cd, Cu, Ni, and Zn may biomagnify in specific marine food chains consisting of bivalves, herbivorous gastropods, and barnacles at TL2 and carnivorous gastropods at TL3. There was an inverse relationship between TTF and exposure concentration for Cd, Cu, Pb, and Zn, a finding that is consistent with previous reviews of bioconcentration factors and bioaccumulation factors for metals. Our analysis also failed to demonstrate a relationship between the magnitude of TTFsand dietary toxicity to consumer organisms. Consequently, we conclude that TTFs for the metals examined are not an inherently useful predictor of potential hazard(i.e., toxic potential) to aquatic organisms. This review identified several uncertainties or data gaps, such as the relatively limited data available for nickel, reliance upon highly structured food chains in laboratory studies compared to the unstructured food webs found in nature, and variability in TTFs between the organisms found in different habitats, and years sampled.
Abstract The wetsalts tiger beetle, Cicindelidia haemorrhagica (LeConte) (Coleoptera: Cicindelidae), is found in several active thermal hot spring areas in Yellowstone National Park (YNP) where ...substrate surface temperatures can exceed 50 °C. However, relationships between surface temperatures and the time adults spend on them remain poorly understood. Therefore, we characterized thermal profiles of Dragon Spring and Rabbit Creek, 2 thermally active research sites containing C. haemorrhagica in YNP, to quantify the time adults spend at different surface temperatures. We took 58 thermal video recordings of adults over 6 total days of observation ranging from 10 to 15 min for each adult. Thermal video analysis results indicated a positive relationship between the total time adult beetles spent on surface temperatures from Dragon Spring and Rabbit Creek as temperatures increased from 20 °C. Once surface temperatures exceeded 40 °C, the total time spent at those surface temperatures declined. Adults were recorded on substrates exceeding 50 °C at one of the 2 research locations. Rabbit Creek had substantially more instances of adults present with surface temperatures exceeding 40 °C, including one individual on a surface temperature of 61.5 °C. There were 3 instances of beetles spending more than 4 min at a particular surface temperature, all within the preferred range of 30–40 °C. Our thermal profile results and previous behavioral observations suggest that adults may be resistant to the heat produced from the thermal waters that influence the substrate temperatures but may not be subject to high surface temperatures as previously reported.
