The objective of this study is to demonstrate the importance of the existing Ways of Saint James in Trás-os-Montes and Alto Douro, in the northern interior of Portugal, and that being integrated in ...soft tourism, can constitute a mechanism that allows the revitalization of rural areas, especially the sparsely populated areas, because it allows a series of benefits such as the increase of tourists and pilgrims, the creation of new services, among other structures, thus boosting tourism development. Above all, these Paths linked to soft tourism, allow us to highlight the predominance of tourism linked to nature tourism and slow mobility. In order to carry out this study, the methodology used will be based on interviews to tourists and pilgrims in order to obtain testimonies that can characterise the impact of the Ways themselves on rural territories.

The vital enzyme O-linked β-N-acetylglucosamine transferase (OGT) catalyzes the O-GlcNAcylation of intracellular proteins coupling the metabolic status to cellular signaling and transcription ...pathways. Aberrant levels of O-GlcNAc and OGT have been linked to metabolic diseases as cancer and diabetes. Here, a new series of peptidomimetic OGT inhibitors was identified highlighting the compound LQMed 330, which presented better IC50 compared to the most potent inhibitors found in the literature. Molecular modeling study of selected inhibitors into the OGT binding site provided insight into the behavior by which these compounds interact with the enzyme. The results obtained in this study provided new perspectives on the design and synthesis of highly specific OGT inhibitors.

This paper investigates the relationship between health outcomes at birth and fetal exposure to temperature shocks in rural Colombia during 1999–2008. We overcome a limitation of previous studies, ...confined by small samples and restricted areas, by using records on nearly 1.5million births. We exploit variations in exposure to temperature shocks by municipality and by year and month of conception in a fixed effects model. We find that exposure to moderate heat waves during the third trimester of pregnancy reduces the birthweight of the infant by about 4.1g. Furthermore, exposure to moderate cold shocks during the first and second trimesters of pregnancy reduces the length at birth by 0.014–0.018cm. We also find evidence of the negative effects of heat weaves in Apgar tests at a magnitude of −0.2 to −0.6 percentage points of the normal Apgar score.

To test whether remote ischaemic conditioning (RIC) as adjuvant to standard of care (SOC) would prevent progression towards heart failure (HF) after ST-elevation myocardial infarction (STEMI). ...Single-centre parallel 1:1 randomized trial (computerized block-randomization, concealed allocation) to assess superiority of RIC (3 cycles of intermittent 5 min lower limb ischaemia) over SOC in consecutive STEMI patients (NCT02313961, clinical trials.gov). From 258 patients randomized to RIC or SOC, 9 and 4% were excluded because of unconfirmed diagnosis and previously unrecognized exclusion criteria, respectively. Combined primary outcome of cardiac mortality and hospitalization for HF was reduced in RIC compared with SOC (
n
= 231 and 217, respectively; HR = 0.35, 95% CI 0.15–0.78) as well as each outcome in isolation. No difference was found in serum troponin I levels between groups. Median and maximum follow-up time were 2.1 and 3.7 years, respectively. In-hospital HF (RR = 0.68, 95% CI 0.47–0.98), need for diuretics (RR = 0.68, 95% CI 0.48–0.97) and inotropes and/or intra-aortic balloon pump (RR = 0.17, 95% CI 0.04–0.76) were decreased in RIC. On planned 12 months follow-up echocardiography (
n
= 193 and 173 in RIC and SOC, respectively) ejection fraction (EF) recovery was enhanced in patients presenting with impaired left ventricular (LV) function (10% absolute difference in median EF compared with SOC;
P
< 0.001). In addition to previously reported improved myocardial salvage index and reduced infarct size RIC was shown beneficial in a combined hard clinical endpoint of cardiac mortality and hospitalization for HF. Improved EF recovery was also documented in patients with impaired LV function.

Abstract Acute myeloid leukaemia (AML) is a severe haematological neoplasm that originates from the transformation of haematopoietic stem cells (HSCs) into leukaemic stem cells (LSCs). The bone ...marrow (BM) microenvironment, particularly that of mesenchymal stromal cells (hMSCs), plays a crucial role in the maintenance of HSCs. In this context, we explored whether alterations in the secretome of hMSCs derived from AML patients (hMSC-AML) could impact HSC gene expression. Proteomic analysis revealed that the secretome of coculture assays with hMSC-AMLs and HSC from healthy donor is altered, with increased levels of secretory leukocyte protease inhibitor (SLPI), a protein associated with important processes for maintenance of the haematopoietic niche that has already been described to be altered in several tumours. Increased SLPI expression was also observed in the BM plasma of AML patients. Transcriptome analysis of HSCs cocultured with hMSC-AML in comparison with HSCs cocultured with hMSC-HD revealed altered expression of SLPI target genes associated with the cell cycle, proliferation, and apoptosis. Important changes were identified, such as increased expression levels of CCNA2, CCNE2, CCND2, CD133 and CDK1 and decreased levels of CDKN2A and IGFBP3, among others. Overall, these findings suggest that the altered secretome of coculture assays with hMSC-AMLs and HSC from healthy donor, particularly increased SLPI expression, can contribute to gene expression changes in HSCs, potentially influencing important molecular mechanisms related to AML development and progression.

Comparing the results of estimates for population, life expectancies, and mortality in children under-5 from GBD 2021 with those from long-standing efforts of the UN, including their Division of ...Economic and Social Affairs (DESA), Population Division's World Population Prospects (WPP),2 and the Inter-agency Group for Child Mortality Estimation (IGME; a consortium of UNICEF, WHO, UNDESA Population Division, and the World Bank Group),3 reveals reassuring similarities in global population numbers. Differences in the estimates from different groups come from several sources: the modelling methods, inclusion and exclusion of nationally provided datapoints, the extent to which data sources are considered to be nationally representative, and whether or not their measurement is accurate and plausible in the context of baseline reference data or similar countries with better data.6 Many countries could benefit from knowing which of their data sources get used in estimation efforts and why. Ultimately, a more solid empirical base for global estimates of mortality, life expectancy, and population projections will only come from efforts to improve country data and statistical systems including household surveys, the Health Management Information Systems (HMIS), and Civil Registration and Vital Statistics (CRVS) systems.7 CRVS is key to producing timely country-reported information, the kind useful to national governments and global donors alike.

The cotton boll weevil, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae), is one of the most important insect pests of cotton, causing significant yield losses. This insect completes ...its life cycle only on the flower structures of some Malvaceae species, with cotton being its preferred host. However, during the cotton off‐season, the boll weevil enters a reproductive dormancy state, in tropical regions, with important alterations in their reproduction organs. During this period, a remaining insect population survives feeding on a variety of other food sources, but the insects do not reproduce. We hypothesized that these alterations in the insect's reproductive organs are associated with the nutritional levels provided by alternative food sources during the cotton off‐season. To test our hypothesis, we first investigated food sources that sustain insect survival for long periods, such as weed species and fruits of cultivated crops. Then, we evaluated how various food sources affect the reproductive organs and reproductive capacity of the boll weevil. Among the alternative food sources tested, insect longevity was highest on banana (Musa paradisiaca L.), mango (Mangifera indica L.), papaya (Carica papaya L.), milkweed (Euphorbia heterophylla L.), and lilac tasselflower (Emilia sonchifolia L.). However, only banana and mango resulted in greater longevity than cotton squares. Banana was the preferred food and resulted in the highest levels of nutrients in the insects. Additionally, insects previously fed on banana made more oviposition punctures on cotton squares than those fed only on cotton squares, which resulted in a significantly higher number of emerged adults. Histological analysis of the insects' reproductive organs showed that feeding on papaya resulted in morphological changes and testicle degradation. In conclusion, we present evidence that boll weevils fed on alternative food sources (other than cotton squares or similar‐quality food) display a nutritional imbalance, associated with substantial alterations in the reproductive tissues of this insect, which may trigger the reproductive dormancy state.
Anthonomus grandis (Coleoptera: Curculionidae) feeds preferentially on cotton and completes its cycle only on a few plants of the Malvaceae family. However, this insect survives the cotton off‐season period feeding on other plant species, such as cultivated fruits or weed flowers. These insects survived longer and preferred banana over cotton squares. When they fed on low‐nutritional food (e.g., papaya fruit), their reproductive organs (testicles and oocytes) showed an abnormal morphology, which impaired their ability to lay viable eggs.

Purpose: The purpose of this systematic review was to evaluate the survival rate, biological complications, technical complications, and clinical behavior of single crowns supported by teeth made up ...in monolithic zirconia with CAD/CAM technology.Study selection: An extensive electronic search was conducted through Medline/PubMed, Embase, and Cochrane Library databases. Additional manual search was performed on the references of included articles to identify relevant publications. Two reviewers independently performed the selection and electronic and manual search.Results: From nine articles included, there was a total of 594 participants and 1657 single-tooth restorations with a mean exposure time of 1.07 years, and follow-up period between 0.3 and 2.1 years. All studies showed a moderate level of quality, with a consequent moderate possibility of associated bias, using the Newcastle-Ottawa Scale (NOS), with survival rate (SR) ranging between 91% to 100%. Bleeding on probing (BOP) were reported with an average value of 29.12%. Marginal integrity showed high success rate values for the observation periods, except for one that included patients with bruxism which obtained a SR of 31.60%. Failures and/or fractures, mostly total and requiring replacement, were observed in three studies. Linear regression showed that there was no statistical correlation between survival rate and type of cementation and the average years of follow-up (p=0.730 e p=0.454). There was high heterogeneity between studies (I2 = 93.74% and Q = 79.672).Conclusion: Within the limitation of this study, monolithic zirconia might be considered as a possible option for restoring single crowns, especially in the posterior zone.
