Coral reef ecosystems have suffered an unprecedented loss of habitat-forming hard corals in recent decades. While marine conservation has historically focused on passive habitat protection, demand ...for and interest in active restoration has been growing in recent decades. However, a disconnect between coral restoration practitioners, coral reef managers and scientists has resulted in a disjointed field where it is difficult to gain an overview of existing knowledge. To address this, we aimed to synthesise the available knowledge in a comprehensive global review of coral restoration methods, incorporating data from the peer-reviewed scientific literature, complemented with grey literature and through a survey of coral restoration practitioners. We found that coral restoration case studies are dominated by short-term projects, with 60% of all projects reporting less than 18 months of monitoring of the restored sites. Similarly, most projects are relatively small in spatial scale, with a median size of restored area of 100 m2. A diverse range of species are represented in the dataset, with 229 different species from 72 coral genera. Overall, coral restoration projects focused primarily on fast-growing branching corals (59% of studies), and report survival between 60 and 70%. To date, the relatively young field of coral restoration has been plagued by similar 'growing pains' as ecological restoration in other ecosystems. These include 1) a lack of clear and achievable objectives, 2) a lack of appropriate and standardised monitoring and reporting and, 3) poorly designed projects in relation to stated objectives. Mitigating these will be crucial to successfully scale up projects, and to retain public trust in restoration as a tool for resilience based management. Finally, while it is clear that practitioners have developed effective methods to successfully grow corals at small scales, it is critical not to view restoration as a replacement for meaningful action on climate change.

Coral-reef ecosystems are experiencing frequent and severe disturbance events that are reducing global coral abundance and potentially overwhelming the natural capacity for reefs to recover. While ...mitigation strategies for climate warming and other anthropogenic disturbances are implemented, coral restoration programmes are being established worldwide as an additional conservation measure to minimise coral loss and enhance coral recovery. Current restoration efforts predominantly rely on asexually produced coral fragments—a process with inherent practical constraints on the genetic diversity conserved and the spatial scale achieved. Because the resilience of coral communities has hitherto relied on regular renewal with natural recruits, the scaling-up of restoration programmes would benefit from greater use of sexually produced corals, which is an approach that is gaining momentum. Here we review the present state of knowledge of scleractinian coral sexual reproduction in the context of reef restoration, with a focus on broadcast- spawning corals. We identify key knowledge gaps and bottlenecks that currently constrain the sexual production of corals and consider the feasibility of using sexually produced corals for scaling-up restoration to the reef- and reef-system scales.

This community-wide tropicalization fundamentally altered key ecological processes, suppressing the recovery of kelp forests.

Abstract
Mesophotic coral ecosystems (MCEs) occur at depths beyond those typically associated with coral reefs. Significant logistical challenges associated with data collection in deep water have ...resulted in a limited understanding of the ecological relevance of these deeper coral ecosystems. We review the trends in this research, covering the geographic spread of MCE research, the focus of these studies, the methods used, how MCEs differ in terms of species diversity and begin to assess connectivity of coral populations. Clear locational biases were observed, with studies concentrated in a few discrete areas mainly around the Atlantic region. The focus of MCE studies has diversified in recent years and more detailed aspects of MCE ecology are now being investigated in particular areas of research. Advances in technology are also reflected in the current range of research, with a wider variety of methods now employed. However, large information gaps are present in entire regions and particularly in relation to the threats, impacts and subsequent management of MCEs. Analysis of species diversity shows that initial definitions based on depth alone may not be appropriate globally, while further taxonomic resolution may also be required to deduce the full biodiversity of major groups in certain regions. Genetic studies to date show species-specific results, although distinct deeper populations do appear to exist, which raises questions regarding the potential of MCEs to act as refugia.
Decadal-scale observations of marine reserves suggest that indirect effects on taxa that occur through cascading trophic interactions take longer to develop than direct effects on target species. ...Combining and analyzing a unique set of long-term time series of ecologic data in and out of fisheries closures from disparate regions, we found that the time to initial detection of direct effects on target species (±SE) was 5.13 ± 1.9 years, whereas initial detection of indirect effects on other taxa, which were often trait mediated, took significantly longer (13.1 ± 2.0 years). Most target species showed initial direct effects, but their trajectories over time were highly variable. Many target species continued to increase, some leveled off, and others decreased. Decreases were due to natural fluctuations, fishing impacts from outside reserves, or indirect effects from target species at higher trophic levels. The average duration of stable periods for direct effects was 6.2 ± 1.2 years, even in studies of more than 15 years. For indirect effects, stable periods averaged 9.1 ± 1.6 years, although this was not significantly different from direct effects. Populations of directly targeted species were more stable in reserves than in fished areas, suggesting increased ecologic resilience. This is an important benefit of marine reserves with respect to their function as a tool for conservation and restoration.

(COTS) collected from the Great Barrier Reef showed average female gonad mass of 16 % (±8 s.d.) and for males 12 % (±6 s.d.). In females ...up to 34 % of the body mass could be devoted to gonad. Based on these data, we also derived relationships between diameter gonad weight and reproductive output for both male and female starfish. Due to the large average size of individuals in this study (39.2 ± 0.3 cm diameter), the potential oocyte production of females was between 29 and 38 million eggs per season for average size starfish. The highest oocyte production was estimated to be >100 million oocytes, which is the highest ever recorded for an individual female starfish. These relationships imply that the largest

We suggest that management targets for maintaining COTS at pre-outbreak thresholds should take account of starfish size as well as starfish density, especially given extreme reproductive potential of large starfish in pre-outbreak populations.

Infantile globoid cell leukodystrophy (GLD, Krabbe disease) is a fatal demyelinating disorder caused by a deficiency in the lysosomal enzyme galactosylceramidase (GALC). GALC deficiency leads to the ...accumulation of the cytotoxic glycolipid, galactosylsphingosine (psychosine). Complementary evidence suggested that psychosine is synthesized via an anabolic pathway. Here, we show instead that psychosine is generated catabolically through the deacylation of galactosylceramide by acid ceramidase (ACDase). This reaction uncouples GALC deficiency from psychosine accumulation, allowing us to test the long-standing “psychosine hypothesis.” We demonstrate that genetic loss of ACDase activity (Farber disease) in the GALC-deficient mouse model of human GLD (twitcher) eliminates psychosine accumulation and cures GLD. These data suggest that ACDase could be a target for substrate reduction therapy (SRT) in Krabbe patients.We show that pharmacological inhibition of ACDase activity with carmofur significantly decreases psychosine accumulation in cells from a Krabbe patient and prolongs the life span of the twitcher (Twi) mouse. Previous SRT experiments in the Twi mouse utilized L-cycloserine, which inhibits an enzyme several steps upstream of psychosine synthesis, thus altering the balance of other important lipids. Drugs that directly inhibit ACDase may have a more acceptable safety profile due to their mechanistic proximity to psychosine biogenesis. In total, these data clarify our understanding of psychosine synthesis, confirm the long-held psychosine hypothesis, and provide the impetus to discover safe and effective inhibitors of ACDase to treat Krabbe disease.
