Autologous and allogeneic cryoimmunological medicine is a brand new branch of biomedical science and clinical practice that examines the features and formation of the immune response to immunogenic ...properties of normal and malignant biological structures altered by ultralow temperature, as well as specific changes in the structural and functional characteristics of immune cells and tissues after cryopreservation. Cryogenic protein denaturation phenomenon provides important insights into the mechanisms underlying the damage to cryogenic lesions immediately after freeze–thawing sessions in bioscience and medicine applications. The newly formed cryocoagulated protein components (cryomodified protein components) are crucial in cryoimmunology from the perspective of the formation of immunological substances at ultralow temperatures. Dendritic cells and cryocell detritus (cryocell debris) formed in living biological tissue after exposure to ultralow temperature in vivo may be an indication of one of the essential mechanisms involved in the cryoimmunological response of living structures to the impact of ultralow temperature exposure. Hence, the formation of new autologous and allogeneic cryoinduced immunogenic substances is a novel concept in biomedical research globally. Accordingly, this review focuses on issues concerning the peculiarities of the interaction of the immune system with a dominant malignant neoplasm tissue after exposure to subzero temperatures, considering the original cryogenic technical approaches. We present an overview of the state-of-the-art methods of cryoimmunology, and their major developments, past and present. The need for the delineation of structural and functional characteristics of the biological substrates of the immune system after cryopreservation that can be used in adoptive cell therapy, especially in cancer patients, is emphasized.

The question of the impact of physical and chemical factors, which are implemented at the stage of lyophilization, on structure and functions of HCBL cells has been considered. The importance of such factors as cell concentration, storage conditions, residual moisture in ensuring the preservation of lyophilized material was disputed. The paper analyzes the data on the use of lyoprotectants and antioxidants during freeze-drying as well as discusses the safety of using lyoprotectant delivery methods in relation to the genomic profile of cells. The review surveys the published data on a strong negative impact of psycho-emotional stress, which accompanies both the military servicemen and civilians of any state in martial law period. Such a ...load can lead to severe disorders of the systems that ensure the human body homeostatic stability, in particular to post-traumatic stress disorder. In this case human body is subjected to a state accompanied by the development of severe oncology and autoimmune diseases, as well as dysfunction of the male reproductive system. The study offers modern approaches to the treatment of the listed pathologies, most of which are based on the international experience of clinical and experimental research as well as achievements of national academic science. Special attention is paid to the development of cryobiological and cryomedical cell-tissue techniques, the use of those can be promising in the strengthening of both mental and physical health of civilian population during the wartime.
The review considers the possibility of using modern nanotechnological developments aimed to achieve alternative cryobiological goals. On the one hand, the use of nanomaterials will increase the ...functional value of thawed cells due to such unique characteristics of nanoparticles as size, shape, surface charge, chemical composition, etc. Nanomaterials can be used as nanocontainers for impermeable cryoprotective agents (CPAs) and cause significant changes in crystal formation, thermal conductivity and other properties of cells, tissues and organs, that increases the efficiency of their cryopreservation. On the other hand, the combined use of nanomaterials and low-temperature freezing factors is considered a promising method of destruction of pathologically altered cells and tissues, as it minimizes the risk of recurrence of oncopathology after insufficient freezing-out of the tumor site.
Проблематика. Атопічний дерматит (АД) – запальне захворювання шкіри, обумовлене зрушенням взаємодії клітин імунної системи та шкіри. Стероїдні протизапальні препарати, що використовуються як терапія ...АД, викликають чимало побічних ефектів. Тому актуальною є розробка нових природних поліфункціональних засобів з імунокоригувальною активністю. Мета. Визначення зміни показників лейкоцитарної ланки периферичної крові, а також встановлення характеру цитоморфологічних змін у селезінці та лімфатичних вузлах (ЛВ) щурів з індукованим АД до та після застосування кріоконсервованих (кЛПКЛ) та ліофілізованих (лЛПКЛ) форм лейкоконцентрату пуповинної крові людини. Методика реалізації. Експерименти були проведені на 6-місячних щурах лінії Вістар. Ініціювали АД шляхом втирання у шкіру спини 5 %-вого спиртово-ацетонового розчину динітрохлорбензолу (ДНХБ) протягом 21-ї доби. Кожну з форм препарату (кЛПКЛ і лЛПКЛ) вводили внутрішньочеревно по 0,5 мл у дозі 5´106 клітин через добу після закінчення застосування ДНХБ. Визначалися кількість лейкоцитів і співвідношення нейтрофилів та лімфоцитів (індекс Н/Л) у крові, а також цитоморфологічні характеристики селезінки та пахових ЛВ. Результати. При АД спостерігали певні особливості зміни цитоморфологічних характеристик структур лімфогемопоетичного комплексу (ЛГК), а саме селезінки і ЛВ, на тлі зміни показників лейкоцитарної ланки. Найбільш істотними змінами в селезінці була перевага білої пульпи над червоною, а в ЛВ – превалювання фолікулів ІІ та ІІІ стадій розвитку. У крові тварин з АД відзначався виражений лейкоцитоз, зростав індекс Н/Л. Показано позитивні коригувальні ефекти кЛККЧ і лЛККЧ щодо морфологічних характеристик ЛГК і зазначених показників крові. Висновки. Доведено ефективність застосування кЛПКЛ і лЛПКЛ з метою корекції показників лейкоцитарної ланки крові та структурної організації лімфогемопоетичних органів при розвитку АД.

) and cholesterol, to inhibit the growth of ...Ehrlich’s ascites carcinoma (EAC). However, the biosafety of these NCs remains unclear. Our objective was to investigate the acute and subchronic toxicity of NCs. NCs were administered to BALB/c mice in NPs concentration of 5.9; 29.5; 59.1; and 118.2 mg/kg. Acute toxicity was induced by a single administration of NCs, subchronic—by repeated daily administration of NCs for 14 days. On day 15 and on day 31 for acute and subchronic toxicity, respectively, the percentage of animal survival, body weight, condition of visceral organs, and activities of γ-glutamyl transferase (GGT) and glucose-6-phosphate dehydrogenase (G-6-PDH) were determined. It was found that administration of NCs in the concentration of 5.9 mg/kg and 29.5 mg/kg of NPs did not influence on survival of animals or have a negative impact on their performance status, morphological and quantitative characteristics of visceral organs, and activities of the GGT and G-6-PDH in the liver. For acute toxicity, the semi-lethal dose (LD

Background. Atopic dermatitis (AD) is an inflammatory skin disease caused by a shift in the interaction between cells of the immune system and the skin. Steroid anti-inflammatory drugs used to treat ...AD often lead to numerous side effects. Therefore, the development of new natural multifunctional preparations with immunomodulatory activity is of great importance.
Objective. To determine the changes in indices of the leukocytes cluster of peripheral blood and determine the peculiarities of cytomorphological changes in the spleen and lymph nodes (LN) of rats with induced AD before and after administration of cryopreserved (cHCBL) and lyophilized (lHCBL) forms of human cord blood leukoconcentrate.
Methods. The experiments were conducted using 6-month-old Wistar rats. AD was induced by topically applying a 5% alcohol-acetone solution of dinitrochlorobenzene (DNCB) to the skin of the rats' back for 21 days. Each form of the preparation (cHCBL and lHCBL) was administered intraperitoneally in a 0.5 ml dose containing 5´106 cells, one day after the final DNCB treatment. The following parameters were assessed in the blood: leukocyte count and the neutrophil-to-lymphocyte ratio (N/L index). Сytomorphological characteristics were determined in the spleen and inguinal LN.
Results. In AD, specific changes in the cytomorphological characteristics of the lymphohematopoietic complex (LHС) structures, namely the spleen and LN, were observed alongside alterations in the leukocyte cluster indices. The most notable changes in the spleen included the dominance of the white pulp over the red pulp and the prevalence of follicles of stages II and III of development in the LN. Animals with AD displayed pronounced leukocytosis and an increased N/L index in their blood. The study demonstrates the positive corrective effects of cHCBL and lHCBL on the morphological characteristics of LHC and the specified blood parameters.
Conclusions. The effectiveness of the cHCBL and lHCBL use for correcting indices of leukocytes cluster of peripheral blood and the structural organization of lymphohemopoietic organs in the development of AD have been proven.
