The desire to increase profit on dairy farms necessitates consideration of the revenue attainable from the sale of surplus calves for meat production. However, the generation of calves that are ...expected to excel in efficiency of growth and carcass merit must not be achieved to the detriment of the dairy female and her ability to calve and re-establish pregnancy early postcalving without any compromise in milk production. Given the relatively high heritability of many traits associated with calving performance and carcass merit, and the tendency for many of these traits to be moderately to strongly antagonistic, a breeding index that encompasses both calving performance and meat production could be a useful tool to fill the void in supporting decisions on bull selection. The objective of the present study was to derive a dairy–beef index (DBI) framework to rank beef bulls for use on dairy females with the aim of striking a balance between the efficiency of valuable meat growth in the calf and the subsequent performance of the dam. Traits considered for inclusion in this DBI were (1) direct calving difficulty; (2) direct gestation length; (3) calf mortality; (4) feed intake; (5) carcass merit reflected by carcass weight, conformation, and fat and the ability to achieve minimum standards for each; (6) docility; and (7) whether the calf was polled. Each trait was weighted by its respective economic weight, most of which were derived from the analyses of available phenotypic data, supplemented with some assumptions on costs and prices. The genetic merit for a range of performance metrics of 3,835 artificial insemination beef bulls from 14 breeds ranked on this proposed DBI was compared with an index comprising only direct calving difficulty and gestation length (the 2 generally most important characteristics of dairy farmers when selecting beef bulls). Within the Angus breed (i.e., the beef breed most commonly used on dairy females), the correlation between the DBI and the index of genetic merit for direct calving difficulty plus gestation length was 0.74; the mean of the within-breed correlations across all other breeds was 0.87. The ranking of breeds changed considerably when ranked based on the top 20 artificial insemination bulls excelling in the DBI versus excelling in the index of calving difficulty and gestation length. Dairy breeds ranked highest on the index of calving difficulty and gestation length, whereas the Holstein and Friesian breeds were intermediate on the DBI; the Jersey breed was one of the poorest breeds on DBI, superior only to the Charolais breed. The results clearly demonstrate that superior carcass and growth performance can be achieved with the appropriate selection of beef bulls for use on dairy females with only a very modest increase in collateral effect on cow performance (i.e., 2–3% greater dystocia expected and a 6-d-longer gestation length).

This assessment by the Environmental Effects Assessment Panel (EEAP) of the United Nations Environment Programme (UNEP) provides the latest scientific update since our most recent comprehensive ...assessment (Photochemical and Photobiological Sciences, 2019, 18, 595–828). The interactive effects between the stratospheric ozone layer, solar ultraviolet (UV) radiation, and climate change are presented within the framework of the Montreal Protocol and the United Nations Sustainable Development Goals. We address how these global environmental changes affect the atmosphere and air quality; human health; terrestrial and aquatic ecosystems; biogeochemical cycles; and materials used in outdoor construction, solar energy technologies, and fabrics. In many cases, there is a growing influence from changes in seasonality and extreme events due to climate change. Additionally, we assess the transmission and environmental effects of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which is responsible for the COVID-19 pandemic, in the context of linkages with solar UV radiation and the Montreal Protocol.

Saccharomyces genomes are highly collinear and show relatively little structural variation, both within and between species of this yeast genus. We investigated the only common inversion polymorphism ...known in S. cerevisiae, which affects a 24-kb 'flip/flop' region containing 15 genes near the centromere of chromosome XIV. The region exists in two orientations, called reference (REF) and inverted (INV). Meiotic recombination in this region is suppressed in crosses between REF and INV orientation strains such as the BY x RM cross. We find that the inversion polymorphism is at least 17 million years old because it is conserved across the genus Saccharomyces. However, the REF and INV isomers are not ancient alleles but are continually being re-created by re-inversion of the region within each species. Inversion occurs due to continual homogenization of two almost identical 4-kb sequences that form an inverted repeat (IR) at the ends of the flip/flop region. The IR consists of two pairs of genes that are specifically and strongly expressed during the late stages of sporulation. We show that one of these gene pairs, YNL018C/YNL034W, codes for a protein that is essential for spore formation. YNL018C and YNL034W are the founder members of a gene family, Centroid, whose members in other Saccharomycetaceae species evolve fast, duplicate frequently, and are preferentially located close to centromeres. We tested the hypothesis that Centroid genes are a meiotic drive system, but found no support for this idea.

Climate models robustly and unanimously simulate narrowing of the intense tropical precipitation under greenhouse gas forcing. We argue that the meridional width of tropical precipitation is ...controlled by the seasonal meridional range of the Intertropical Convergence Zone (ITCZ). The contraction of tropical precipitation under greenhouse forcing results from a reduced seasonal range of ITCZ migration. An energetic theory—similar to the energetic theory for ITCZ shifts based on the hemispheric contrast of energy input to the atmosphere—is developed. The meridional width of tropical precipitation is proportional to the seasonal range of the interhemispheric contrast in atmospheric heating divided by the efficiency of atmospheric cross‐equatorial heat transport. Climate models are biased toward overly expansive tropical precipitation resulting from an exaggerated seasonal atmospheric heating. The robust contraction of tropical precipitation under global warming results from increased efficiency of interhemispheric energy transport consistent with enhanced gross moist stability of the tropical atmosphere.

Dementia with Lewy bodies (DLB) was considered to be an uncommon cause of dementia until improved neuropathological staining methods for ubiquitin were developed in the late 1980's. Subsequent ...recognition that 10-15% of dementia cases in older people were associated with Lewy body pathology led to the publication in 1996 of Consensus clinical and pathological diagnostic criteria for the disorder. These have greatly raised global awareness of DLB and helped to generate a body of knowledge which informs modern clinical management of this pharmacologically sensitive group of patients. They have also enabled important issues surrounding the relationships of DLB with Alzheimer's disease and Parkinson's disease to be addressed and partially resolved. A recent re-evaluation of the Consensus criteria has confirmed many aspects of the original recommendations, supplementing these with suggestions for improved pathological characterisation, clinical detection and management. Virtu-ally unrecognised 20 years ago, DLB could within this decade be one of the best characterised and potentially treatable neurodegenerative disorders of late life.
Serological testing is emerging as a powerful tool to progress our understanding of COVID-19 exposure, transmission and immune response. Large-scale testing is limited by the need for in-person blood ...collection by staff trained in venepuncture, and the limited sensitivity of lateral flow tests. Capillary blood self-sampling and postage to laboratories for analysis could provide a reliable alternative. Two-hundred and nine matched venous and capillary blood samples were obtained from thirty nine participants and analysed using a COVID-19 IgG ELISA to detect antibodies against SARS-CoV-2. Thirty eight out of thirty nine participants were able to self-collect an adequate sample of capillary blood (≥ 50 µl). Using plasma from venous blood collected in lithium heparin as the reference standard, matched capillary blood samples, collected in lithium heparin-treated tubes and on filter paper as dried blood spots, achieved a Cohen's kappa coefficient of > 0.88 (near-perfect agreement, 95% CI 0.738-1.000). Storage of capillary blood at room temperature for up to 7 days post sampling did not affect concordance. Our results indicate that capillary blood self-sampling is a reliable and feasible alternative to venepuncture for serological assessment in COVID-19.

The Environmental Effects Assessment Panel of the Montreal Protocol under the United Nations Environment Programme evaluates effects on the environment and human health that arise from changes in the ...stratospheric ozone layer and concomitant variations in ultraviolet (UV) radiation at the Earth’s surface. The current update is based on scientific advances that have accumulated since our last assessment (Photochem and Photobiol Sci 20(1):1–67, 2021). We also discuss how climate change affects stratospheric ozone depletion and ultraviolet radiation, and how stratospheric ozone depletion affects climate change. The resulting interlinking effects of stratospheric ozone depletion, UV radiation, and climate change are assessed in terms of air quality, carbon sinks, ecosystems, human health, and natural and synthetic materials. We further highlight potential impacts on the biosphere from extreme climate events that are occurring with increasing frequency as a consequence of climate change. These and other interactive effects are examined with respect to the benefits that the Montreal Protocol and its Amendments are providing to life on Earth by controlling the production of various substances that contribute to both stratospheric ozone depletion and climate change.

Summary Background Blood biomarkers of neuronal damage could facilitate clinical management of and therapeutic development for Huntington's disease. We investigated whether neurofilament light ...protein NfL (also known as NF-L) in blood is a potential prognostic marker of neurodegeneration in patients with Huntington's disease. Methods We did a retrospective analysis of healthy controls and carriers of CAG expansion mutations in HTT participating in the 3-year international TRACK-HD study. We studied associations between NfL concentrations in plasma and clinical and MRI neuroimaging findings, namely cognitive function, motor function, and brain volume (global and regional). We used random effects models to analyse cross-sectional associations at each study visit and to assess changes from baseline, with and without adjustment for age and CAG repeat count. In an independent London-based cohort of 37 participants (23 HTT mutation carriers and 14 controls), we further assessed whether concentrations of NfL in plasma correlated with those in CSF. Findings Baseline and follow-up plasma samples were available from 97 controls and 201 individuals carrying HTT mutations. Mean concentrations of NfL in plasma at baseline were significantly higher in HTT mutation carriers than in controls (3·63 SD 0·54 log pg/mL vs 2·68 0·52 log pg/mL, p<0·0001) and the difference increased from one disease stage to the next. At any given timepoint, NfL concentrations in plasma correlated with clinical and MRI findings. In longitudinal analyses, baseline NfL concentration in plasma also correlated significantly with subsequent decline in cognition (symbol-digit modality test r =–0·374, p<0·0001; Stroop word reading r =–0·248, p=0·0033), total functional capacity ( r =–0·289, p=0·0264), and brain atrophy (caudate r =0·178, p=0·0087; whole-brain r =0·602, p<0·0001; grey matter r =0·518, p<0·0001; white matter r =0·588, p<0·0001; and ventricular expansion r =–0·589, p<0·0001). All changes except Stroop word reading and total functional capacity remained significant after adjustment for age and CAG repeat count. In 104 individuals with premanifest Huntington's disease, NfL concentration in plasma at baseline was associated with subsequent clinical onset during the 3-year follow-up period (hazard ratio 3·29 per log pg/mL, 95% CI 1·48–7·34, p=0·0036). Concentrations of NfL in CSF and plasma were correlated in mutation carriers ( r =0·868, p<0·0001). Interpretation NfL in plasma shows promise as a potential prognostic blood biomarker of disease onset and progression in Huntington's disease. Aims To assess the incidence, timing, and relation of drug-eluting stent (DES) thrombosis to discontinuation of clopidogrel therapy. Methods and results This prospective observational cohort study ...included 6816 consecutive patients that underwent successful DES implantation. Primary endpoint was definite stent thrombosis (ST). During 4 years of follow-up, definite ST was observed in 73 patients, corresponding to a cumulative incidence of 1.2%. Cumulative incidence of ST at 30 days was 0.5 and 0.8% at 1 year, respectively. Discontinuation of clopidogrel therapy was significantly associated with ST only in the first 6 months after the procedure (P < 0.001). During that period, the median time interval from clopidogrel discontinuation to ST was 9 days interquartile range (IQR) 5.5–22.5 while thereafter it was 104.3 days (IQR 7.4–294.8). Conclusion The 4 year incidence of ST after DES implantation is low. A relevant number of ST occur early after discontinuation of clopidogrel therapy. The dependence of ST on discontinuation of clopidogrel therapy seems to be mostly confined to the first 6 months after DES implantation. However, specifically designed randomized studies are required to establish the optimal length of clopidogrel therapy after DES implantation.
