Electrospray ionization mass spectrometry (ESI-MS) experiments, including ion mobility spectrometry mass spectrometry (ESI-IMS-MS) and electron capture dissociation (ECD) of proteins ionized from ...aqueous solutions, have been used for the study of solution-like structures of intact proteins. By mixing aqueous proteins with denaturants online before ESI, the amount of protein unfolding can be precisely controlled and rapidly analyzed, permitting the characterization of protein folding intermediates in protein folding pathways. Herein, we mixed various pH solutions online with aqueous cytochrome C for unfolding and characterizing its unfolding intermediates with ESI-MS charge state distribution measurements, IMS, and ECD. The presence of folding intermediates and unfolded cytochrome c structures were detected from changes in charge states, arrival time distributions (ATDs), and ECD. We also compared structures from nondenaturing and denaturing solution mixtures measured under “gentle” (i.e., low energy) ion transmission conditions with structures measured under “harsh” (i.e., higher energy) transmission. This work confirms that when using “gentle” instrument conditions, the gas-phase cytochrome c ions reflect attributes of the various solution-phase structures. However, “harsh” conditions that maximize ion transmission produce extended structures that no longer correlate with changes in solution structure.

Despite the potential use of digital storytelling with marginalised groups, there are few examples of its application in homelessness or examinations of co-creative relationships in this context. ...Along with digital storytelling, this research used immersive media (virtual reality and 360 degree video) to explore place-based social exclusion. In the feasibility study, with four doctoral researchers at Loughborough University as participants, immersive digital stories were co-created. The aim of this study was to understand how to create place-based immersive digital stories, through adapting existing digital storytelling methods and the co-creation of virtual reality, to inform best practices for future studies involving participants who have experienced homelessness. Participants created maps and empathy timelines, shared stories, recorded voiceovers and edited footage. The researcher facilitated this and recorded the 360-degree filmed footage. The final stories proved to explore place-based social exclusion. Co-creative relationships were found to be more significant between the researcher and individual participant than amongst the participants as a group. With immersive media, the researcher’s experience formed an active part of the finished pieces. Despite this, participants described their role as director, being ultimately in control. These findings will influence the methods that will be used in the future with those who have experienced homelessness in Loughborough. They also show how immersive media in digital storytelling can strengthen co-creation and acknowledge the researcher in the story.
For more than a century, the Boone and Crockett Club has kept records of harvest locations for 38 categories of North American big game, which includes measurement details and sizes of enduring ...characteristics (e.g., antlers, skulls, horns). All white-tailed deer (Odocoileus virginianus) entered in the Club records (i.e., record deer) have large antlers because the measurements must meet or exceed a minimum size to qualify for entry. We used the records from the Club to test the hypothesis that the spatial distribution in the harvest of record deer was related to the number of antlered deer harvested, variations in land productivity, soil mineral content, and attributes of land cover for 9 states in the midwestern United States. We used a Bayesian theoretical framework to develop a spatial model that examined the influence of explanatory variables on the number of record deer harvested in each county. In our study area, 3,658 record deer were harvested in 692 of the 856 counties (80.8%). More record deer were harvested in counties that had more high-contrast edges, less contiguous land cover, and greater variation in soil productivity. These results provide information for managers and hunters to better understand the spatial distribution of record deer and factors that are correlated with their distribution.

Technology has the potential to provide more up-to-date information and customised services to train passengers and therefore improve the rail journey experience. However, there is a lack knowledge ...about which innovations and services are preferred by the travelling public. The purpose of this study was to understand the value which passengers placed on technological innovations to improve the overall passenger journey experience. A conjoint analysis survey based on the best-worst scale of preference was developed to evaluate how passengers (N = 398) value different system features proposed to improve passenger experience in the UK. Results show that the automatic compensation for delayed or cancelled trains was valued the highest, and the ability to pre-order special services ranked as least value from a set of ten features. Additional results include the segmentation of responses according to passenger type (commuters, business and leisure) and the similarities and differences in responses from the public versus those working directly in the rail industry. The insights gained from this study suggest which features should be prioritised to improve rail passenger journey experiences.

Cancer-induced bone pain (CIBP) is the most common and devastating symptom of bone metastatic cancer that substantially disrupts patients' quality of life. Currently, there are few effective ...analgesic treatments for CIBP other than opioids which come with severe side effects. In order to better understand the factors and mechanisms responsible for CIBP it is essential to have clinically relevant animal models that mirror pain-related symptoms and disease progression observed in patients with bone metastatic cancer. In the current study, we characterize a syngeneic mouse model of prostate cancer induced bone pain. We transfected a prostate cancer cell line (RM1) with green fluorescent protein (GFP) and luciferase reporters in order to visualize tumor growth longitudinally
and to assess the relationship between sensory neurons and tumor cells within the bone microenvironment. Following intra-femoral injection of the RM1 prostate cancer cell line into male C57BL/6 mice, we observed a progressive increase in spontaneous guarding of the inoculated limb between 12 and 21 days post inoculation in tumor bearing compared to sham operated mice. Daily running wheel performance was evaluated as a measure of functional impairment and potentially movement evoked pain. We observed a progressive reduction in the distance traveled and percentage of time at optimal velocity between 12 and 21 days post inoculation in tumor bearing compared to sham operated mice. We utilized histological, radiographic and μCT analysis to examine tumor induced bone remodeling and observed osteolytic lesions as well as extra-periosteal aberrant bone formation in the tumor bearing femur, similar to clinical findings in patients with bone metastatic prostate cancer. Within the tumor bearing femur, we observed reorganization of blood vessels, macrophage and nerve fibers within the intramedullary space and periosteum adjacent to tumor cells. Tumor bearing mice displayed significant increases in the injury marker ATF3 and upregulation of the neuropeptides SP and CGRP in the ipsilateral DRG as well as increased measures of central sensitization and glial activation in the ipsilateral spinal cord. This immunocompetent mouse model will be useful when combined with cell type selective transgenic mice to examine tumor, immune cell and sensory neuron interactions in the bone microenvironment and their role in pain and disease progression associated with bone metastatic prostate cancer.

People have emotional reactions to the built environment, and the sonic environment is one of the major contributing factors of people’s experiences of places. It is useful for decision makers such ...as planners, architects, engineers and designers to understand the link between the soundscapes of built environments, and their resultant emotional reactions within users of those environments. This understanding can allow these decision makers to make better informed decisions about built environment design, and achieve the desired positive emotional responses from users. The issue under question, and the subject of this paper, was to understand and define the emotional dimensions of a soundscape, and then to explore how the resultant dimensional space could have a practical application in decision making. Through data generated through the Positive Soundscapes Project, a Principal Component Analysis was conducted to decompose descriptors of the urban soundscape into two independent emotional dimensions. It was found that the two independent emotional dimensions of a soundscape relate to its “Calmness” and “Vibrancy”, and these could be positioned to describe a 2-Dimensional (2-D) perceptual space. Listening evaluations were then conducted to explore how perceptions of different types of urban soundscape could be plotted with the 2D emotional perceptual space, and to illustrate how (i) introducing design interventions can change the position of soundscapes within this space and (ii) how metrics such as dB(A) do not necessarily correlate with calmness and vibrancy scores. Through illustrative examples, we suggest how such a 2-D perceptual space might have a practical use in the planning process, primarily as a means for presenting users’ perceptions of soundscapes in a simple, visual, and easy to compare manner, and for setting targets for current and future soundscape design. We conclude with recommendations for further work required to fully develop these emotional dimensions and the 2-D perceptual space into a useful tool.

This book develops a new account of Socratic method, based on a psychological model of Plato's dramatic depiction of Socrates' character and conduct. Socratic method is seen as a blend of three types ...of philosophical discourse: refutation, truth-seeking, and persuasion. Cain focuses on the persuasive features of the method since, in her view, it is this aspect of Socrates' method that best explains the content and the value of the dialectical arguments. Emphasizing the persuasive aspect of Socratic method helps us uncover the operative standards of dialectical argumentation in fifth-century Athens. Cain considers both the sophistic style of rhetoric and contentious debate in Socrates' time, and Aristotle's perspective on the techniques of argument and their purposes. An informal, pragmatic analysis of argumentation appropriate to the dialectical context is developed. We see that Socrates uses ambiguity and other strategic fallacies with purposeful play, and for moral ends. Taking specific examples of refutations from Plato's dialogues, Cain links the interlocutors' characters and situations with the dialectical argument that Socrates constructs to refute them. The merit of this interpretation is that it gives broad range, depth, and balance to Socrates' argumentative style; it also maintains a keen sensitivity to the interlocutors' emotional reactions, moral values, and attitudes. The book concludes with a discussion of the overall value, purpose, and success of Socratic method, and draws upon a Platonic/Socratic conception of the soul and a dialectical type of self-knowledge.
In order to determine how the interior of a car should sound, automotive manufacturers often rely on obtaining data from individual evaluations of vehicle sounds. Company identity could play a role ...in these appraisals, particularly when individuals are comparing cars from opposite ends of the performance spectrum. This research addressed the question: does company identity influence the evaluation of automotive sounds belonging to cars of a similar performance level and from the same market segment? Participants listened to car sounds from two competing manufacturers, together with control sounds. Before listening to each sound, participants were presented with the correct company identity for that sound, the incorrect identity or were given no information about the identity of the sound. The results showed that company identity did not influence appraisals of high performance cars belonging to different manufacturers. These results have positive implications for methodologies employed to capture the perceptions of individuals.
Statement of Relevance: A challenge in automotive design is to set appropriate targets for vehicle sounds, relying on understanding subjective reactions of individuals to such sounds. This paper assesses the role of company identity in influencing these subjective reactions and will guide sound evaluation studies, in which the manufacturer is often apparent.

Customer-facing train crew members have to follow strict procedures to guarantee that trains are safe and run on time. They are also responsible for revenue protection and customer care. Human ...factors and ergonomics research are instrumental to understand the safety-critical aspects and improve work. We bring user experience research and personas to describe how train crew perceive their routines and how new technology may impact them. We conducted 7 hours of interviews and 30 hours of shadowing observations with the train crew (N = 22) to provide an understanding of who are they and to define their experiences. We present the crew's current routines and created two personas to represent them. One is slightly reluctant to adopt the proposed technology, whereas the other is more accepting. Results indicate how such technology may affect crew work ergonomics and experiences, and suggest which valuable aspects should be maintained, for example the positive interactions with passengers.
Practitioner summary: This study investigated the work routines of the customer-facing train crew. Interviews and shadowing were conducted with 22 crew from a large operator in the UK. Personas were created to represent them. Results show their preferred activities and how these would be affected by the introduction of new technology.
