Therapeutic dressings to enhance burn wound repair and regeneration are required. Silk fibroin (SF), a natural protein, induces cell migration and serves as a biomaterial in various dressings. SF ...dressings usually contain α-helices and β-sheets. The former has been confirmed to improve cell proliferation and migration, but the wound healing effect and related mechanisms of β-sheet SF remain unclear. We investigated the effects of β-sheet SF in vivo and in vitro. Alcohol-treated α-helix SF transformed into the β-sheet form, which promoted granulation formation and re-epithelialization when applied as lyophilized SF dressing (LSFD) in a rat burn model. Our in vitro results showed that β-sheet SF increased human dermal fibroblast (HDF) migration and promoted the expression of extracellular matrix (ECM) proteins (fibronectin and type III collagen), matrix metalloproteinase-12, and the cell adhesion molecule, integrin β1, in rat granulation tissue and HDFs. This confirms the role of crosstalk between integrin β1 and ECM proteins in cell migration. In summary, we demonstrated that β-sheet SF facilitates tissue regeneration by modulating cell adhesion molecules in dermal fibroblasts. LSFD could find clinical application for burn wound regeneration. Moreover, β-sheet SF could be combined with anti-inflammatory materials, growth factors, or antibiotics to develop novel dressings.

Bladder cancer (BC) is the second most common urological tumour in Western countries. Approximately, 80% of patients with BC will present with non-muscle invasive bladder cancer (NMIBC), whereas a ...quarter will have muscle invasive disease (MIBC) at the time of BC diagnosis. However, patients with NMIBC are at risk of BC recurrence or progression into MIBC, and an MIBC prognosis is determined by the presence of progression and metastasis. Matrix metalloproteinase 2 (MMP2), a type of matrix metalloproteinase (MMP), plays a major role in tumour invasion and is well-characterized in BC prognosis. In BC, the mechanisms regulating MMP2 expression, and, in turn, promote cancer invasion, have hardly been explored. Thrombospondin-4 (THBS4/TSP4) is a matricellular glycoprotein that regulates multiple biological functions, including proliferation, angiogenesis, cell adhesion and extracellular matrix modelling. Based on the results of a meta-analysis in the Gene Expression Profiling Interactive Analysis 2 database, we observed that TSP4 expression levels were consistent with overall survival (OS) rate and BC progression, with the highest expression levels observed in the advanced stages of BC and associated with poor OS rate. In our pilot experiments, incubation with recombinant TSP4 promoted the migration and invasion in BC cells. Furthermore, MMP2 expression levels increased after recombinant TSP4 incubation. TSP4-induced-MMP2 expression and cell motility were regulated via the AKT signalling pathway. Our findings facilitate further investigation into TSP4 silencing-based therapeutic strategies for BC.

Purpose Cancer cells adapt to stress by activation of the autophagy pathway primed for survival. A high basal level of autophagic activity was found in human bladder cancer cell lines. We studied the ...significance of the phenomenon on cancer cell survival. Materials and Methods The immortalized human bladder epithelial cell line SV-HUC-1 and the human bladder cancer cell lines RT-4 and 5637 together with human bladder cancer specimens collected from patients were used. A commercially available bladder cancer microarray was applied to confirm the findings. LC3 (light chain-3) II protein detection was done to determine the presence of autophagy. Caspase 3 and DNA fragmentation was performed to detect apoptosis. Results Bladder cancer cell lines showed activated autophagic flux comparing to SV-HUC-1 cells, prostate cancer cells and breast cancer cells. Results were confirmed in human bladder cancer specimens. Autophagy inhibition by Baf (bafilomycin) A1, or by knockdown of ATG (autophagy related protein) 7 or 12 induced cytotoxicity in multiple human bladder cell lines. Induction of apoptosis was found in cells with autophagy inhibition. Although the disruption of mitochondria membrane potential or the generation of reactive oxygen species was detected in Baf A1 treated cells, intensity was mild and not thought to be related to apoptosis of bladder cancer cells. Conclusions Our results indicate that autophagy is required for the growth and survival of human bladder cancer cells.
Maize tassel detection is essential for future agronomic management in maize planting and breeding, with application in yield estimation, growth monitoring, intelligent picking, and disease ...detection. However, detecting maize tassels in the field poses prominent challenges as they are often obscured by widespread occlusions and differ in size and morphological color at different growth stages. This study proposes the SEYOLOX-tiny Model that more accurately and robustly detects maize tassels in the field. Firstly, the data acquisition method ensures the balance between the image quality and image acquisition efficiency and obtains maize tassel images from different periods to enrich the dataset by unmanned aerial vehicle (UAV). Moreover, the robust detection network extends YOLOX by embedding an attention mechanism to realize the extraction of critical features and suppressing the noise caused by adverse factors (e.g., occlusions and overlaps), which could be more suitable and robust for operation in complex natural environments. Experimental results verify the research hypothesis and show a mean average precision (mAP@0.5) of 95.0%. The mAP@0.5, mAP@0.5–0.95, mAP@0.5–0.95 (area=small), and mAP@0.5–0.95 (area=medium) average values increased by 1.5, 1.8, 5.3, and 1.7%, respectively, compared to the original model. The proposed method can effectively meet the precision and robustness requirements of the vision system in maize tassel detection.

Bladder cancer (BC) is the second most common urologic cancer in western countries. New strategies for managing high-grade muscle-invasive bladder cancer (MIBC) are urgently required because MIBC has ...a high risk of recurrence and poor survival. A growing body of evidence indicates that microRNA has potent antitumorigenic properties in various cancers, and thus, therapeutic strategies based on microRNA may show promising results in cancer therapy. Analysis of The Cancer Genome Atlas (TCGA) database indicated that hsa-miR-30a-3p is downregulated in human BC. Our in vitro investigation demonstrated that hsa-miR-30a-3p suppresses the expression of matrix metalloproteinase-2 (MMP-2) and MMP-9 and reduces the cell invasive potential of BC cells. Furthermore, hsa-miR-30a-3p directly targets ATG5, ATG12, and Beclin 1; this in turn improves the chemosensitivity of BC cells to cisplatin through the repression of protective autophagy. In a tumor-xenograft mice model, hsa-miR-30a-3p suppressed muscle invasion. Cotreatment with hsa-miR-30a-3p enhanced the antitumor effect of cisplatin in reducing tumor growth in BC. The current study provides a novel strategy of using hsa-miR-30a-3p as an adjuvant or replacement therapy in future BC treatment.
In this work, we demonstrate a novel In 0.52 Al 0.48 As based top-illuminated avalanche photodiode (APD), designed to circumvent the problem of serious bandwidth degradation under high gain (>100) ...and high power operation and significantly enhance the dynamic range in the established frequency modulated continuous wave (FMCW) lidar system. In our APD design, the carriers transiting through the dual multiplication (M-)layers are subjected to a stepped-up electric field profile, so they can be energized by the first step and propagate to the second step to trigger the avalanche processes. Such a cascade avalanche process leads to an ultra-high gain bandwidth product (460 GHz) with a 1 A/W responsivity at unit gain. Compared to the high-performance and commercial p-i-n PD and photo-receiver (PD + trans-impedance amplifier (TIA)) installed in the same lidar test bed, our demonstrated APD receiver (without TIA) has a larger S/N ratio under high operation gain (33 A/W) with less optical local-oscillator (LO) power required (0.25 vs. 0.5 mW), while exhibiting a wider dynamic range in each pixel. These advantages in turn lead to the construction of a better quality of 3-D lidar image by using the demonstrated APD.
This research deals with the single machine scheduling problem of minimizing the makespan with sequence dependent setup times and delayed precedence constraints. A makespan calculation model is first ...proposed. When given a feasible job sequence, the proposed model can calculate the makespan. Then a variable neighbourhood search (VNS) with four phases is proposed for optimizing the job sequence. The proposed VNS adopts five operations to search for new solutions, and modifies all solutions to satisfy precedence constraints. The proposed VNS will accept a worse solution over a better solution with a certain probability, in order to escape from a local optimum. The experimental results show that the proposed VNS provides the best results with less than 10 s of computation time. Therefore it is efficient and effective in solving the single machine scheduling problems.
When addressing the urgent task of improving vaccine supply chains, especially to accommodate the introduction of new vaccines, there is often a heavy emphasis on stationary storage. Currently, ...donations to vaccine supply chains occur largely in the form of storage equipment.
This study utilized a HERMES-generated detailed, dynamic, discrete event simulation model of the Niger vaccine supply chain to compare the impacts on vaccine availability of adding stationary cold storage versus transport capacity at different levels and to determine whether adding stationary storage capacity alone would be enough to relieve potential bottlenecks when pneumococcal and rotavirus vaccines are introduced by 2015.
Relieving regional level storage bottlenecks increased vaccine availability (by 4%) more than relieving storage bottlenecks at the district (1% increase), central (no change), and clinic (no change) levels alone. Increasing transport frequency (or capacity) yielded far greater gains (e.g., 15% increase in vaccine availability when doubling transport frequency to the district level and 18% when tripling). In fact, relieving all stationary storage constraints could only increase vaccine availability by 11%, whereas doubling the transport frequency throughout the system led to a 26% increase and tripling the frequency led to a 30% increase. Increasing transport frequency also reduced the amount of stationary storage space needed in the supply chain. The supply chain required an additional 61,269L of storage to relieve constraints with the current transport frequency, 55,255L with transport frequency doubled, and 51,791L with transport frequency tripled.
When evaluating vaccine supply chains, it is important to understand the interplay between stationary storage and transport. The HERMES-generated dynamic simulation model showed how augmenting transport can result in greater gains than only augmenting stationary storage and can reduce stationary storage needs.
