To biomechanically characterize superior capsule reconstruction (SCR) using fascia lata allograft, double-layer dermal allograft, and single-layer dermal allograft for a clinically relevant massive ...irreparable rotator cuff tear involving the entire supraspinatus and 50% of the infraspinatus tendons.
Eight cadaveric specimens were tested in 0°, 30°, and 60° abduction for (1) intact, (2) massive rotator cuff tear, (3) SCR using fascia lata, (4) SCR using double-layer dermis, and (5) SCR using single-layer dermis. Superior translation and subacromial contact pressure were measured. Statistical analysis was conducted using repeated measures ANOVA or paired t test with P < .05.
Massive rotator cuff tear significantly increased superior translation of the humeral head at all abduction angles (P < .05). At 0° abduction, all SCR conditions significantly decreased superior translation compared with the massive tear but did not restore translation (P < .05) to intact. Fascia lata and double-layer dermis SCR restored superior translation to intact at 30° and 60° of abduction, but single-layer dermis did not. Subacromial contact pressure at 0° of abduction significantly decreased with SCR with fascia lata and double-layer dermis compared with tear. At 30°, all SCR conditions significantly decreased subacromial contact pressure. Single-layer dermis graft thickness significantly decreased more than fascia lata during testing (P = .02).
For SCR tensioned at 20° glenohumeral abduction, all 3 graft types may restore superior translation and subacromial contact pressure depending on the glenohumeral abduction angle; fascia lata and double-layer dermis may be more effective than single-layer dermis.
If a dermal graft is to be used for SCR, consideration should be given to doubling the graft for increased thickness and better restorative biomechanical properties, which may improve clinical outcomes following SCR.
UVB radiation-induced signaling in mammalian cells involves two major pathways: one that is initiated through the generation of DNA photoproducts in the nucleus and a second one that occurs ...independently of DNA damage and is characterized by cell surface receptor activation. The chromophore for the latter one has been unknown. Here, we report that the UVB response involves tryptophan as a chromophore. We show that through the intracellular generation of photoproducts, such as the arylhydrocarbon receptor (AhR) ligand 6-formylindolo3,2-bcarbazole, signaling events are initiated, which are transferred to the nucleus and the cell membrane via activation of the cytoplasmatic AhR. Specifically, AhR activation by UVB leads to (i) transcriptional induction of cytochrome P450 1A1 and (ii) EGF receptor internalization with activation of the EGF receptor downstream target ERK1/2 and subsequent induction of cyclooxygenase-2. The role of the AhR in the UVB stress response was confirmed in vivo by studies employing AhR KO mice.

Canopy soils occur on tree branches throughout the temperate rainforests of the Pacific Northwest Coast and are recognized as a defining characteristic of these ecosystems. Certain tree species ...extend adventitious roots into these canopy soil environments. Yet, research on adventitious root-associated fungi remains limited. Our study used microscopy to compare fungal colonization intensity between canopy and forest floor roots of old-growth bigleaf maple (Acer macrophyllum) trees. Subsequently, two high-throughput sequencing platforms were used to explore the spatial and seasonal variation of root-associated fungi between the two soil environments over one year. We found that canopy and forest floor roots had similar colonization intensity and were associating with a diversity of arbuscular mycorrhizal fungi and other potential symbionts, many of which were resolved to species level. Soil environment and seasonality affected root-associated fungal community composition, and several fungal species were indicative of the canopy soil environment. In Washington State's (USA) temperate old-growth rainforests, these canopy soil environments host a unique suite of root-associated fungi. The presence of arbuscular mycorrhizae provides further evidence that adventitious roots form fungal associations to exploit canopy soils for resources, and there may be novel relationships forming with other fungi. These soils may be providing a redundancy compartment (i.e., "nutrient reserve"), imparting a resiliency to disturbances for certain old-growth trees.

• The influence of mature trees on colonization of Douglas-fir (Pseudotsuga menziesii) seedlings by ectomycorrhizal fungi (EMF) is not well understood. Here, the EMF communities of seedlings planted ...near and far from trees are compared with each other, with EMF of seedlings potted in field soils and with EMF of mature trees. • Seedlings were planted within 6 m, or beyond 16 m, from residual Douglas-fir trees in recently harvested green-tree retention units in Washington State, USA, or potted in soils gathered from near each residual tree. Mature tree roots were sampled by partly excavating the root system. The EMF communities were assessed by polymerase chain reaction-restriction fragment length polymorphism and sequence analysis of ribosomal RNA genes. • Seedlings near trees had higher species richness and diversity of EMF communities compared with seedlings far from trees. The EMF communities of seedlings near trees were more similar to those of mature trees, while seedlings far from trees were more similar to glasshouse seedlings. • By enhancing the EMF diversity of seedlings, residual trees may maintain or accelerate the re-establishment of mycorrhizal communities associated with mature forests.

Sarg. (red spruce) in the Appalachian Mountains of New England has been correlated with emissions transported from the Great Lakes region, including acids, metals, and ...oxidants. In 1994–1995, metal stress in red spruce foliage evidenced by phytochelatin concentrations increased with red spruce damage index in spruce-fir dominated stands in high elevation forests. In this study, we revisited those same forests after two decades to examine metal stress impacts on high-elevation forests following reductions in atmospheric pollutant loading. We measured metal concentrations in soils, lichens, and foliage, and concentrations of phytochelatin and its precursors in foliage of red spruce trees at 1000 m along a west–east transect from New York to New Hampshire, and along an 800–1000 m elevational transect on Whiteface Mountain, NY. Path analysis showed that foliar Hg had a direct positive effect on foliar phytochelatins, metal-binding peptides produced by the metal stress response in plants. Essential metals Cu and Zn decreased the concentration of Hg in foliage. However, we could not determine the relative importance of atmospheric vs soil pathways for metal exposure. While metal stress was still occurring on Whiteface Mountain in 2013, the overall visual health of red spruce trees across the region was significantly improved compared to 1993–1995. Thus, although metal stress is still measurable in red spruce, the physiological impact may be lessened by decreases in the deposition of metals and acids, thus providing evidence of positive forest health outcomes from improvements in regional air quality in the Northeastern US.

Much of the attention paid to jihadist groups in the greater Sahel region have focused on the Tuareg movements. More recently, however, members of the Fulani ethnic group have become much more ...significant in the larger jihadist movement in the region. The Fulani-dominated Katiba Macina and Ansarul Islam have conducted major attacks and created a significant level of civilian displacement. Many of the Fulani operations to date clearly are a continuation of their earlier conflicts both with regional governments and other ethnic groups. The increased identification of the Fulani in the jihadist movement also provides a further example of how preexisting group grievances can be operationalized by leaders of violent groups into an ideological movement.

Low-frequency sound from large vessels is a major, global source of ocean noise that can interfere with acoustic communication for a variety of marine animals. Changes in vessel activity provide ...opportunities to quantify relationships between vessel traffic levels and soundscape conditions in biologically important habitats. Using continuous deep-sea (890 m) recordings acquired ∼20 km (closest point of approach) from offshore shipping lanes, we observed reduction of low-frequency noise within Monterey Bay National Marine Sanctuary (California, United States) associated with changes in vessel traffic during the onset of the COVID-19 pandemic. Acoustic modeling shows that the recording site receives low-frequency vessel noise primarily from the regional shipping lanes rather than via the Sound Fixing and Ranging (SOFAR) channel. Monthly geometric means and percentiles of spectrum levels in the one-third octave band centered at 63 Hz during 2020 were compared with those from the same months of 2018–2019. Spectrum levels were persistently and significantly lower during February through July 2020, although a partial rebound in ambient noise levels was indicated by July. In what is now largely considered a footnote in history, Americans invaded Canada along the Niagara Frontier in 1866. The group behind the invasion—the Fenian Brotherhood—was formed in 1858 by Irish ...nationalists in New York City in order to fight for Irish independence from Britain. At the end of the American Civil War, Fenian leaders attempted to use Irish Americans, many of them combat veterans, to seize Canada and make it the "New Ireland" as a means to force the British from "old" Ireland. New York State was both the epicenter of Fenian leadership and a key support base and staging area for the military operations. Although relatively short-lived and with some of its military operations being somewhere between farce and tragedy, the Fenian Brotherhood had a very important impact on nineteenth-century New York and America, but remains largely forgotten. In Rebels on the Niagara Lawrence E. Cline examines not only the Fenian operations and their impact on Canada, but also the role the United States and New York played in both the initial support for the Fenian movement and its subsequent collapse in America.
Analgesic medications compounded for topical use are gaining popularity for the management of chronic pain. The advantages of topical pain medications include reduction of systemic adverse effects, ...improved patient acceptance, few drug interactions, ease of dose determination, avoidance of first-pass metabolism, and direct access to the target site. Compounded topical medications typically use a mixture of 3 or more single medications to achieve multiple complementary effects at lower doses of each individual medication. Herein, we review the mechanisms, adverse effects, and evidence for some of the most commonly used medications in topical compounds for pain management. Because more topical medications are used for chronic pain, dermatologists can expect an increase in irritant and allergic contact dermatitis related to these medications.
We present the first measurements of the double ratio of the polarization-transfer components (Px′/Pz′)p/(Px′/Pz′)s for knock-out protons from the s and p shells in C12 measured by the C12(e→,e′p→) ...reaction in quasi-elastic kinematics. The data are compared to theoretical predictions in the relativistic distorted-wave impulse approximation. Our results show that the differences between s- and p-shell protons, observed when compared at the same initial momentum (missing momentum), largely disappear when the comparison is done at the same proton virtuality. We observe no difference in medium modifications between protons from the s and p shells with the same virtuality in spite of the large differences in the respective nuclear densities.
