We used molecular subtyping to investigate an outbreak of listeriosis involving residents of 24 US states. We defined a case as infection with Listeria monocytogenes serotype 4b yielding one of ...several closely related patterns when subtyped by pulsed-field gel electrophoresis. Patients infected with strains yielding different patterns were used as controls. A total of 108 cases were identified with 14 associated deaths and four miscarriages or stillbirths. A case-control study implicated meat frankfurters as the likely source of infection (OR 17·3, 95% CI 2·4–160). The outbreak ended abruptly following a manufacturer-issued recall, and the outbreak strain was later detected in low levels in the recalled product. A second strain was recovered at higher levels but was not associated with human illness. Our findings suggest that L. monocytogenes strains vary widely in virulence and confirm that large outbreaks can occur even when only low levels of contamination are detected in sampled food. Standardized molecular subtyping and coordinated, multi-jurisdiction investigations can greatly facilitate detection and control of listeriosis outbreaks.

The literature base regarding the development of sporting talent is extensive, and includes empirical articles, reviews, position papers, academic books, governing body documents, popular books, ...unpublished theses and anecdotal evidence, and contains numerous models of talent development. With such a varied body of work, the task for researchers, practitioners and policy makers of generating a clear understanding of what is known and what is thought to be true regarding the development of sporting talent is particularly challenging. Drawing on a wide array of expertise, we address this challenge by avoiding adherence to any specific model or area and by providing a reasoned review across three key overarching topics: (a) the performer; (b) the environment; and (c) practice and training. Within each topic sub-section, we review and calibrate evidence by performance level of the samples. We then conclude each sub-section with a brief summary, a rating of the quality of evidence, a recommendation for practice and suggestions for future research. These serve to highlight both our current level of understanding and our level of confidence in providing practice recommendations, but also point to a need for future studies that could offer evidence regarding the complex interactions that almost certainly exist across domains.

Tractography based on non-invasive diffusion imaging is central to the study of human brain connectivity. To date, the approach has not been systematically validated in ground truth studies. Based on ...a simulated human brain data set with ground truth tracts, we organized an open international tractography challenge, which resulted in 96 distinct submissions from 20 research groups. Here, we report the encouraging finding that most state-of-the-art algorithms produce tractograms containing 90% of the ground truth bundles (to at least some extent). However, the same tractograms contain many more invalid than valid bundles, and half of these invalid bundles occur systematically across research groups. Taken together, our results demonstrate and confirm fundamental ambiguities inherent in tract reconstruction based on orientation information alone, which need to be considered when interpreting tractography and connectivity results. Our approach provides a novel framework for estimating reliability of tractography and encourages innovation to address its current limitations.
Introduction Infective endocarditis (IE) is a severe and highly prevalent infection among people who inject drugs (PWID). While short-term (30-day) outcomes are similar between PWID and non-PWID, the ...long-term outcomes among PWID after IE are poor, with 1-year mortality rates in excess of 25%. Novel clinical interventions are needed to address the unique needs of PWID with IE, including increasing access to substance use treatment and addressing structural barriers and social determinants of health. Methods and analysis PWID with IE will be connected to a multidisciplinary team that will transition with them from hospital to the community. The six components of the Second Heart Team are: (1) peer support worker with lived experience, (2) systems navigator, (3) addiction medicine physician, (4) primary care physician, (5) infectious diseases specialist, (6) cardiovascular surgeon. A convergent mixed-methods study design will be used to test the feasibility of this intervention. We will concurrently collect quantitative and qualitative data and 'mix' at the interpretation stage of the study to answer our research questions. Ethics and dissemination This study has been approved by the Hamilton Integrated Research Ethics Board (Project No. 7012). Dynamics of herbivore populations can be influenced both by density-dependent processes and climate. We used age-at-harvest data for adult female white-tailed deer (Odocoileus virginianus) collected ...over 23 years to estimate survival and reproduction by age class and to identify effects of environmental factors. The study population was located on Anticosti Island (QC, Canada), at the northern limit of the species' range; the population was at high density, and the landscape had scarce forage and abundant snow during winter. Despite severe environmental conditions, population growth apparently increased during the study; adult survival was similar to other populations, although reproduction appeared lower. Winter severity was not related to survival, but density affected adult female survival. Density at estrus was the main factor influencing reproduction of 2- and 3–4-year-olds and also affected reproduction of prime-aged females (5–9-yr-olds), but not of older females. Reproductive rate of younger females was influenced by environmental conditions in autumn, such as high density or snow conditions that limited forage availability. Reproductive success of 5–9- and ≥10-year-old females appeared dependent on spring conditions favoring high-quality forage, probably through effects on neonatal survival. Relative to other studies on northern ungulates, demographic processes in our study appeared to be more affected by autumn and spring climate, in addition to population density, than by winter climate. We thus propose that population density, as well as autumn and spring climate, should be considered in management strategies. Harvest data offered a unique opportunity to study forest ungulates, for which individual monitoring is rarely possible.

The objective of this study was to determine whether a suite of wood quantity and quality attributes of balsam fir and black spruce forests could be predicted using airborne laser scanner data. In ...situ estimates of stand structure and wood fibre attributes were derived from measurements at sample plots covering a wide range of forest conditions of insular Newfoundland. Models developed to predict field estimates explained 52–90 per cent of the variation in structure attributes, including mean and quadratic mean diameter at breast height, average and dominant height, stem density, basal area, total and merchantable volume and above-ground total biomass. Cross-validated root mean square errors were <24 per cent of mean values, with the exception of stem density, for which errors were 27–32 per cent. Models of fibre attributes explained 18–53 per cent of the variation in fibre length, wood density, radial diameter, coarseness, microfibril angle, modulus of elasticity, wall thickness and specific surface with cross-validated root mean square errors of <14 per cent of mean values. Similar results were achieved for fibre attribute models derived using geographic, climate and vegetation variables. The results demonstrate potential for inventory of quantity and quality attributes over a large region of boreal forests in Newfoundland, Canada.
The Pan-Andromeda Archaeological Survey is a survey of >400 square degrees centered on the Andromeda (M31) and Triangulum (M33) galaxies that has provided the most extensive panorama of an L galaxy ...group to large projected galactocentric radii. Here, we collate and summarize the current status of our knowledge of the substructures in the stellar halo of M31, and discuss connections between these features. We estimate that the 13 most distinctive substructures were produced by at least 5 different accretion events, all in the last 3 or 4 Gyr. We suggest that a few of the substructures farthest from M31 may be shells from a single accretion event. We calculate the luminosities of some prominent substructures for which previous estimates were not available, and we estimate the stellar mass budget of the outer halo of M31. We revisit the problem of quantifying the properties of a highly structured data set; specifically, we use the OPTICS clustering algorithm to quantify the hierarchical structure of M31's stellar halo and identify three new faint structures. M31's halo, in projection, appears to be dominated by two "mega-structures," which can be considered as the two most significant branches of a merger tree produced by breaking M31's stellar halo into increasingly smaller structures based on the stellar spatial clustering. We conclude that OPTICS is a powerful algorithm that could be used in any astronomical application involving the hierarchical clustering of points. The publication of this article coincides with the public release of all PAndAS data products.
RBM15, an RNA binding protein, determines cell-fate specification of many tissues including blood. We demonstrate that RBM15 is methylated by protein arginine methyltransferase 1 (PRMT1) at residue ...R578, leading to its degradation via ubiquitylation by an E3 ligase (CNOT4). Overexpression of PRMT1 in acute megakaryocytic leukemia cell lines blocks megakaryocyte terminal differentiation by downregulation of RBM15 protein level. Restoring RBM15 protein level rescues megakaryocyte terminal differentiation blocked by PRMT1 overexpression. At the molecular level, RBM15 binds to pre-messenger RNA intronic regions of genes important for megakaryopoiesis such as GATA1, RUNX1, TAL1 and c-MPL. Furthermore, preferential binding of RBM15 to specific intronic regions recruits the splicing factor SF3B1 to the same sites for alternative splicing. Therefore, PRMT1 regulates alternative RNA splicing via reducing RBM15 protein concentration. Targeting PRMT1 may be a curative therapy to restore megakaryocyte differentiation for acute megakaryocytic leukemia.
