The forecasting of high-dimensional, spatiotemporal nonlinear systems has made tremendous progress with the advent of model-free machine learning techniques. However, in real systems it is not always ...possible to have all the information needed; only partial information is available for learning and forecasting. This can be due to insufficient temporal or spatial samplings, to inaccessible variables, or to noisy training data. Here, we show that it is nevertheless possible to forecast extreme event occurrences in incomplete experimental recordings from a spatiotemporally chaotic microcavity laser using reservoir computing. Selecting regions of maximum transfer entropy, we show that it is possible to get higher forecasting accuracy using nonlocal data vs local data, thus allowing greater warning times of at least twice the time horizon predicted from the nonlinear local Lyapunov exponent.

Antimalarial drugs that can block the transmission of Plasmodium gametocytes to mosquito vectors would be highly beneficial for malaria elimination efforts. Identifying transmission-blocking drugs ...currently relies on evaluation of their activity against gametocyte-producing laboratory parasite strains and would benefit from a testing pipeline with genetically diverse field isolates. The aims of this study were to develop a pipeline to test drugs against P. falciparum gametocyte field isolates and to evaluate the transmission-blocking activity of a set of novel compounds. Two assays were designed so they could identify both the overall transmission-blocking activity of a number of marketed and experimental drugs by direct membrane feeding assays (DMFA), and then also discriminate between those that are active against the gametocytes (gametocyte killing or sterilizing) or those that block development in the mosquito (sporontocidal). These DMFA assays used venous blood samples from naturally infected Plasmodium falciparum gametocyte carriers and locally reared Anopheles gambiae s.s. mosquitoes. Overall transmission-blocking activity was assessed following a 24 hour incubation of compound with gametocyte infected blood (TB-DMFA). Sporontocidal activity was evaluated following addition of compound directly prior to feeding, without incubation (SPORO-DMFA); Gametocyte viability was retained during 24-hour incubation at 37°C when gametocyte infected red blood cells were reconstituted in RPMI/serum. Methylene-blue, MMV693183, DDD107498, atovaquone and P218 showed potent transmission-blocking activity in the TB-DMFA, and both atovaquone and the novel antifolate P218 were potent inhibitors of sporogonic development in the SPORO-DMA. This work establishes a pipeline for the integral use of field isolates to assess the transmission-blocking capacity of antimalarial drugs to block transmission that should be validated in future studies.

The pioneering paper 'Optical rogue waves' by Solli et al (2007 Nature 450 1054) started the new subfield in optics. This work launched a great deal of activity on this novel subject. As a result, ...the initial concept has expanded and has been enriched by new ideas. Various approaches have been suggested since then. A fresh look at the older results and new discoveries has been undertaken, stimulated by the concept of 'optical rogue waves'. Presently, there may not by a unique view on how this new scientific term should be used and developed. There is nothing surprising when the opinion of the experts diverge in any new field of research. After all, rogue waves may appear for a multiplicity of reasons and not necessarily only in optical fibers and not only in the process of supercontinuum generation. We know by now that rogue waves may be generated by lasers, appear in wide aperture cavities, in plasmas and in a variety of other optical systems. Theorists, in turn, have suggested many other situations when rogue waves may be observed. The strict definition of a rogue wave is still an open question. For example, it has been suggested that it is defined as 'an optical pulse whose amplitude or intensity is much higher than that of the surrounding pulses'. This definition (as suggested by a peer reviewer) is clear at the intuitive level and can be easily extended to the case of spatial beams although additional clarifications are still needed. An extended definition has been presented earlier by N Akhmediev and E Pelinovsky (2010 Eur. Phys. J. Spec. Top. 185 1-4). Discussions along these lines are always useful and all new approaches stimulate research and encourage discoveries of new phenomena. Despite the potentially existing disagreements, the scientific terms 'optical rogue waves' and 'extreme events' do exist. Therefore coordination of our efforts in either unifying the concept or in introducing alternative definitions must be continued. From this point of view, a number of the scientists who work in this area of research have come together to present their research in a single review article that will greatly benefit all interested parties of this research direction. Whether the authors of this 'roadmap' have similar views or different from the original concept, the potential reader of the review will enrich their knowledge by encountering most of the existing views on the subject. Previously, a special issue on optical rogue waves (2013 J. Opt. 15 060201) was successful in achieving this goal but over two years have passed and more material has been published in this quickly emerging subject. Thus, it is time for a roadmap that may stimulate and encourage further research.
Extreme events such as rogue waves in optics and fluids are often associated with the merging dynamics of coherent structures. We present experimental and numerical results on the physics of extreme ...event appearance in a spatially extended semiconductor microcavity laser with an intracavity saturable absorber. This system can display deterministic irregular dynamics only, thanks to spatial coupling through diffraction of light. We have identified parameter regions where extreme events are encountered and established the origin of this dynamics in the emergence of deterministic spatiotemporal chaos, through the correspondence between the proportion of extreme events and the dimension of the strange attractor.

Heterologous prime boost immunization with chimpanzee adenovirus 63 (ChAd63) and Modified Vaccinia Virus Ankara (MVA) vectored vaccines is a strategy previously shown to provide substantial ...protective efficacy against P. falciparum infection in United Kingdom adult Phase IIa sporozoite challenge studies (approximately 20-25% sterile protection with similar numbers showing clear delay in time to patency), and greater point efficacy in a trial in Kenyan adults.
We conducted the first Phase IIb clinical trial assessing the safety, immunogenicity and efficacy of ChAd63 MVA ME-TRAP in 700 healthy malaria exposed children aged 5-17 months in a highly endemic malaria transmission area of Burkina Faso.
ChAd63 MVA ME-TRAP was shown to be safe and immunogenic but induced only moderate T cell responses (median 326 SFU/106 PBMC (95% CI 290-387)) many fold lower than in previous trials. No significant efficacy was observed against clinical malaria during the follow up period, with efficacy against the primary endpoint estimate by proportional analysis being 13.8% (95%CI -42.4 to 47.9) at sixth month post MVA ME-TRAP and 3.1% (95%CI -15.0 to 18.3; p = 0.72) by Cox regression.
This study has confirmed ChAd63 MVA ME-TRAP is a safe and immunogenic vaccine regimen in children and infants with prior exposure to malaria. But no significant protective efficacy was observed in this very highly malaria-endemic setting.

ABSTRACT
Observational analyses of changing climate extremes over the West Africa region have been limited by the availability of long and high‐quality datasets. To help address this gap, a climate ...extremes indices workshop was held in the Gambia in December 2011 with participants from 14 West African countries. The resulting analysis utilized 15 annual indices derived from observed daily temperatures and 10 annual indices derived from observed daily precipitation. The analysis was conducted for 166 meteorological stations in 13 countries for 2 periods: 1960–2010 and 1981–2010. The analyses of trends in the annual mean temperature indices have identified statistically significant increases of 0.16 °C/decade and 0.28 °C/decade for mean annual maximum and mean annual minimum temperatures, respectively, averaged over all available land stations in the region during the last 50 years. The seasonal‐temperature‐related indices show significant patterns of warming in all seasons. The annual mean of daily minimum temperature has increased more than the annual mean of daily maximum temperature leading to a decreasing trend in the diurnal temperature range. Warm days and warm nights have become more frequent, and cold days and cold nights have become less frequent. The analyses of precipitation‐based indices indicate spatially non‐coherent changes throughout the study area with few statistically significant trends over the longer period. Exceptions to this are the simple daily intensity index and maximum 5‐day precipitation, which show significant increasing regional trends over both the shorter and longer periods. Additionally, over the recent period (1981–2010) most of the precipitation related indices show significant trends towards wetter conditions. However, this period of increased rainfall follows a decade of significantly drier conditions in the region – it is not clear whether the recent upward trends reflect the ‘recovery’ from this long drought period or represents a long‐term response to warming.
Climate extremes indices were derived from daily temperature and precipitation data from 166 observing stations in 13 West African countries, and combined to provide a regional assessment of changing extremes for 1960–2010 and 1981–2010. Statistically significant warming trends indicate more frequent warm days and nights and less frequent cold days and nights. Precipitation‐based indices indicate weak spatial patterns with few significant trends over 1960–2010, but over 1981–2010 show significant trends towards wetter conditions.
