In December 2019, cases of unidentified pneumonia with a history of exposure in the Huanan Seafood Market were reported in Wuhan, Hubei Province. A novel coronavirus, SARS-CoV-2, was identified to be ...accountable for this disease. Human-to-human transmission is confirmed, and this disease (named COVID-19 by World Health Organization (WHO)) spread rapidly around the country and the world. As of 18 February 2020, the number of confirmed cases had reached 75,199 with 2009 fatalities. The COVID-19 resulted in a much lower case-fatality rate (about 2.67%) among the confirmed cases, compared with Severe Acute Respiratory Syndrome (SARS) and Middle East Respiratory Syndrome (MERS). Among the symptom composition of the 45 fatality cases collected from the released official reports, the top four are fever, cough, short of breath, and chest tightness/pain. The major comorbidities of the fatality cases include hypertension, diabetes, coronary heart disease, cerebral infarction, and chronic bronchitis. The source of the virus and the pathogenesis of this disease are still unconfirmed. No specific therapeutic drug has been found. The Chinese Government has initiated a level-1 public health response to prevent the spread of the disease. Meanwhile, it is also crucial to speed up the development of vaccines and drugs for treatment, which will enable us to defeat COVID-19 as soon as possible.
The focus of this study was to investigate the expression status of Circ‐vimentin (VIM) and further analyze its pathogenesis and clinical significance in acute myeloid leukemia (AML) patients. ...Real‐time quantitative polymerase chain reaction was carried on Circ‐VIM in 113 AML patients and 42 healthy controls. Circ‐VIM was significantly upregulated in AML compared with control and was positively correlated with white blood cells (WBC) count. Receiver operating characteristic curve analysis indicated that the performance of Circ‐VIM expression could serve as a promising biomarker for differentiating AML patients from controls. Significant correlations of Circ‐VIM expression were found with WBC and French–American–British classifications. Survival analyses further showed that over‐expressed Circ‐VIM were associated with markedly shorter overall survival (OS) and leukemia‐free survival (LFS) in whole‐cohort AML, nonacute promyelocytic leukemia AML and cytogenetically normal‐AML patients. Multivariate analysis also disclosed that Circ‐VIM over‐expression was an independent poor prognostic factor for OS and LFS in AML patients. Remarkably, Pearson correlation analysis evidenced that the expression of Circ‐VIM was positively correlated with VIM expression in all AML patients. These results indicated that overexpression Circ‐VIM could serve as a significant biomarker.

A fiber-optic surface plasmon resonance (SPR) sensor for temperature detection has been proposed by utilizing a thermosensitive liquid as the intermediate and combining with the fiber SPR structure. ...The sensing element of the sensor has been fabricated by packaging the fiber probe coated with a silver layer into a capillary filled with thermosensitive anhydrous ethanol. This packaging can protect the metal layer from oxidation and damage. Moreover, this proposed sensor achieves a sensitivity of 1.5745 nm/°C, which is much higher than that of the traditional fiber SPR sensor according to the comparative experiments.
The Foxo3 gene, belonging to the forkhead family, is one of the classes of transcription factors characterized by a forkhead DNA-binding domain, which usually considered being a cancer suppressor ...gene. Circ-Foxo3 is a circular structure which connects the 3'end to the 5'end. Scholars detected that circ-Foxo3 could compete with Foxo3 for binding to some miRNAs.
In this study, we will test the expression of Foxo3 and circ-Foxo3 in de novo acute myeloid leukemia (AML) patients to explore the relationship between Foxo3 gene and circ-Foxo3. All the de novo AML samples and normal control samples was measured by real-time quantitative PCR. A receiver operating characteristic curve was conducted to differentiate AML patients from control people. Association of Foxo3 expression and overall survival was conducted by Kaplan-Meier survival analysis.
We found that the expression of Foxo3 gene in de novo patients was significantly lower than control samples (P = 0.009). Meanwhile, circ-Foxo3 also expressed lower in de novo AML patients than in control samples (P = 0.040). In different classifications, this trend could be observed more remarkably. In non-M3 patients, the Foxo3 high patients' survival time was longer than Foxo3 low patients (P = 0.002). Besides, in non-favorable risk groups, patients with low expression of Foxo3 had longer survival time than Foxo3 high patients (P = 0.004). Furthermore, in normal Karyotypic patients, the overall survival time of patients with high-expressed Foxo3 was significantly longer than those with low expression (P = 0.034). Besides, Pearson analysis was also conducted between these two genes in AML patients. Results revealed that they were positively correlated (R = 0.63, P < 0.001).
In conclusion, we found that low expression of circ-Foxo3 and Foxo3 were frequent in AML patients, and patients with high expression of Foxo3 often had a trend of better prognosis.

Type 2 diabetes mellitus and metabolic disorders have become an epidemic globally. However, the pathogenesis remains largely unclear and the prevention and treatment are still limited. In addition to ...environmental factors during adulthood, early life is the critical developmental window with high tissue plasticity, which might be modified by external environmental cues. Substantial evidence has demonstrated the vital role of early-life nutrition in programming the metabolic disorders in later life. In this review, we aim to overview the concepts of fetal programming and investigate the effects of early-life nutrition on energy metabolism in later life and the potential epigenetic mechanism. The related studies published on PubMed database up to March 2020 were included. The results showed that both maternal overnutrition and undernutrition increased the riskes of metabolic disorders in offspring and epigenetic modifications, including DNA methylation, miRNAs, and histone modification, might be the vital mediators. The beneficial effects of early-life lifestyle modifications as well as dietary and nutritional interventions on these deleterious metabolic remolding were initially observed. Overall, characterizing the early-life malnutrition that reshapes metabolic disease trajectories may yield novel targets for early prevention and intervention and provide a new point of view to the energy metabolism.
Dengue virus (DENV) has become a global health threat with about half of the world's population at risk of infection. Since there is no specific efficient medicine in treatment, it is urgent to develop vaccines to prevent infection and disease progression. Currently, only a live attenuated vaccine, chimeric yellow fever 17D-tetravalent dengue vaccine (CYD-TDV), has been licensed for clinical use in some countries, and many candidate vaccines are still under research and development. This review discusses the progress, strengths, and weaknesses of the five types of vaccines including live attenuated vaccine, inactivated virus vaccine, recombinant subunit vaccine, viral vectored vaccine, and DNA vaccine.
Obesity has become a global health problem. Lifestyle modification and medical treatment only appear to yield short-term weight loss. Roux-en-Y gastric bypass (RYGB) is the most popular bariatric ...procedure, and it sustains weight reduction and results in the remission of obesity-associated comorbidities for obese individuals. However, patients who undergo this surgery may develop hypoglycemia. To date, the diagnosis is challenging and the prevalence of post-RYGB hypoglycemia (PRH) is unclear. RYGB alters the anatomy of the upper gastrointestinal tract and has a combined effect of caloric intake restriction and nutrient malabsorption. Nevertheless, the physiologic changes after RYGB are complex. Although hyperinsulinemia, incretin effects, dysfunction of β-cells and α-cells, and some other factors have been widely investigated and are reported to be possible mediators of PRH, the pathogenesis is still not completely understood. In light of the important role of the gut microbiome in metabolism, we hypothesized that the gut microbiome might also be a critical link between RYGB and hypoglycemia. In this review, we mainly highlight the current possible factors predisposing individuals to PRH, particularly related to the gut microbiota, which may yield significant insights into the intestinal regulation of glucose metabolic homeostasis and provide novel clues to improve the treatment of type 2 diabetes mellitus.
Heparin is an important anticoagulant drug, and microbial heparin biosynthesis is a potential alternative to animal-derived heparin production. However, effectively using heparin synthesis enzymes ...faces challenges, especially with microbial recombinant expression of active heparan sulfate N-deacetylase/N-sulfotransferase. Here, we introduce the monosaccharide N-trifluoroacetylglucosamine into Escherichia coli K5 to facilitate sulfation modification. The Protein Repair One-Stop Service-Focused Rational Iterative Site-specific Mutagenesis (PROSS-FRISM) platform is used to enhance sulfotransferase efficiency, resulting in the engineered NST-M8 enzyme with significantly improved stability (11.32-fold) and activity (2.53-fold) compared to the wild-type N-sulfotransferase. This approach can be applied to engineering various sulfotransferases. The multienzyme cascade reaction enables the production of active heparin from bioengineered heparosan, demonstrating anti-FXa (246.09 IU/mg) and anti-FIIa (48.62 IU/mg) activities. This study offers insights into overcoming challenges in heparin synthesis and modification, paving the way for the future development of animal-free heparins using a cellular system-based semisynthetic strategy.
Research developments of the photonic crystal fiber based surface plasmon resonance (PCF-SPR) chemical sensors were intensively reviewed. Photonic crystal fibers, such as the microstructured optical ...fiber, the photonic bandgap fiber and the Bragg fiber with various structures were applied to the SPR sensors, including fuse-tapered fiber structure, D-type fiber structure and cladding-off fiber structure. Those sensors were classified as three kinds of configurations which were respectively based on the inner metal layer, the metallic nanowire and the outer metal film. What's more, the principles, superiorities and problems of the PCF-SPR sensors were also discussed in detail.

Objective: SIRT1 is an NAD+‐dependent deacetylase and its enzymatic activity may be regulated by cellular energy. SIRT1 overexpression reduces the level of oxygen consumption, which is correlative ...with nonalcoholic fatty liver disease (NAFLD). To elucidate the role of SIRT1 on the development of NAFLD, we investigated the expression of SIRT1 in NAFLD induced by high‐fat diet in rats and the effects of calorie restriction.
Methods: Thirty‐one male Wistar rats were divided at random into four groups. The rats in the normal control group NC (n=7) and in the NAFLD model group HF (n=9) were fed ad libitum with normal chow and high‐fat diet, respectively, for 3 months, the rats in the calorie restriction (CR) group HCR (n=9) were fed with a high‐fat diet for 2 months and then 60% CR with normal chow for 1 month, and the rats in group CRH (n=6) were firstly fed with 60% CR with normal chow for 1 month and then fed a high‐fat diet for 2 months. At the end of the experiment, some parameters and expressions of SIRT1 were detected.
Results: The rats in group HF displayed NAFLD. Compared with group NC, the expression of SIRT1 protein was significantly decreased (P<0.01). However, the lower body weight and visceral fat mass of rats in group HCR were showed. Compared with group HF, CR increased the expression of SIRT1 in liver significantly (P<0.01). Consequently, the ultramicropathology changes of NAFLD prominently improved in this group. Meanwhile, the rats in group CRH displayed higher expression of SIRT1 protein and very gentle pathology changes of NAFLD.
Conclusion: The expression of SIRT1 is reduced significantly in NAFLD induced by high‐fat diet in rats. CR increase‐SIRT1 protein expression may be an important mechanism by which CR improves NAFLD.
