Dabrafenib is a potent BRAF-kinase inhibitor. Its activity was evaluated on 40 consecutive metastatic melanoma patients (pts) harboring the V600BRAF mutations. Dabrafenib was administered orally at ...the dosage of 150 mg b.i.d. daily. ORR was 82%, with 7% CR, 62% PR, 13% SD and 18% PD. The median PFS and OS were seven and 17 months, respectively (median follow-up: 8.5 months). Increased risk of progression was found in pts with elevated LDH, ECOG PS >1 and more than two metastatic sites. Grade 3-4 adverse events were recorded in 4 pts. In this retrospective analysis, Dabrafenib confirmed its role as the standard clinical option in metastatic melanoma pts.

Adverse outcome pathway (AOP) is a conceptual framework that links a molecular initiating event (MIE) via intermediate key events (KEs) with adverse effects (adverse outcomes, AO) relevant for risk ...assessment, through defined KE relationships (KERs). The aim of the present work is to describe a linear AOP, supported by experimental data, for skeletal craniofacial defects as the AO. This AO was selected in view of its relative high incidence in humans and the suspected relation to chemical exposure. We focused on inhibition of CYP26, a retinoic acid (RA) metabolizing enzyme, as MIE, based on robust previously published data. Conazoles were selected as representative stressors. Intermediate KEs are RA disbalance, aberrant HOX gene expression, disrupted specification, migration, and differentiation of neural crest cells, and branchial arch dysmorphology.
We described the biological basis of the postulated events and conducted weight of evidence (WoE) assessments. The biological plausibility and the overall empirical evidence were assessed as high and moderate, respectively, the latter taking into consideration the moderate evidence for concordance of dose-response and temporal relationships. Finally, the essentiality assessment of the KEs, considered as high, supported the robustness of the presented AOP. This AOP, which appears of relevance to humans, thus contributes to mechanistic underpinning of selected test methods, thereby supporting their application in integrated new approach test methodologies and strategies and application in a regulatory context.

The trilith consists of two vertical elements (columns) supporting a horizontal element (lintel). The understanding of the dynamic behaviour of triliths is an important step towards their ...preservation and starts with the knowledge of the dynamics of rigid blocks. A passive method based on a dynamic mass absorber is used to protect a trilith from overturning. The protection system is modelled as a pendulum, hinged on the lintel, with the mass lumped at the end. The equations of rocking motion, uplift and the impact conditions are obtained for the coupled system trilith-mass absorber. An extensive parametric analysis is performed with the aim to compare the behaviour of the system with and without the pendulum, under impulsive one-sine (or one-cosine) base excitations. In order to point out the effectiveness of the protection system, overturning spectra, providing the amplitude of the excitation versus its frequency, are obtained. The pendulum mass absorber results effective in avoiding overturning in specific ranges of the frequency of the excitation. However, outside these ranges the mass absorber never compromises the safety of the trilith.
