The landforms of northern Gale crater on Mars expose thick sequences of sedimentary rocks. Based on images obtained by the Curiosity rover, we interpret these outcrops as evidence for past fluvial, ...deltaic, and lacustrine environments. Degradation of the crater wall and rim probably supplied these sediments, which advanced inward from the wall, infilling both the crater and an internal lake basin to a thickness of at least 75 meters. This intracrater lake system probably existed intermittently for thousands to millions of years, implying a relatively wet climate that supplied moisture to the crater rim and transported sediment via streams into the lake basin. The deposits in Gale crater were then exhumed, probably by wind-driven erosion, creating Aeolis Mons (Mount Sharp).

Abstract
We perform a joint analysis of intrinsic alignments and cosmology using tomographic weak lensing, galaxy clustering, and galaxy–galaxy lensing measurements from Year 1 (Y1) of the Dark ...Energy Survey. We define early- and late-type subsamples, which are found to pass a series of systematics tests, including for spurious photometric redshift error and point spread function correlations. We analyse these split data alongside the fiducial mixed Y1 sample using a range of intrinsic alignment models. In a fiducial non-linear alignment model analysis, assuming a flat Λ cold dark matter cosmology, we find a significant difference in intrinsic alignment amplitude, with early-type galaxies favouring $A_\mathrm{IA} = 2.38^{+0.32}_{-0.31}$ and late-type galaxies consistent with no intrinsic alignments at $0.05^{+0.10}_{-0.09}$. The analysis is repeated using a number of extended model spaces, including a physically motivated model that includes both tidal torquing and tidal alignment mechanisms. In multiprobe likelihood chains in which cosmology, intrinsic alignments in both galaxy samples and all other relevant systematics are varied simultaneously, we find the tidal alignment and tidal torquing parts of the intrinsic alignment signal have amplitudes $A_1 = 2.66 ^{+0.67}_{-0.66}$, $A_2=-2.94^{+1.94}_{-1.83}$, respectively, for early-type galaxies and $A_1 = 0.62 ^{+0.41}_{-0.41}$, $A_2 = -2.26^{+1.30}_{-1.16}$ for late-type galaxies. In the full (mixed) Y1 sample the best constraints are $A_1 = 0.70 ^{+0.41}_{-0.38}$, $A_2 = -1.36 ^{+1.08}_{-1.41}$. For all galaxy splits and IA models considered, we report cosmological parameter constraints consistent with the results of the main DES Y1 cosmic shear and multiprobe cosmology papers.
• Background Electrical capacitance, measured between an electrode inserted at the base of a plant and an electrode in the rooting substrate, is often linearly correlated with root mass. Electrical ...capacitance has often been used as an assay for root mass, and is conventionally interpreted using an electrical model in which roots behave as cylindrical capacitors wired in parallel. Recent experiments in hydroponics show that this interpretation is incorrect and a new model has been proposed. Here, the new model is tested in solid substrates. • Methods The capacitances of compost and soil were determined as a function of water content, and the capacitances of cereal plants growing in sand or potting compost in the glasshouse, or in the field, were measured under contrasting irrigation regimes. • Key Results Capacitances of compost and soil increased with increasing water content. At water contents approaching field capacity, compost and soil had capacitances at least an order of magnitude greater than those of plant tissues. For plants growing in solid substrates, wetting the substrate locally around the stem base was both necessary and sufficient to record maximum capacitance, which was correlated with stem cross-sectional area: capacitance of excised stem tissue equalled that of the plant in wet soil. Capacitance measured between two electrodes could be modelled as an electrical circuit in which component capacitors (plant tissue or rooting substrate) are wired in series. • Conclusions The results were consistent with the new physical interpretation of plant capacitance. Substrate capacitance and plant capacitance combine according to standard physical laws. For plants growing in wet substrate, the capacitance measured is largely determined by the tissue between the surface of the substrate and the electrode attached to the plant. Whilst the measured capacitance can, in some circumstances, be correlated with root mass, it is not a direct assay of root mass.

Schwann cells are essential for the maintenance and function of motor neurons, axonal networks, and the neuromuscular junction. In amyotrophic lateral sclerosis, where motor neuron function is ...progressively lost, Schwann cell function may also be impaired. Recently, important signaling and potential trophic activities of Schwann cell-derived exosomal vesicles have been reported. This case report describes the treatment of a patient with advanced amyotrophic lateral sclerosis using serial intravenous infusions of allogeneic Schwann cell-derived exosomal vesicles, marking, to our knowledge, the first instance of such treatment. An 81-year-old male patient presented with a 1.5-year history of rapidly progressive amyotrophic lateral sclerosis. After initial diagnosis, the patient underwent a combination of generic riluzole, sodium phenylbutyrate for the treatment of amyotrophic lateral sclerosis, and taurursodiol. The patient volunteered to participate in an FDA-approved single-patient expanded access treatment and received weekly intravenous infusions of allogeneic Schwann cell-derived exosomal vesicles to potentially restore impaired Schwann cell and motor neuron function. We confirmed that cultured Schwann cells obtained from the amyotrophic lateral sclerosis patient via sural nerve biopsy appeared impaired (senescent) and that exposure of the patient’s Schwann cells to allogeneic Schwann cell-derived exosomal vesicles, cultured expanded from a cadaver donor improved their growth capacity in vitro. After a period of observation lasting 10 weeks, during which amyotrophic lateral sclerosis Functional Rating Scale-Revised and pulmonary function were regularly monitored, the patient received weekly consecutive infusions of 1.54 × 10 12 (×2), and then consecutive infusions of 7.5 × 10 12 (×6) allogeneic Schwann cell-derived exosomal vesicles diluted in 40 mL of Dulbecco’s phosphate-buffered saline. None of the infusions were associated with adverse events such as infusion reactions (allergic or otherwise) or changes in vital signs. Clinical lab serum neurofilament and cytokine levels measured prior to each infusion varied somewhat without a clear trend. A more sensitive in-house assay suggested possible inflammasome activation during the disease course. A trend for clinical stabilization was observed during the infusion period. Our study provides a novel approach to address impaired Schwann cells and possibly motor neuron function in patients with amyotrophic lateral sclerosis using allogeneic Schwann cell-derived exosomal vesicles. Initial findings suggest that this approach is safe.
Context. Determination of cluster masses is a fundamental tool for cosmology. Comparing mass estimates obtained by different probes allows to understand possible systematic uncertainties. Aims. The ...cluster Abell 315 is an interesting test case, since it has been claimed to be underluminous in X-ray for its mass (determined via kinematics and weak lensing). We have undertaken new spectroscopic observations with the aim of improving the cluster mass estimate, using the distribution of galaxies in projected phase space. Methods. We identified cluster members in our new spectroscopic sample. We estimated the cluster mass from the projected phase-space distribution of cluster members using the MAMPOSSt method. In doing this estimate we took into account the presence of substructures that we were able to identify. Results. We identify several cluster substructures. The main two have an overlapping spatial distribution, suggesting a (past or ongoing) collision along the line-of-sight. After accounting for the presence of substructures, the mass estimate of Abell 315 from kinematics is reduced by a factor 4, down to M200 = 0.8+0.6-0.4 × 1014M⊙. We also find evidence that the cluster mass concentration is unusually low, c200 ≡ r200/r-2 ≲ 1. Using our new estimate of c200 we revise the weak lensing mass estimate down to M200 = 1.8+1.7-0.9 × 1014M⊙. Our new mass estimates are in agreement with that derived from the cluster X-ray luminosity via a scaling relation, M200 = 0.9 ± 0.2 × 1014M⊙. Conclusions. Abell 315 no longer belongs to the class of X-ray underluminous clusters. Its mass estimate was inflated by the presence of an undetected subcluster in collision with the main cluster. Whether the presence of undetected line-of-sight structures can be a general explanation for all X-ray underluminous clusters remains to be explored using a statistically significant sample.

The laser-plasma accelerator (LPA) presently provides electron beams with a typical current of a few kA, a bunch length of a few fs, energy in the few hundred MeV to several GeV range, a divergence ...of typically 1 mrad, an energy spread of the order of 1%, and a normalized emittance of the order of π.mm.mrad. One of the first applications could be to use these beams for the production of radiation: undulator emission has been observed but the rather large energy spread (1%) and divergence (1 mrad) prevent straightforward free-electron laser (FEL) amplification. An adequate beam manipulation through the transport to the undulator is then required. The key concept proposed here relies on an innovative electron beam longitudinal and transverse manipulation in the transport towards an undulator: a 'demixing' chicane sorts the electrons according to their energy and reduces the spread from 1% to one slice of a few ‰ and the effective transverse size is maintained constant along the undulator (supermatching) by a proper synchronization of the electron beam focusing with the progress of the optical wave. A test experiment for the demonstration of FEL amplification with an LPA is under preparation. Electron beam transport follows different steps with strong focusing with permanent magnet quadrupoles of variable strength, a demixing chicane with conventional dipoles, and a second set of quadrupoles for further focusing in the undulator. The FEL simulations and the progress of the preparation of the experiment are presented.

The aim of this article is to present the experiences of our center certified by the ERAS® Society for colorectal resections 18 months after successful implementation.
Since the beginning of the certification 261 patients have been treated in our clinic according to the specifications of the ERAS® concept. As a comparison group the last 50 patients prior to implementation were evaluated in terms of compliance with ERAS® requirements, length of hospital stay and readmission rate, the need for care in an intensive or intermediate care ward, the number of necessary reoperations and the complication rate.
Compliance increased from 39.3% preERAS® to 81.1% after ERAS® implementation (p < 0.001). At the same time the length of stay of ERAS® patients was reduced from 7 days to 5 days (p = 0.001). While the rate of surgical complications was the same between the two groups (p = 0.236), nonsurgical complications occurred significantly less frequently in the ERAS® cohort (p = 0.018).
There are well-known stumbling blocks in implementing and maintaining an ERAS® concept; however, it is worthwhile for the patient to circumnavigate this and establish ERAS® as the standard treatment path.
Summary Background The calcitonin gene-related peptide (CGRP) pathway is a promising target for preventive therapies in patients with migraine. We assessed the safety and efficacy of AMG 334, a fully ...human monoclonal antibody against the CGRP receptor, for migraine prevention. Methods In this multicentre, randomised, double-blind, placebo-controlled, phase 2 trial, patients aged 18–60 years with 4 to 14 migraine days per month were enrolled at 59 headache and clinical research centres in North America and Europe, and randomly assigned in a 3:2:2:2 ratio to monthly subcutaneous placebo, AMG 334 7 mg, AMG 334 21 mg, or AMG 334 70 mg using a sponsor-generated randomisation sequence centrally executed by an interactive voice response or interactive web response system. Study site personnel, patients, and the sponsor study personnel were masked to the treatment assignment. The primary endpoint was the change in monthly migraine days from baseline to the last 4 weeks of the 12-week double-blind treatment phase. The primary endpoint was calculated using the least squares mean at each timepoint from a generalised linear mixed-effect model for repeated measures. Safety endpoints were adverse events, clinical laboratory values, vital signs, and anti-AMG 334 antibodies. The study is registered with ClinicalTrials.gov , number NCT01952574 . An open-label extension phase of up to 256 weeks is ongoing and will assess the long-term safety of AMG 334. Findings From Aug 6, 2013, to June 30, 2014, 483 patients were randomly assigned to placebo (n=160), AMG 334 7 mg (n=108), AMG 334 21 mg (n=108), or AMG 334 70 mg (n=107). The mean change in monthly migraine days at week 12 was −3·4 (SE 0·4) days with AMG 334 70 mg versus −2·3 (0·3) days with placebo (difference −1·1 days 95% CI −2·1 to −0·2, p=0·021). The mean reductions in monthly migraine days with the 7 mg (−2·2 SE 0·4) and the 21 mg (−2·4 0·4) doses were not significantly different from that with placebo. Adverse events were recorded in 82 (54%) patients who received placebo, 54 (50%) patients in the AMG 334 7 mg group, 54 (51%) patients in the AMG 334 21 mg group, and 57 (54%) patients in the AMG 334 70 mg group. The most frequently reported adverse events were nasopharyngitis, fatigue, and headache. Serious adverse events were reported for one patient in the AMG 334 7 mg group (ruptured ovarian cyst) and one patient in the AMG 334 70 mg group (migraine and vertigo); these events were judged to be unrelated to AMG 334 treatment. Nine (3%) of 317 patients had neutralising antibodies. No apparent association was recorded between patients with positive anti-AMG 334 antibodies and adverse events. No clinically significant vital signs, laboratory, or electrocardiogram findings were recorded. Interpretation These results suggest that AMG 334 70 mg might be a potential therapy for migraine prevention in patients with episodic migraine and support further investigation of AMG 334 in larger phase 3 trials. The systematic comparison of genomic sequences from different organisms represents a central focus of contemporary genome analysis. Comparative analyses of vertebrate sequences can identify coding ...and conserved non-coding regions, including regulatory elements, and provide insight into the forces that have rendered modern-day genomes. As a complement to whole-genome sequencing efforts, we are sequencing and comparing targeted genomic regions in multiple, evolutionarily diverse vertebrates. Here we report the generation and analysis of over 12 megabases (Mb) of sequence from 12 species, all derived from the genomic region orthologous to a segment of about 1.8 Mb on human chromosome 7 containing ten genes, including the gene mutated in cystic fibrosis. These sequences show conservation reflecting both functional constraints and the neutral mutational events that shaped this genomic region. In particular, we identify substantial numbers of conserved non-coding segments beyond those previously identified experimentally, most of which are not detectable by pair-wise sequence comparisons alone. Analysis of transposable element insertions highlights the variation in genome dynamics among these species and confirms the placement of rodents as a sister group to the primates.
