Background
Triple negative breast cancer (TNBC) is a poor prognosis breast cancer with the highest mutation rate and limited treatment options. MiR-155 is highly expressed in TNBC, but its role and ...potential mechanism in TNBC remain to be elucidated.
Objective
The aim of this study is to examine the effect of interfering with miRNA-155 on the inflammatory pathway of NLRP 3 in TNBC (MDA-MB-231).
Methods
MiRNA-155-specific interference (Si-miR-155) on MDA-MB-231 cell was manifested by transfection of miRNA-155 inhibitor. Meanwhile, blank control (Blank) and negative control (NC) were set. Cell growth and proliferation rate were detected by MTT; apoptosis rate were detected by flow cytometry; colony forming test was used to detected cell viability; cell migration ability was detected by Wound healing assay; TNF-α, IL-18, IL-6 and IL-1β levels were detected by ELISA. The mRNA of miRNA-155, NLRP3, ASC, caspase-1 and Ki67 were detected by qRT-PCR. The expression levels of NLRP3, caspase-1, ASC and Ki67 were detected by Western blotting.
Results
The proliferation rate of Si-miRNA-155 group decreased, while the apoptosis rate increased significantly. After interfering with miRNA-155, the number of cancer cell colonies and the migration ability was decreased, and the secretion levels of IL-18, TNF-α, IL-6 and IL-1β were also inhibited. Moreover the mRNA and protein expression of NLRP3, caspase-1, ASC and Ki67 were significantly suppressed.
Conclusions
Interference with miRNA-155 can inhibit the NLRP3 pathway of MDA-MB-231 cells, as well as the proliferation, migration and inflammatory factor secretion of MDA-MB-231 cell, and can accelerate its apoptosis.

Alzheimer’s disease (AD) is the most common type of dementia but lacks effective treatment at present. Gastrodin (GAS) is a phenolic glycoside extracted from the traditional Chinese herb—Gastrodia ...elata—and has been reported as a potential therapeutic agent for AD. However, its efficiency is reduced for AD patients due to its limited BBB permeability. Studies have demonstrated the feasibility of opening the blood-brain barrier (BBB) via focused ultrasound (FUS) to overcome the obstacles preventing medicines from blood flow into the brain tissue. We explored the therapeutic potential of FUS-mediated BBB opening combined with GAS in an AD-like mouse model induced by unilateral intracerebroventricular (ICV) injection of Aβ1-42. Mice were divided into 5 groups: control, untreated, GAS, FUS and FUS+GAS. Combined treatment (FUS+GAS) rather than single intervention (GAS or FUS) alleviated memory deficit and neuropathology of AD-like mice. The time that mice spent in the novel arm was prolonged in the Y-maze test after 15-day intervention, and the waste-cleaning effect was remarkably increased. Contents of Aβ, tau, and P-tau in the observed (also the targeted) hippocampus were reduced. BDNF, synaptophysin (SYN), and PSD-95 were upregulated in the combined group. Overall, our results demonstrate that FUS-mediated BBB opening combined with GAS injection exerts the potential to alleviate memory deficit and neuropathology in the AD-like experimental mouse model, which may be a novel strategy for AD treatment.

This article interrogates the early reception history of the late Qing Chinese novel The Travels of Lao Can authored by Liu E (1857–1909). It argues that for decades this novel was widely read as a ...book of prophecy that revealed the future of China based on traditional Chinese occult wisdom. It was this alleged mystique, instead of its literary merits, that accounted for the high popularity of the text in the late Qing and Republican book market. This argument is predicated upon the discovery and reappraisal of a 1916 “fake edition” of the book until now unremarked in Chinese literary historiography. Investigating this forgery reveals that it was arguably the most widely circulated edition of The Travels of Lao Can in the Republican period. Bound with a newly fabricated sequel that explicitly manifested its “divinatory efficacy”, this edition greatly shaped the popular reception of the text as a book of prophecy. Revisiting this “fake edition” and the forgotten stories behind it not only enables the rereading of The Travels of Lao Can in its full complexity, but also points to deeper questions about the relationship between print culture, literary consumption, and the production of occult knowledge in China's modern transition.
This essay proposes a religious approach to reading modern Chinese fiction. Departing from existing scholarship that seeks overt religious elements in literature, it argues that non-religious and ...even anti-religious texts can evoke psychological states akin to religious experiences, particularly those associated with Buddhism. To this end, it analyzes three “secular” examples from the canon of modern Chinese fiction. The first example, Lu Xun’s short story “My Old Home” (1921), instantiates this essay’s basic analytical framework, which identifies two fundamental impulses in modern Chinese fictional narrative: the impulse of representing—the mimesis of reality in a familiar past-present-future temporal progression, and the impulse of invoking, which creates a suspended temporality apart from the narrative’s natural temporal universe, and thereby allows the readers to witness something that redeems reality rather than representing it. The second example, Lao She’s short story “Soul-Slaying Spear” (1935), is written during a period when realism was criticized for its impotence in changing social reality. This story challenges this view by showing that realism’s transformative power is inherent in its impulse of invoking. The third example, The Travels of Lao Can (1903–1906), points to an indigenous provenance of the impulse of invoking, often masked by post-Literary Revolution foreign influences, which Frederic Jameson calls “the antinomies of realism.” The religious approach exemplified by these three works echoes Francisca Cho’s Buddhist theory of cinema, which argues that the medium of cinema, regardless of its content, can activate Buddhist enlightenment. Similarly, this essay reveals how the medium of language, even in the cloak of realistic mimesis, can give rise to similar experiences. Recognizing this Buddhist connection does not imply that Buddhism is the sole source of this literary phenomenon but foregrounds how Buddhism can provide new perspectives to literary criticism which is often trapped in Eurocentric views. This article presents the findings of an empirical study exploring a set of system characteristics that influence users’ acceptance of electronic recordkeeping systems. Thirty-four semi-structured ...interviews were conducted with general users (i.e., non-specialists in information and records management), from which 10 system characteristics were identified, for example, sharing, integrated, and complete. In addition, this study has identified three moderating factors (i.e., gender, organizational context, and occupational difference) that influence the strength of the relationship between the set of system characteristics and users’ perceptions of electronic recordkeeping systems. Based on the results of this study and the Technology Acceptance Model, this article proposes a research model describing how the set of system characteristics and the moderating factors influence users’ perceptions of electronic recordkeeping systems. Given that records management work is no longer purely the responsibility of information and records management specialists but rather is increasingly part of every employee’s duties, it is vital to gain an understanding of the external variables that have an impact on users’ perceptions of electronic recordkeeping systems and that may enhance users’ intention to use these systems.

Focused ultrasound (FUS) is a potential tool for treating chronic pain by modulating the central nervous system. Herein, we aimed to determine whether transcranial FUS stimulation of the anterior ...cingulate cortex (ACC) effectively improved chronic pain in the chronic compress injury mice model at different stages of neuropathic pain. The mechanical threshold of pain was recorded in the nociceptive tests. We found FUS stimulation elevated the mechanical threshold of pain in both short-term (p<0.01) and long-term (p<0.05) experiments. Furthermore, we determined protein expression differences in ACC between the control group, the intervention group, and the Sham group to analyze the underlying mechanism of FUS stimulation in improving neuropathic pain. Additionally, the results showed FUS stimulation led to alterations in differential proteins in long-term experiments, including cellular processes, cellular signaling, and information storage and processing. Our findings indicate FUS may effectively alleviate mechanical neuropathic pain via the ACC’s stimulation, especially in the chronic state.
