IPv4 is now officially deprecated by the IETF. A significant amount of effort has already been expended by the HEPiX IPv6 Working Group on testing dual-stacked hosts and IPv6-only CPU resources. ...Dual-stack adds complexity and administrative overhead to sites that may already be starved of resource. This has resulted in a very slow uptake of IPv6 from WLCG sites. 464XLAT (RFC6877) is intended for IPv6 single-stack environments that require the ability to communicate with IPv4-only endpoints. This paper will present a deployment strategy for 464XLAT, operational experiences of using 464XLAT in production at a WLCG site and important information to consider prior to deploying 464XLAT.
The Queen Mary University of London Grid site's Lustre file system has recently undergone a major upgrade from version 1.8 to the most recent 2.8 release, and a capacity increase to over 3 PB. Lustre ...is an open source, POSIX compatible, clustered file system presented to the Grid using the StoRM Storage Resource Manager. The motivation and benefits of upgrading including hardware and software choices, are discussed. The testing, performance improvements and data migration procedure are outlined as is the source code modifications needed for StoRM compatibility. Benchmarks and real world performance are presented and future plans discussed.

IPv4 network addresses are running out and the deployment of IPv6 networking in many places is now well underway. Following the work of the HEPiX IPv6 Working Group, a growing number of sites in the ...Worldwide Large Hadron Collider Computing Grid (WLCG) are deploying dual-stack IPv6/IPv4 services. The aim of this is to support the use of IPv6-only clients, i.e. worker nodes, virtual machines or containers. The IPv6 networking protocols while they do contain features aimed at improving security also bring new challenges for operational IT security. The lack of maturity of IPv6 implementations together with the increased complexity of some of the protocol standards raise many new issues for operational security teams. The HEPiX IPv6 Working Group is producing guidance on best practices in this area. This paper considers some of the security concerns for WLCG in an IPv6 world and presents the HEPiX IPv6 working group guidance for the system administrators who manage IT services on the WLCG distributed infrastructure, for their related site security and networking teams and for developers and software engineers working on WLCG applications.
The fraction of Internet traffic carried over IPv6 continues to grow rapidly. IPv6 support from network hardware vendors and carriers is pervasive and becoming mature. A network infrastructure ...upgrade often offers sites an excellent window of opportunity to configure and enable IPv6. There is a significant overhead when setting up and maintaining dual-stack machines, so where possible sites would like to upgrade their services directly to IPv6 only. In doing so, they are also expediting the transition process towards its desired completion. While the LHC experiments accept there is a need to move to IPv6, it is currently not directly affecting their work. Sites are unwilling to upgrade if they will be unable to run LHC experiment workflows. This has resulted in a very slow uptake of IPv6 from WLCG sites. For several years the HEPiX IPv6 Working Group has been testing a range of WLCG services to ensure they are IPv6 compliant. Several sites are now running many of their services as dual-stack. The working group, driven by the requirements of the LHC VOs to be able to use IPv6-only opportunistic resources, continues to encourage wider deployment of dual-stack services to make the use of such IPv6-only clients viable. This paper presents the working group's plan and progress so far to allow sites to deploy IPv6-only CPU resources. This includes making experiment central services dual-stack as well as a number of storage services. The monitoring, accounting and information services that are used by jobs also need to be upgraded. Finally the VO testing that has taken place on hosts connected via IPv6-only is reported.
Optimising network performance is key to high bandwidth data transfers required for a Tier-2 site. We describe the techniques we have used to obtain good performance. Multiple parallel transfers allowed us to saturate a l Gbit/s link for 24 hours - whilst still achieving acceptable download speeds. Source based routing and multiple data transfer servers allowed us to use an otherwise unused "resilient" link.
Obesity has become a serious public health problem and a major risk factor for the development of illnesses, such as insulin resistance and hypertension. Human homeostatic systems have adapted to ...daily changes in light and dark in a way that the body anticipates the sleep and activity periods. Mammals have developed an endogenous circadian clock located in the suprachiasmatic nuclei of the anterior hypothalamus that responds to the environmental light-dark cycle. Similar clocks have been found in peripheral tissues, such as the liver, intestine, and adipose tissue, regulating cellular and physiological functions. The circadian clock has been reported to regulate metabolism and energy homeostasis in the liver and other peripheral tissues. This is achieved by mediating the expression and/or activity of certain metabolic enzymes and transport systems. In return, key metabolic enzymes and transcription activators interact with and affect the core clock mechanism. In addition, the core clock mechanism has been shown to be linked with lipogenic and adipogenic pathways. Animals with mutations in clock genes that disrupt cellular rhythmicity have provided evidence for the relationship between the circadian clock and metabolic homeostasis. In addition, clinical studies in shift workers and obese patients accentuate the link between the circadian clock and metabolism. This review will focus on the interconnection between the circadian clock and metabolism, with implications for obesity and how the circadian clock is influenced by hormones, nutrients, and timed meals.

Presently, about 12% of the population is 65 years or older and by the year 2030 that figure is expected to reach 21%. In order to promote the well-being of the elderly and to reduce the costs ...associated with health care demands, increased longevity should be accompanied by ageing attenuation. Energy restriction, which limits the amount of energy consumed to 60-70% of the daily intake, and intermittent fasting, which allows the food to be available ad libitum every other day, extend the life span of mammals and prevent or delay the onset of major age-related diseases, such as cancer, diabetes and cataracts. Recently, we have shown that well-being can be achieved by resetting of the circadian clock and induction of robust catabolic circadian rhythms via timed feeding. In addition, the clock mechanism regulates metabolism and major metabolic proteins are key factors in the core clock mechanism. Therefore, it is necessary to increase our understanding of circadian regulation over metabolism and longevity and to design new therapies based on this regulation. This review will explore the present data in the field of circadian rhythms, ageing and metabolism.
Abstract The master clock located in the suprachiasmatic nuclei (SCN) of the anterior hypothalamus regulates circadian rhythms in mammals. The clock is an intracellular, transcriptional mechanism ...sharing the same molecular components in SCN neurons and in peripheral cells, such as the liver, intestine, and retina. The circadian clock controls food processing and energy homeostasis by regulating the expression and/or activity of enzymes involved in cholesterol, amino acid, lipid, glycogen, and glucose metabolism. In addition, many hormones involved in metabolism, such as insulin, glucagon, adiponectin, corticosterone, leptin, and ghrelin, exhibit circadian oscillation. Furthermore, disruption of circadian rhythms is involved in the development of cancer, metabolic syndrome, and obesity. Metabolism and food intake also feed back to influence the biological clock. Calorie restriction (CR) entrains the SCN clock, whereas timed meals entrain peripheral oscillators. Furthermore, the cellular redox state, dictated by food metabolism, and several nutrients, such as glucose, ethanol, adenosine, caffeine, thiamine, and retinoic acid, can phase-shift circadian rhythms. In conclusion, there is a large body of evidence that links feeding regimens, food components, and the biological clock.

Abstract
Obesity is a major risk factor for the development of illnesses, such as insulin resistance and hypertension, and has become a serious public health problem. Mammals have developed a ...circadian clock located in the hypothalamic suprachiasmatic nuclei (SCN) that responds to the environmental light-dark cycle. Clocks similar to the one located in the SCN are found in peripheral tissues, such as the kidney, liver, and adipose tissue. The circadian clock regulates metabolism and energy homeostasis in peripheral tissues by mediating activity and/or expression of key metabolic enzymes and transport systems. Knockouts or mutations in clock genes that lead to disruption of cellular rhythmicity have provided evidence to the tight link between the circadian clock and metabolism. In addition, key proteins play a dual role in regulating the core clock mechanism, as well as adipose tissue metabolism, and link circadian rhythms with lipogenesis and lipolysis. Adipose tissues are distinguished as white, brown, and beige (or brite), each with unique metabolic characteristics. Recently, the role of the circadian clock in regulating the differentiation into the different adipose tissues has been investigated. In this review, the role of clock proteins and the downstream signaling pathways in white, brown, and brite adipose tissue function and differentiation will be reviewed. In addition, chronodisruption and metabolic disorders and clinical aspects of circadian adiposity will be addressed.
The role of the circadian clock in regulating adipose tissue function and differentiation and key proteins that play a dual role in regulating clock mechanism and adipose tissue metabolism are addressed.
