Osteoarthritis (OA), the most prevalent age-related joint disorder, is characterized by chronic inflammation, progressive articular cartilage destruction, and subchondral bone sclerosis. Accumulating ...evidences indicate that circular RNAs (circRNAs) play a critical role in various diseases, but the function of circRNAs in OA remains largely unknown. Here we showed that circRNA.33186 was significantly upregulated in IL-1β)-treated chondrocytes and in cartilage tissues of a destabilized medial meniscus (DMM)-induced OA mouse model. Knockdown of circRNA.33186 increased anabolic factor (type II collagen) expression and decreased catabolic factor (MMP-13) expression. Knockdown of circRNA.33186 also promoted proliferation and inhibited apoptosis in IL-1β-treated chondrocytes. Silencing of circRNA.33186 in vivo markedly alleviated DMM-induced OA. Mechanistic study showed that circRNA.33186 directly binds to and inhibits miR-127-5p, thereby increasing MMP-13 expression, and contributes to OA pathogenesis. Taken together, our findings demonstrated a fundamental role of circRNA.33186 in OA progression and provide a potential drug target in OA therapy.

Very recently Xue et al. reported an important detection of the X-ray transient, CDF-S XT2, whose light curve is analogous to X-ray plateau features of gamma-ray burst afterglows. They suggested that ...this transient is powered by a remnant stable magnetar from a binary neutron star merger because several pieces of evidence (host galaxy, location, and event rate) all point toward such an assumption. In this Letter, we revisit this scenario and confirm that this X-ray emission can be well explained by the internal gradual magnetic dissipation process in an ultra-relativistic wind of the newborn magnetar. We show that both the light curve and spectral evolution of CDF-S XT2 can be well fitted by such a model. Furthermore, we can probe some key properties of the central magnetar, such as its initial spin period, surface magnetic field strength, and wind saturation Lorentz factor.
ATPase associated with a variety of cellular activities (AAA ATPase) family members are closely linked to tumor formation and progression. However, their roles in hepatocellular carcinoma (HCC) ...largely remain unclear.
Bioinformatic analyses of public databases were used to excavate the potential AAA ATPases that may contribute to HCC, and thyroid hormone receptor interactor 13 (TRIP13) was selected to following researches because of its most prominently differential expression. Western blot, qRT-PCR and immunohistochemistry were used to detect the expression of TRIP13 in HCC tissues, and then the relationship between TRIP13 expression and clinicopathological parameters were evaluated. Finally, its functions and potential mechanisms were investigated through a series gain- and loss-of-function strategies both in vitro and in vivo.
TRIP13 was significantly overexpressed in HCC tissues and high level of TRIP13 was closely correlated with a worse clinical outcome. Functionally, elevated TRIP13 facilitated cell proliferation, migration, invasion, and promoted cellular epithelial-mesenchymal transition (EMT) in vitro, while promote tumor growth and lung metastasis in vivo. Mechanistically, TRIP13 interacted with ACTN4 and positively regulated its expression, thus activating the AKT/mTOR pathway to drive tumor progression. Moreover, miR-192-5p served as an upstream regulator of TRIP13 by directly binding to TRIP13 mRNA 3' UTR, which may partially explain the high expression of TRIP13 in HCC.
Our findings identified TRIP13 as a promising candidate oncogene in HCC, and TRIP13 induced cell migration, invasion and metastasis of HCC through the AKT/mTOR signaling via interacting with ACTN4.

Counting everything, also named few-shot counting, requires a model to be able to count objects with any novel (unseen) category giving few exemplar boxes. However, the existing few-shot counting ...methods are sub-optimal due to weak feature representation, such as the correlation between the exemplar patch and query feature, and contextual dependencies in density map prediction. In this paper, we propose a very simple but effective method, CSTrans, consisting of a Correlation-guided Self-Activation (CSA) module and a Local Dependency Transformer (LDT) module, to mitigate the above two issues, respectively. The CSA utilizes the correlation map to activate the semantic features and suppress the noisy influence of the query features, aiming at mining the potential relation while enriching correlation representation. Furthermore, the LDT incorporates a Transformer to explore local contextual dependencies and predict the density map. Our method achieves competitive performance on FSC-147 and CARPK datasets. We hope its simple implementation and superior performance can serve as a new and strong baseline for few-shot counting tasks and attract more interest in designing simple but effective models in future studies. Abstract Very recently, a particularly long gamma-ray burst (GRB) 230307A was reported and proposed to originate from a compact binary merger based on its host galaxy property, kilonova, and heavy ...elements. More intriguingly, a very early plateau followed by a rapid decline in the soft X-ray band was detected in its light curve by the Lobster Eye Imager for Astronomy, indicating strong evidence of the existence of a magnetar as the merger product. This Letter explores the magnetar wind internal gradual magnetic dissipation model, in which the radiative efficiency evolves over time, and successfully fits it to the observed data. Our results reinforce the notion that the X-ray plateau serves as a powerful indicator of a magnetar and imply that an evolving efficiency is likely to be a common feature in X-ray plateaus of GRB afterglows. In addition, we also discuss the explanations for the prompt emission, GRB afterglows, as well as kilonova, and predict possible kilonova afterglows in a magnetar central engine.

A symbolic C++ library-Tinned-has been developed for symbolic differentiation and manipulation in response theory. By recognizing different key building blocks in the density matrix-based ...(Thorvaldsen et al., J. Chem. Phys. 2008, 129, 214108) and coupled-cluster response theories, we have implemented their corresponding C++ symbolic classes, including but not limited to one- and two-electron operators, exchange-correlation energy and potential, and coupled-cluster operator. Formulas of response theory can be well expressed in terms of the symbolic classes in the library Tinned. Their high-order perturbation-strength derivatives can be straightforwardly computed and extracted afterwards for numerical evaluation. The library Tinned will greatly facilitate the development work of response theory and may lead to a unified framework for response theory at different levels of electronic structure theory.

Designing effective electrocatalysts for the carbon dioxide reduction reaction (CO2RR) is an appealing approach to tackling the challenges posed by rising CO2 levels and realizing a closed carbon ...cycle. However, fundamental understanding of the complicated CO2RR mechanism in CO2 electrocatalysis is still lacking because model systems are limited. We have designed a model nickel single‐atom catalyst (Ni SAC) with a uniform structure and well‐defined Ni‐N4 moiety on a conductive carbon support with which to explore the electrochemical CO2RR. Operando X‐ray absorption near‐edge structure spectroscopy, Raman spectroscopy, and near‐ambient X‐ray photoelectron spectroscopy, revealed that Ni+ in the Ni SAC was highly active for CO2 activation, and functioned as an authentic catalytically active site for the CO2RR. Furthermore, through combination with a kinetics study, the rate‐determining step of the CO2RR was determined to be *CO2−+H+→*COOH. This study tackles the four challenges faced by the CO2RR; namely, activity, selectivity, stability, and dynamics.
Ni‐che reaction: In situ reduction of nickel(II) 2,9,16,23‐tetra(amino)phthalocyanine, anchored on the surface of carbon nanotubes, yields nickel single atoms. Advanced spectroscopy of the single‐atom catalyst reveals that Ni+ is a highly active catalytic site for CO2 activation and reduction.
