Upregulation of the proto-oncogene plasminogen activator inhibitor-1 (PAI-1) is a common hallmark of various solid tumours, but the mechanisms controlling its expression are not fully understood.
We ...investigate microRNAs (miRNAs) regulating PAI-1 in a panel of normal bladder urothelial biopsies, superficial Ta bladder tumours and invasive T1-T4 tumours using expression microarrays and qRT-PCR. The prognostic implications of PAI-1 deregulation are established by tissue microarray staining of non-muscle-invasive bladder tumours. MicroRNA repression of PAI-1 is assayed by ectopic miRNA expression, argonaute immunoprecipitation and luciferase assays.
We found that the miR-143/-145 cluster is downregulated in all stages of bladder cancer and inversely correlated with PAI-1 expression. Mature miR-143 and miR-145 are coordinately expressed, and both directly target the PAI-1 3'UTR, leading to reduced PAI-1 mRNA and protein levels. Furthermore, we show that PAI-1 and miR-145 levels may serve as useful prognostic markers for non-muscle-invasive bladder tumours for which accurate progressive outcome is currently difficult to predict.
This report provides the first evidence for direct miRNA regulation of PAI-1 in bladder cancer. We also demonstrate mRNA co-targeting by a cluster of non-family miRNAs, and suggest miR-145 and PAI-1 as clinically relevant biomarkers in bladder cancer.

beta decay of proton-rich nuclei plays an important role in exploring isospin mixing. The beta decay of P-26 at the proton drip line is studied using double-sided silicon strip detectors operating in ...conjunction with high-purity germanium detectors. The T = 2 isobaric analog state (IAS) at 13 055 keV and two new high-lying states at 13 380 and 11 912 keV in Si-26 are unambiguously identified through beta-delayed two-proton emission (beta 2p). Angular correlations of two protons emitted from Si-26 excited states populated by P-26 beta decay are measured, which suggests that the two protons are emitted mainly sequentially. We report the first observation of a strongly isospin-mixed doublet that deexcites mainly via two-proton decay. The isospin mixing matrix element between the Si-26 IAS and the nearby 13 380-keV state is determined to be 130(21) keV, and this result represents the strongest mixing, highest excitation energy, and largest level spacing of a doublet ever observed in beta-decay experiments.

We present a single-dish mapping algorithm with a number of advantages over traditional techniques. (1) Our algorithm makes use of weighted modeling, instead of weighted averaging, to interpolate ...between signal measurements. This smooths the data, but without blurring the data beyond instrumental resolution. Techniques that rely on weighted averaging blur point sources sometimes by as much as 40%. (2) Our algorithm makes use of local, instead of global, modeling to separate astronomical signal from instrumental and/or environmental signal drift along the telescope's scans. Other techniques, such as basket weaving, model this drift with simple functional forms (linear, quadratic, etc.) across the entirety of scans, limiting their ability to remove such contaminants. (3) Our algorithm makes use of a similar, local modeling technique to separate astronomical signal from radio-frequency interference (RFI), even if only continuum data are available. (4) Unlike other techniques, our algorithm does not require data to be collected on a rectangular grid or regridded before processing. (5) Data from any number of observations, overlapping or not, may be appended and processed together. (6) Any pixel density may be selected for the final image. We present our algorithm and evaluate it using both simulated and real data. We are integrating it into the image-processing library of the Skynet Robotic Telescope Network, which includes optical telescopes spanning four continents, and now also Green Bank Observatory's 20 m diameter radio telescope in West Virginia. Skynet serves hundreds of professional users, and additionally tens of thousands of students, of all ages. Default data products are generated on the fly, but will soon be customizable after the fact.
In this study, the Arabidopsis thaliana NPR1 (non‐expressor of PR genes) gene was integrated into an elite wheat cultivar, and the response of the transgenic wheat expressing NPR1 to inoculation with ...Fusarium asiaticum was analysed. With seedling inoculation, the transgenic lines showed significantly increased fusarium seedling blight (FSB) susceptibility, whereas floret inoculation resulted in enhanced fusarium head blight (FHB) resistance. Quantitative real‐time PCR revealed that expression of two defence genes, PR3 and PR5, was associated with susceptible reactions to FSB and FHB, whereas the PR1 gene was activated in resistance responses. This inverse modulation by the constitutively expressed NPR1 gene suggests that NPR1 has a bifunctional role in regulating defence responses in plants. Therefore, it is unsuitable for improving overall resistance to FSB and FHB in wheat.

We report direct evidence of room-temperature ferromagnetic ordering in O-deficient ZnO:Cu films by using soft x-ray magnetic circular dichroism and x-ray absorption. Our measurements have revealed ...unambiguously two distinct features of Cu atoms associated with (i) magnetically ordered Cu ions present only in the oxygen-deficient samples and (ii) magnetically disordered regular Cu2+ ions present in all the samples. We find that a sufficient amount of both oxygen vacancies (V(O)) and Cu impurities is essential to the observed ferromagnetism, and a non-negligible portion of Cu impurities is uninvolved in the magnetic order. Based on first-principles calculations, we propose a microscopic "indirect double-exchange" model, in which alignments of localized large moments of Cu in the vicinity of the V(O) are mediated by the large-sized vacancy orbitals.

The Galactic center black hole Sagittarius A* is a variable near-infrared (NIR) source that exhibits bright flux excursions called flares. When flux from Sgr A* is detected, the light curve has been ...shown to exhibit red noise characteristics and the distribution of flux densities is non-linear, non-Gaussian, and skewed to higher flux densities. However, the low-flux density turnover of the flux distribution is below the sensitivity of current single-aperture telescopes. For this reason, the median NIR flux has only been inferred indirectly from model fitting, but it has not been directly measured. In order to explore the lowest flux ranges, to measure the median flux density, and to test if the previously proposed flux distributions fit the data, we use the unprecedented resolution of the GRAVITY instrument at the VLTI. We obtain light curves using interferometric model fitting and coherent flux measurements. Our light curves are unconfused, overcoming the confusion limit of previous photometric studies. We analyze the light curves using standard statistical methods and obtain the flux distribution. We find that the flux distribution of Sgr A* turns over at a median flux density of (1.1 ± 0.3) mJy. We measure the percentiles of the flux distribution and use them to constrain the NIR
K
-band spectral energy distribution. Furthermore, we find that the flux distribution is intrinsically right-skewed to higher flux density in log space. Flux densities below 0.1 mJy are hardly ever observed. In consequence, a single powerlaw or lognormal distribution does not suffice to describe the observed flux distribution in its entirety. However, if one takes into account a power law component at high flux densities, a lognormal distribution can describe the lower end of the observed flux distribution. We confirm the rms–flux relation for Sgr A* and find it to be linear for all flux densities in our observation. We conclude that Sgr A* has two states: the bulk of the emission is generated in a lognormal process with a well-defined median flux density and this quiescent emission is supplemented by sporadic flares that create the observed power law extension of the flux distribution.

We report the time-resolved spectral analysis of a bright near-infrared and moderate X-ray flare of Sgr A⋆. We obtained light curves in the M, K, and H bands in the mid- and near-infrared and in the ...2 − 8 keV and 2 − 70 keV bands in the X-ray. The observed spectral slope in the near-infrared band is νLν ∝ ν0.5 ± 0.2; the spectral slope observed in the X-ray band is νLν ∝ ν−0.7 ± 0.5. Using a fast numerical implementation of a synchrotron sphere with a constant radius, magnetic field, and electron density (i.e., a one-zone model), we tested various synchrotron and synchrotron self-Compton scenarios. The observed near-infrared brightness and X-ray faintness, together with the observed spectral slopes, pose challenges for all models explored. We rule out a scenario in which the near-infrared emission is synchrotron emission and the X-ray emission is synchrotron self-Compton. Two realizations of the one-zone model can explain the observed flare and its temporal correlation: one-zone model in which the near-infrared and X-ray luminosity are produced by synchrotron self-Compton and a model in which the luminosity stems from a cooled synchrotron spectrum. Both models can describe the mean spectral energy distribution (SED) and temporal evolution similarly well. In order to describe the mean SED, both models require specific values of the maximum Lorentz factor γmax, which differ by roughly two orders of magnitude. The synchrotron self-Compton model suggests that electrons are accelerated to γmax ∼ 500, while cooled synchrotron model requires acceleration up to γmax ∼ 5 × 104. The synchrotron self-Compton scenario requires electron densities of 1010 cm−3 that are much larger than typical ambient densities in the accretion flow. Furthermore, it requires a variation of the particle density that is inconsistent with the average mass-flow rate inferred from polarization measurements and can therefore only be realized in an extraordinary accretion event. In contrast, assuming a source size of 1 RS, the cooled synchrotron scenario can be realized with densities and magnetic fields comparable with the ambient accretion flow. For both models, the temporal evolution is regulated through the maximum acceleration factor γmax, implying that sustained particle acceleration is required to explain at least a part of the temporal evolution of the flare.

Understanding the photoexcited carrier-relaxation actions in ultrasmall black phosphorus quantum dots (BPQDs) will play a crucial role in the fields of electronics and optoelectronics. Herein, we ...report the ultraviolet (UV) saturable absorption and ultrafast photoexcited carrier-relaxation dynamics of BPQDs. The ultrasmall BPQDs are synthesized using a facile liquid-exfoliation method and possess a diameter of 3.8 ± 0.6 nm and a thickness of 1.5 ± 0.4 nm. Femtosecond open-aperture (OA) Z-scan measurements showed typical saturable absorption properties in the UV band. A negative nonlinear optical (NLO) absorption coefficient of -(1.4 ± 0.3) × 10
cm GW
and a saturable intensity of 6.6 ± 1.3 GW cm
were determined. Using a degenerate pump-probe technique, an ultrafast photoexcited carrier-recombination time was observed in the range of 216-305 fs, which was 3 orders of magnitude faster than that of BP nanosheets. Such an ultrafast relaxation component may be attributable to the edge- and step-mediated recombination and was confirmed by our density functional theory (DFT) calculations. This work provides fundamental insight into the underlying mechanism of the photoexcited carrier relaxation dynamic action in BPQDs which can enable UV photonic devices.
