Adhesion of foodborne pathogens to materials of industrial surfaces is an important step in their transmission through the food chain. Adhesion is also a prerequisite for bacterial ...colonisation within a host, to enable intracellular invasion. We define a strategy to reduce contamination and infection by Listeria monocytogenes, Campylobacter jejuni and Escherichia coli using an ethanol extract of Alpinia katsumadai seeds (AlpE) and epigallocatechin gallate (EGCG) as anti‐adhesive agents. We show for the first time that AlpE and EGCG reduce adhesion of individual cultures to polystyrene (AlpE, up to 10.6%; EGCG, up to 39.7%) and to the amoeba Acanthamoeba castellanii (AlpE, up to 52.6%; EGCG, up to 53.4%). The combination of AlpE/EGCG significantly reduced C. jejuni adherence to the abiotic (45.5%) and biotic (52.2%) surfaces. Thus, using natural agents from plants at low doses, we can potentially reduce the primary source of food contamination and a frequent source of infections.

Mycobacterium avium complex-related diseases are often associated with poorly maintained hot water systems. This calls for the development of new control strategies. The aim of this study was to ...investigate the activity of essential oils (EOs) from the Mediterranean plants, common juniper, immortelle, sage, lavandin, laurel, and white cedar against Mycobacterium avium ssp. avium, Mycobacterium intracellulare, and Mycobacterium gordonae in culturing broth and freshwater as their most common habitat. To do that, we developed a new method of water microdilution to determine their minimal effective concentrations (MEC). The most active EO was the one from the common juniper with the MEC of 1.6 mg mL
. Gas chromatography / mass spectrometry the juniper EO identified monoterpenes (70.54 %) and sesquiterpenes (25.9 %) as dominant component groups. The main monoterpene hydrocarbons were α-pinene, sabinene, and β-pinene. The juniper EO significantly reduced the cell viability of M. intracellulare and M. gordonae at MEC, and of M. avium at 2xMEC. Microscopic analysis confirmed its inhibitory effect by revealing significant morphological changes in the cell membrane and cytoplasm of all three bacteria. The mode of action of the juniper EO on the cell membrane was confirmed by a marked leakage of intracellular material. Juniper EO has a great practical potential as a complementary or alternative water disinfectant in hot water systems such as baths, swimming pools, spa pools, hot tubs, or even foot baths/whirlpools.
Nontuberculous mycobacteria, like other opportunistic premise plumbing pathogens, produce resistant biofilms on various surfaces in the plumbing system including pipes, tanks, and fittings. Since ...standard methods of water disinfection are ineffective in eradicating biofilms, research into new agents is necessary. Essential oils (EOs) have great potential as anti-biofilm agents. Therefore, the purpose of this research was to investigate the potential anti-biofilm effect of common juniper (Juniperus communis) and immortelle (Helichrysum italicum) EOs. Minimum inhibitory concentrations (MIC), minimum bactericidal concentrations (MBC), and minimum effective concentrations of EOs on Mycobacterium avium, M. intracellulare, and M. gordonae were tested. Additionally, biofilms on the surface of a stainless steel disc were treated with single or mixed concentration of EOs, in order to investigate their degeneration via the bacterial count and confocal laser scanning microscopy (CLSM). H. italicum EO showed the strongest biofilm degradation ability against all Mycobacteria strains that were tested. The strongest effect in the biofilm degradation after the single or mixed applications of EOs was observed against M. gordonae, followed by M. avium. The most resistant was the M. intracellulare biofilm. Synergistic combinations of J. communis and H. italicum EOs therefore seem to be an effective substance in biofilm degradation for use in small water systems such as baths or hot tubs.
Cilj: Ispitati antimikrobni i antioksidacijski učinak te sposobnost inhibicije biofilma eteričnog ulja (EU) smilja Helichrysum italicum (Roth) G. Don (H. italicum) prema netuberkuloznim ...mikobakterijama M. avium i M. intracellulare. Materijali i metode: Za određivanje minimalne inhibicijske koncentracije (MIK) i minimalne baktericidne koncentracije (MBK) EU H. italicum prema M. avium i M. intracellulare korištena je metoda mikrodilucije u bujonu. Preživljavanje mikobakterija u Middlebrook 7H9 bujonu pod utjecajem EU smilja praćeno je subkultivacijom suspenzija 0, 1, 4 i 8 dan. Za određivanje inhibicije stvaranja biofilma u vodi primjenjena je metoda bojenja biofilma s kristal violetom. Antioksidacijski učinak MIK koncentracije EU određen je DPPH (2,2-difenil-1-pikril-hidrazil) metodom. Rezultati: MIK i MBK vrijednost EU smilja iznosila je 3,2 mg/ml. Koncentracija ½ MIK vrijednosti dovela je nakon prvog dana do redukcije vijabilnosti M. avium za približno 2 log10 odnosno 2,5 log10 za M. intracellulare, dok je četvrti dan zabilježena potpuna inhibicija porasta obje vrste mikobakterija. M. avium je pokazao veću tendenciju stvaranja biofilma. Koncentracija od ¼ i ½ MIK vrijednosti dovela je do statistički značajne inhibicije stvaranja biofilma ispitivanih mikobakterija. MIK koncentracija EU H. italicum je nakon 60 minuta inhibirala 38,2 % DPPH radikala. Zaključak: Subinhibitorne koncentracije EU smilja značajno reduciraju vijabilnost M. avium i M. intracellulare i dovode do značajne inhibicije stvaranja biofilma ispitivanih mikobakterija u vodi što bi omogućilo primjenu niskih efektivnih koncentracija ovog EU, koje nemaju citotoksični učinak, kao prirodnog dezinficijensa u vodi.

Antibacterial and antibiofilm efficacy through a synergistic effect of herbal leaf extracts of bearberry (BE), bearberry tea (BTE), parsley (PE), and the strawberry tree (STE) was evaluated to discover an alternative approach to control urinary tract infections caused by Enterococci. UHPLC–DAD MS/MS analysis indicated that herbal extracts were rich in ellagic and gallic acids, catechin, quercetin, and arbutin, which are strong antioxidants. Mostly, Enterococcus faecalis isolates showed resistance only to gentamicin and/or norfloxacin (26.3 %) and moderate biofilm production (31.5 %), while 12 isolates of Enterococcus faecium showed multiple antibiotic resistance. The minimum inhibitory concentration for the majority of isolates was 1.25 mg ml−1 for STE, but 2.5 mg ml−1 for BE and BTE. Although individual extracts gave higher MIC values, for most isolates, a synergistic effect at lower concentrations was achieved, when BE or BTE (0.625 mg ml−1) was combined with STE (0.31 mg ml−1). The BTE extract was shown to have the strongest individual effect on initial adhesiveness and on biofilm formation to all selected isolates, while the synergism of BTE and STE caused significant biofilm inhibition at all concentrations tested. The synergistic effect achieved of the tested extract combinations might be a starting point in the development of alternative products, effective against antibiotic resistant Enterococcus isolates.

Cilj: Istražiti odabrane čimbenike virulencije te antimikrobnu osjetljivost/rezistenciju uropatogenih bakterija Enterococcus faecalis i Enterococcus faecium. Materijali i metode: Ukupno 96 urinarnih ...izolata enterokoka identificirano je standardnim i automatiziranim biokemijskim postupcima. Fenotipskim metodama dokazana je prisutnost kazeinaze i želatinaze te lipaze. Metodom agar dilucije određena je rezistencija prema lizozimu. Disk difuzijskom i mikrodilucijskom metodom ispitana je osjetljivost/rezistencija izolata prema odabranim antibioticima. Rezultati: Od ukupnog broja uropatogenih enterokoka, E. faecalis je identificiran u 79,2%, a E. faecium u 20,8% slučajeva. Većina sojeva uropatogenih enterokoka pokazivala je lipolitičku aktivnost, dok je produkcija želatinaze i kazeinaze dominantna karakteristika E. faecalis, a izostaje u E. faecium. Rezistencija prema nižim koncentracijama lizozima uočena je u 97% od ukupnog broja testiranih uropatogenih enterokoka. Visoki stupanj rezistencije na lizozim, zabilježen je u 72% svih ispitanih sojeva. Svi izolati E. faecalis bili su osjetljivi prema glikopeptidima i imipenemu. Vankomicin (VanA fenotip) rezistentni enterokoki (VRE) prisutni su isključivo među sojevima E. faecium. Isti sojevi su rezistentni prema imipenemu, ampicilinu i gentamicinu. Zaključak: S obzirom na rastuću važnost vrsta iz roda Enterococcus kao uzročnika hospitalnih i vanbolničkih infekcija te sve veću učestalost glikopeptidne rezistencije, utvrđivanje čimbenika virulencije i njihove uloge u patogenezi infekcija mokraćnog sustava od posebnog je značenja.
Globalni problem bakterijske rezistencije na antibiotike uzrok je potrage za novim antimikrobnim lijekovima. Zbog toga se mnogo očekuje od meda koji ima antimikrobno djelovanje na bakterije otporne ...na djelovanje antibiotika. Za antibakterijsku aktivnost meda odgovorno je više komponenti prisutnih u medu, u koncentracijama koje se razlikuju kod raznih vrsti meda. Nepredvidiva antibakterijska aktivnost meda, koja nije standardizirana, može otežati njegovu primjenu kao antimikrobnog agensa radi uočenih varijacija u in vitro antibakterijskoj aktivnosti različitih vrsta meda. Trenutačno se na tržištu može naći nekoliko vrsti meda sa standardiziranim razinama antibakterijskog djelovanja, od kojih je najpoznatiji med manuke (lat. Leptospermum scoparium).
Prijenos uzročnika infekcija rukama bolničkog osoblja vodeći je način prijenosa bolničkih infekcija. Uzročnici infekcija na ruke mogu dospjeti izravnim kontaktom osoblja s pacijentom, ali i kontaktom ...s različitim površinama u zdravstvenoj ustanovi. Bakterije koje su najčešći uzročnici bolničkih infekcija mogu mjesecima preživjeti na različitim površinama i materijalima koji tako mogu postati kontinuirani izvor zaraze za pacijente i osoblje u bolnici. U radu je opisano kako sušenje utječe na bakterijsku stanicu, navedeni su mehanizmi pomoću kojih pojedine bakterijske vrste uspješno preživljavaju u suhom okolišu, a objašnjeno je i kako različiti okolišni uvjeti utječu na preživljavanje bakterija u bolničkoj sredini. Navedeni su rezultati ispitivanja osjetljivosti na sušenje nekih češćih bakterijskih uzročnika bolničkih infekcija. Poznavanje čimbenika koji olakšavaju preživljavanja bakterija izloženih sušenju omogućuje izbor učinkovitih mjera dezinfekcije i sterilizacije kojima će se uništiti otporne vrste u zdravstvenim ustanovama, naročito bolnicama.
