Abstract
We present LIMFAST, a seminumerical code for simulating high-redshift galaxy formation and cosmic reionization as revealed by multitracer line intensity mapping (LIM) signals. LIMFAST builds ...upon and extends the 21cmFAST code widely used for 21 cm cosmology by implementing state-of-the-art models of galaxy formation and evolution. The metagalactic radiation background, including the production of various star formation lines, together with the 21 cm line signal tracing the neutral intergalactic medium (IGM), is self-consistently described by photoionization modeling and stellar population synthesis coupled to the galaxy formation model. We introduce basic structure and functionalities of the code, and demonstrate its validity and capabilities by showing broad agreements between the predicted and observed evolution of cosmic star formation, IGM neutral fraction, and metal enrichment. lines simulated by LIMFAST, and compare them with results from the literature. We elaborate on how several major aspects of our modeling framework, including models of star formation, chemical enrichment, and photoionization, may impact different LIM observables and thus become testable once applied to observational data. LIMFAST aims at being an efficient and resourceful tool for intensity mapping studies in general, exploring a wide range of scenarios of galaxy evolution and reionization and frequencies over which useful cosmological signals can be measured.
Background Biologic therapies with anti–tumor necrosis factor agents are promising treatments for hidradenitis suppurativa (HS). Objective We assessed the efficacy and safety of infliximab (IFX) for ...the treatment of moderate to severe HS. Methods A prospective double-blind treatment phase of 8 weeks where patients received IFX or placebo was followed by an open-label phase where patients taking placebo were given the opportunity to cross over to IFX, and an observational phase. Primary treatment efficacy was based on HS Severity Index. Secondary end points included Dermatology Life Quality Index, visual analog scale, and Physician Global Assessment scores. Inflammatory markers erythrocyte sedimentation rate and C-reactive protein were also assessed. Results More patients in the IFX than in the placebo group showed a 50% or greater decrease from baseline HS Severity Index score. In addition, statistically and clinically significant improvement from baseline was observed at week 8 in Dermatology Life Quality Index score, visual analog scale score, erythrocyte sedimentation rate, and C-reactive protein compared with placebo. Patients in the placebo group treated with IFX after week 8 (crossover) responded similarly to the original IFX group. Many patients withdrew during the observational phase to continue anti–tumor necrosis factor-alfa therapy. No unexpected serious adverse events were observed. Limitations Results are representative of a single center, patients were treated by a single physician, some patients did not return after their last infusion, and the HS Severity Index requires validation. Conclusions This clinical study represents the first formal assessment of IFX for treatment of moderate to severe HS. IFX was well tolerated, no unexpected safety issues were identified, and improvements in pain intensity, disease severity, and quality of life were demonstrated with concomitant reduction in clinical markers of inflammation.

Abstract
The epoch of reionization (EoR) offers a unique window into the dawn of galaxy formation, through which high-redshift galaxies can be studied by observations of both themselves and their ...impact on the intergalactic medium. Line intensity mapping (LIM) promises to explore cosmic reionization and its driving sources by measuring intensity fluctuations of emission lines tracing the cosmic gas in varying phases. Using LIMFAST, a novel seminumerical tool designed to self-consistently simulate LIM signals of multiple EoR probes, we investigate how building blocks of galaxy formation and evolution theory, such as feedback-regulated star formation and chemical enrichment, might be studied with multitracer LIM during the EoR. On galaxy scales, we show that the star formation law and the feedback associated with star formation can be indicated by both the shape and redshift evolution of LIM power spectra. For a baseline model of metal production that traces star formation, we find that lines highly sensitive to metallicity are generally better probes of galaxy formation models. On larger scales, we demonstrate that inferring ionized bubble sizes from cross-correlations between tracers of ionized and neutral gas requires a detailed understanding of the astrophysics that shape the line luminosity–halo mass relation. Despite various modeling and observational challenges, wide-area, multitracer LIM surveys will provide important high-redshift tests for the fundamentals of galaxy formation theory, especially the interplay between star formation and feedback by accessing statistically the entire low-mass population of galaxies as ideal laboratories, complementary to upcoming surveys of individual sources by new-generation telescopes.
Accurate estimates of natural and/or vaccine-induced antibodies to SARS-CoV-2 are difficult to obtain. Although model-based estimates of seroprevalence have been proposed, they require inputting ...unknown parameters including viral reproduction number, longevity of immune response, and other dynamic factors. In contrast to a model-based approach, the current study presents a data-driven detailed statistical procedure for estimating total seroprevalence (defined as antibodies from natural infection or from full vaccination) in a region using prospectively collected serological data and state-level vaccination data. Specifically, we conducted a longitudinal statewide serological survey with 88,605 participants 5 years or older with 3 prospective blood draws beginning September 30, 2020. Along with state vaccination data, as of October 31, 2021, the estimated percentage of those 5 years or older with naturally occurring antibodies to SARS-CoV-2 in Texas is 35.0% (95% CI = (33.1%, 36.9%)). This is 3x higher than, state-confirmed COVID-19 cases (11.83%) for all ages. The percentage with naturally occurring or vaccine-induced antibodies (total seroprevalence) is 77.42%. This methodology is integral to pandemic preparedness as accurate estimates of seroprevalence can inform policy-making decisions relevant to SARS-CoV-2.

Abstract
Understanding the duration of antibodies to the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus that causes COVID-19 is important to controlling the current pandemic. ...Participants from the Texas Coronavirus Antibody Response Survey (Texas CARES) with at least 1 nucleocapsid protein antibody test were selected for a longitudinal analysis of antibody duration. A linear mixed model was fit to data from participants (n = 4553) with 1 to 3 antibody tests over 11 months (1 October 2020 to 16 September 2021), and models fit showed that expected antibody response after COVID-19 infection robustly increases for 100 days postinfection, and predicts individuals may remain antibody positive from natural infection beyond 500 days depending on age, body mass index, smoking or vaping use, and disease severity (hospitalized or not; symptomatic or not).

The role of somatic genetic variants in the pathogenesis of intracranial-aneurysm formation is unknown. We identified a 23-year-old man with progressive, right-sided intracranial aneurysms, ...ipsilateral to an impressive cutaneous phenotype. The index individual underwent a series of genetic evaluations for known connective-tissue disorders, but the evaluations were unrevealing. Paired-sample exome sequencing between blood and fibroblasts derived from the diseased areas detected a single novel variant predicted to cause a p.Tyr562Cys (g.149505130T>C GRCh37/hg19; c.1685A>G) change within the platelet-derived growth factor receptor β gene (PDGFRB), a juxtamembrane-coding region. Variant-allele fractions ranged from 18.75% to 53.33% within histologically abnormal tissue, suggesting post-zygotic or somatic mosaicism. In an independent cohort of aneurysm specimens, we detected somatic-activating PDGFRB variants in the juxtamembrane domain or the kinase activation loop in 4/6 fusiform aneurysms (and 0/38 saccular aneurysms; Fisher’s exact test, p < 0.001). PDGFRB-variant, but not wild-type, patient cells were found to have overactive auto-phosphorylation with downstream activation of ERK, SRC, and AKT. The expression of discovered variants demonstrated non-ligand-dependent auto-phosphorylation, responsive to the kinase inhibitor sunitinib. Somatic gain-of-function variants in PDGFRB are a novel mechanism in the pathophysiology of fusiform cerebral aneurysms and suggest a potential role for targeted therapy with kinase inhibitors.

Natural variation and changing climate in coastal oceans subject meroplanktonic organisms to broad ranges of pH and oxygen (O₂) levels. In controlled‐laboratory experiments we explored the ...interactive effects of pH, O₂, and semidiurnal pH fluctuations on the survivorship, development, and size of early life stages of two mytilid mussels, Mytilus californianus and M. galloprovincialis. Survivorship of larvae was unaffected by low pH, low O₂, or semidiurnal fluctuations for both mytilid species. Low pH (<7.6) resulted in delayed transition from the trochophore to veliger stage, but this effect of low pH was absent when incorporating semidiurnal fluctuations in both species. Also at low pH, larval shells were smaller and had greater variance; this effect was absent when semidiurnal fluctuations of 0.3 units were incorporated at low pH for M. galloprovincialis but not for M. californianus. Low O₂ in combination with low pH had no effect on larval development and size, indicating that early life stages of mytilid mussels are largely tolerant to a broad range of O₂ reflective of their environment (80–260 μmol kg⁻¹). The role of pH variability should be recognized as an important feature in coastal oceans that has the capacity to modulate the effects of ocean acidification on biological responses.

The prevalence of long-term symptoms of coronavirus disease 2019 (COVID-19) in nonhospitalized pediatric populations in the United States is not well described. The objective of this analysis was to ...examine the presence of persistent COVID symptoms in children by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibody status.
Data were collected between October 2020 and May 2022 from the Texas Coronavirus Antibody REsponse Survey, a statewide prospective population-based survey among 5-90 years old. Serostatus was assessed by the Roche Elecsys Anti-SARS-CoV-2 Immunoassay for detection of antibodies to the SARS-CoV-2 nucleocapsid protein. Self-reported antigen/polymerase chain reaction COVID-19 test results and persistent COVID symptom status/type/duration were collected simultaneously. Risk ratios for persistent COVID symptoms were calculated versus adults and by age group, antibody status, symptom presence/severity, variant, body mass index and vaccine status.
A total of 82 (4.5% of the total sample n = 1813, 8.0% pre-Delta, 3.4% Delta and beyond) participants reported persistent COVID symptoms (n = 27 1.5% 4-12 weeks, n = 58 3.3% >12 weeks). Compared with adults, all pediatric age groups had a lower risk for persistent COVID symptoms regardless of length of symptoms reported. Additional increased risk for persistent COVID symptoms >12 weeks included severe symptoms with initial infection, not being vaccinated and having unhealthy weight (body mass index ≥85th percentile for age and sex).
These findings highlight the existence of nonhospitalized youth who may also experience persistent COVID symptoms. Children and adolescents are less likely to experience persistent COVID symptoms than adults and more likely to be symptomatic, experience severe symptoms and have unhealthy weight compared with children/adolescents without persistent COVID symptoms.
