promoter, which is active over the flowering period, produces higher yields than wild type. This yield increase occurs with or without ...drought conditions during flowering. To understand the mechanistic basis of the increased yield, we characterized gene expression and metabolite profiles in leaves and developing female reproductive tissue, comprising florets, node, pith, and shank, over the flowering period with and without drought. The
promoter was most active in the vasculature, particularly phloem companion cells in florets and pith, consistent with the largest decreases in trehalose 6-phosphate (T6P) levels (2- to 3-fold) being found in pith and florets. Low T6P led to decreased gene expression for primary metabolism and increased gene expression for secondary metabolism, particularly lipid-related pathways. Despite similar changes in gene expression, the pith and floret displayed opposing assimilate profiles: sugars, sugar phosphates, amino acids, and lipids increased in florets, but decreased in pith. Possibly explaining this assimilate distribution, seven
genes were found to be up-regulated in the transgenic plants. SnRK1 activity and the expression of the gene for the SnRK1 beta subunit, expression of SnRK1 marker genes, and endogenous trehalose pathway genes were also altered. Furthermore, leaves of the transgenic maize maintained a higher photosynthetic rate for a longer period compared to wild type. In conclusion, we found that decreasing T6P in reproductive tissues down-regulates primary metabolism and up-regulates secondary metabolism, resulting in different metabolite profiles in component tissues. Our data implicate T6P/ SnRK1 as a major regulator of whole-plant resource allocation for crop yield improvement.

A critical early step in drug discovery is the screening of a chemical library. Typically, promising compounds are identified in a primary screen and then more fully characterized in a dose—response ...analysis with 7—10 data points per compound. Here, we describe a robust microfluidic approach that increases the number of data points to approximately 10,000 per compound. The system exploits Taylor—Aris dispersion to create concentration gradients, which are then segmented into picoliter microreactors by droplet-based microfluidics. The large number of data points results in IC50 values that are highly precise (±2.40% at 95% confidence) and highly reproducible (CV = 2.45%, n = 16). In addition, the high resolution of the data reveals complex dose—response relationships unambiguously. We used this system to screen a chemical library of 704 compounds against protein tyrosine phosphatase 1B, a diabetes, obesity, and cancer target. We identified a number of novel inhibitors, the most potent being sodium cefsulodine, which has an IC50 of 27 ± 0.83 μM.

Theories of how people learn relationships between continuous variables have tended to focus on two possibilities: one, that people are estimating explicit functions, or two that they are performing ...associative learning supported by similarity. We provide a rational analysis of function learning, drawing on work on regression in machine learning and statistics. Using the equivalence of Bayesian linear regression and Gaussian processes, which provide a probabilistic basis for similarity-based function learning, we show that learning explicit rules and using similarity can be seen as two views of one solution to this problem. We use this insight to define a rational model of human function learning that combines the strengths of both approaches and accounts for a wide variety of experimental results.

An examination of five temperature and five precipitation extreme indicators reveals an increase in both temperature and precipitation extremes over the 1926–2000 period in the northeast United ...States, with most of this increase occurring over the past four decades. Empirical orthogonal function (EOF) analysis of winter frost days (FD) and warm nights (TN90) and also winter consecutive dry days (CDD) and very wet days (R95T) over the 1950–2000 period reveals that some of the variability associated with changes in these extremes may be explained by variations in the Arctic Oscillation (AO), El Niño–Southern Oscillation (ENSO), and Pacific–North American (PNA) pattern. The most prominent feature of these results was the high correlation between the leading EOF of frost days and warm nights and the AO. Winter composites of temperature and precipitation extreme indicators were examined for different phases of the AO and ENSO during the 1926–2000 period. Overall, the AO is a better predictor of winter warm nights, while the ENSO is a better predictor of consecutive dry days in the northeast United States.

To determine the clinical manifestations and outcome of shigellosis among children infected with different species of Shigella.
We identified all patients <15 years infected with Shigella admitted to ...the icddr, b Dhaka hospital during one year. Study staff reviewed admission records and repeated the physical examinations and history of patients daily.
Of 792 children with shigellosis 63% were infected with S. flexneri, 20% with S. dysenteriae type 1, 10% with S. boydii, 4% with S. sonnei, and 3% with S. dysenteriae types 2-10. Children infected with S. dysenteriae type 1, when compared to children infected with other species, were significantly (P<0.05) more likely to have severe gastrointestinal manifestations: grossly bloody stools (78% vs. 33%), more stools in the 24 h before admission (median 25 vs. 11), and rectal prolapse (52% vs. 15%)--and extra-intestinal manifestations--leukemoid reaction (22% vs. 2%), hemolytic-uremic syndrome (8% vs. 1%), severe hyponatremia (58% vs. 26%) and neurologic abnormalities (24% vs. 16%). The overall fatality rate was 10% and did not differ significantly by species. In a multiple regression analysis young age, malnutrition, hyponatremia, lesser stool frequency, documented seizure, and unconsciousness were predictive of death.
Both severe intestinal disease and extra-intestinal manifestations of shigellosis occur with infection by any of the four species of Shigella, but are most common with S. dysenteriae type 1. Among these inpatient children, the risk of death was high with infection of any of the four Shigella species.

Extinct lamniform sharks (Elasmobranchii: Lamniformes) are well represented in the late Mesozoic‒Cenozoic fossil record, yet their biology is poorly understood because they are mostly represented ...only by their teeth. Here, we present measurements taken from specimens of all 13 species of extant macrophagous lamniforms to generate functions that would allow estimations of body, jaw, and dentition lengths of extinct macrophagous lamniforms from their teeth. These quantitative functions enable us to examine the body size distribution of all known macrophagous lamniform genera over geologic time. Our study reveals that small body size is plesiomorphic for Lamniformes. There are four genera that included at least one member that reached >6 m during both the Mesozoic and Cenozoic, most of which are endothermic. The largest form of the genus Otodus, O. megalodon ('megatooth shark') that reached at least 14 m, is truly an outlier considering that all other known macrophagous lamniforms have a general size limit of 7 m. Endothermy has previously been proposed to be the evolutionary driver for gigantism in Lamniformes. However, we contend that ovoviviparous reproduction involving intrauterine cannibalism, a possible synapomorphy of Lamniformes, to be another plausible driver for the evolution of endothermy achieved by certain lamniform taxa.

Most paleoclimate studies of Mainland Southeast Asia hydroclimate focus on the summer monsoon, with few studies investigating rainfall in other seasons. Here, we present a multiproxy stalagmite ...record (45,000 to 4,000 years) from central Vietnam, a region that receives most of its annual rainfall in autumn (September-November). We find evidence of a prolonged dry period spanning the last glacial maximum that is punctuated by an abrupt shift to wetter conditions during the deglaciation at ~14 ka. Paired with climate model simulations, we show that sea-level change drives autumn monsoon rainfall variability on glacial-orbital timescales. Consistent with the dry signal in the stalagmite record, climate model simulations reveal that lower glacial sea level exposes land in the Gulf of Tonkin and along the South China Shelf, reducing convection and moisture delivery to central Vietnam. When sea level rises and these landmasses flood at ~14 ka, moisture delivery to central Vietnam increases, causing an abrupt shift from dry to wet conditions. On millennial timescales, we find signatures of well-known Heinrich Stadials (HS) (dry conditions) and Dansgaard-Oeschger Events (wet conditions). Model simulations show that during the dry HS, changes in sea surface temperature related to meltwater forcing cause the formation of an anomalous anticyclone in the Western Pacific, which advects dry air across central Vietnam, decreasing autumn rainfall. Notably, sea level modulates the magnitude of millennial-scale dry and wet phases by muting dry events and enhancing wet events during periods of low sea level, highlighting the importance of this mechanism to autumn monsoon variability.
Languages are transmitted from person to person and generation to generation via a process of iterated learning: people learn a language from other people who once learned that language themselves. ...We analyze the consequences of iterated learning for learning algorithms based on the principles of Bayesian inference, assuming that learners compute a posterior distribution over languages by combining a prior (representing their inductive biases) with the evidence provided by linguistic data. We show that when learners sample languages from this posterior distribution, iterated learning converges to a distribution over languages that is determined entirely by the prior. Under these conditions, iterated learning is a form of Gibbs sampling, a widely‐used Markov chain Monte Carlo algorithm. The consequences of iterated learning are more complicated when learners choose the language with maximum posterior probability, being affected by both the prior of the learners and the amount of information transmitted between generations. We show that in this case, iterated learning corresponds to another statistical inference algorithm, a variant of the expectation‐maximization (EM) algorithm. These results clarify the role of iterated learning in explanations of linguistic universals and provide a formal connection between constraints on language acquisition and the languages that come to be spoken, suggesting that information transmitted via iterated learning will ultimately come to mirror the minds of the learners.

The question of how much the outcomes of cultural evolution are shaped by the cognitive capacities of human learners has been explored in several disciplines, including psychology, anthropology and ...linguistics. We address this question through a detailed investigation of transmission chains, in which each person passes information to another along a chain. We review mathematical and empirical evidence that shows that under general conditions, and across experimental paradigms, the information passed along transmission chains will be affected by the inductive biases of the people involved-the constraints on learning and memory, which influence conclusions from limited data. The mathematical analysis considers the case where each person is a rational Bayesian agent. The empirical work consists of behavioural experiments in which human participants are shown to operate in the manner predicted by the Bayesian framework. Specifically, in situations in which each person's response is used to determine the data seen by the next person, people converge on concepts consistent with their inductive biases irrespective of the information seen by the first member of the chain. We then relate the Bayesian analysis of transmission chains to models of biological evolution, clarifying how chains of individuals correspond to population-level models and how selective forces can be incorporated into our models. Taken together, these results indicate how laboratory studies of transmission chains can provide information about the dynamics of cultural evolution and illustrate that inductive biases can have a significant impact on these dynamics.
