A versatile terpene synthase (LcTPS2) producing unconventional macrocyclic terpenoids was characterized from Leucosceptrum canum. Engineered Escherichia coli and Nicotiana benthamiana expressing ...LcTPS2 produced six 18‐/14‐membered sesterterpenoids including five new ones and two 14‐membered diterpenoids. These products represent the first macrocyclic sesterterpenoids from plants and the largest sesterterpenoid ring system identified to date. Two variants F516A and F516G producing approximately 3.3‐ and 2.5‐fold, respectively, more sesterterpenoids than the wild‐type enzyme were engineered. Both 18‐ and 14‐membered ring sesterterpenoids displayed significant inhibitory activity on the IL‐2 and IFN‐γ production of T cells probably via inhibition of the MAPK pathway. The findings will contribute to the development of efficient biocatalysts to create bioactive macrocyclic sesterterpenoids, and also herald a new potential in the well‐trodden territory of plant terpenoid biosynthesis.
A cryptic plant terpene cyclase producing unconventional macrocyclic sesterterpenoids and diterpenoids was characterized from Leucosceptrum canum. Two variants producing more sesterterpenoids were engineered, and the immunosuppressive activity of these sesterterpenoids was found.

Glycoside hydrolase 12 (GH12) proteins act as virulence factors and pathogen‐associated molecular patterns (PAMPs) in oomycetes. However, the pathogenic mechanisms of fungal GH12 proteins ...have not been characterized. In this study, we demonstrated that two of the six GH12 proteins produced by the fungus Verticillium dahliae Vd991, VdEG1 and VdEG3 acted as PAMPs to trigger cell death and PAMP‐triggered immunity (PTI) independent of their enzymatic activity in Nicotiana benthamiana. A 63‐amino‐acid peptide of VdEG3 was sufficient for cell death‐inducing activity, but this was not the case for the corresponding peptide of VdEG1. Further study indicated that VdEG1 and VdEG3 trigger PTI in different ways: BAK1 is required for VdEG1‐ and VdEG3‐triggered immunity, while SOBIR1 is specifically required for VdEG1‐triggered immunity in N. benthamiana. Unlike oomycetes, which employ RXLR effectors to suppress host immunity, a carbohydrate‐binding module family 1 (CBM1) protein domain suppressed GH12 protein‐induced cell death. Furthermore, during infection of N. benthamiana and cotton, VdEG1 and VdEG3 acted as PAMPs and virulence factors, respectively indicative of host‐dependent molecular functions. These results suggest that VdEG1 and VdEG3 associate differently with BAK1 and SOBIR1 receptor‐like kinases to trigger immunity in N. benthamiana, and together with CBM1‐containing proteins manipulate plant immunity.

We proposed a method to evaluate the reliability of gantry cranes and address current shortcomings in reliability evaluations. First, according to gantry crane characteristics, an evaluation system ...and fault tree model were established. Fault tree analysis (FTA) was then transformed into a Bayesian network topology model. Second, the prior probability of basic events was determined from expert scoring. Finally, the reliability of the whole crane was evaluated. The proposed method was applied to a company’s gantry crane. The result indicated that failure probability could be calculated, and the root cause of failure could be precisely diagnosed. Fuzzy mathematical analysis could only evaluate risk levels of gantry cranes, and FTA provided only the minimum cut sets. The proposed method could precisely identify more risk factors than fuzzy mathematical analysis and FTA. This study confirmed that the proposed methodology was a more efficient reliability evaluation tool.

Currently, there is no consensus on the most appropriate technique for obtaining lateral hip radiographs after cephalomedullary nail (CMN) surgery. The aim of this study was to investigate the ...distribution of two commonly used postoperative lateral hip radiographic methods (classic lateral view and modified lateral view) and try to find out which one is better suited for this situation.
A retrospective analysis was conducted on 146 patients who underwent surgical fixation for extracapsular hip fractures between January 2018 and June 2022. The main outcome measured was the angle between the straight part of the CMN and the lag screw/blade on hip lateral X-rays (CMNA). The lateral hip radiographs were categorized into two groups based on different lateral hip radiographic methods. CMNA, patient age, gender, fracture classification based on the 2018 AO classification, nail length (short/long), surgical side (left/right), height, weight, BMI, preoperative waiting time, postoperative imaging interval were collected and compared between the two groups.
The distribution trend of CMNA significantly differs between two types of hip joint lateral radiographic methods. Specifically, the classic lateral method exhibits a significantly bimodal and skewed distribution with a median (p25, p75) of -21.6° (-31.2°, -8°), whereas the modified lateral method presents a normal distribution with a mean ± SD of +7.57° ± 14.4°. The difference in the Mean Rank between the classic (47.10) and the modified (102.96) lateral methods is statistically significant (P < 0.001).
The CMNA method is an excellent tool for studying the lateral distribution.We recommend using the modified lateral view as the preferred option for obtaining lateral hip radiographs after CMN surgery due to its superior distribution of CMNA and greater patient-friendliness.

Verticillium dahliae is a broad host-range pathogen that causes vascular wilts in plants. Interactions between three hosts and specific V. dahliae genotypes result in severe defoliation. The ...underlying mechanisms of defoliation are unresolved.
Genome resequencing, gene deletion and complementation, gene expression analysis, sequence divergence, defoliating phenotype identification, virulence analysis, and quantification of V. dahliae secondary metabolites were performed.
Population genomics previously revealed that G-LSR2 was horizontally transferred from the fungus Fusarium oxysporum f. sp. vasinfectum to V. dahliae and is exclusively found in the genomes of defoliating (D) strains. Deletion of seven genes within G-LSR2, designated as VdDf genes, produced the nondefoliation phenotype on cotton, olive, and okra but complementation of two genes restored the defoliation phenotype. Genes VdDf5 and VdDf6 associated with defoliation shared homology with polyketide synthases involved in secondary metabolism, whereas VdDf7 shared homology with proteins involved in the biosynthesis of N-lauroylethanolamine (N-acylethanolamine (NAE) 12:0), a compound that induces defoliation. NAE overbiosynthesis by D strains also appears to disrupt NAE metabolism in cotton by inducing overexpression of fatty acid amide hydrolase.
The VdDfs modulate the synthesis and overproduction of secondary metabolites, such as NAE 12:0, that cause defoliation either by altering abscisic acid sensitivity, hormone disruption, or sensitivity to the pathogen.

Prolyl thioesters have shown significantly lower reactivities in native chemical ligation (NCL) in comparison to that of the alanyl thioester. This report describes a mild and efficient internal ...activation protocol of peptidyl prolyl thioesters in NCL without using any thiol-based additives, where the introduction of a 4-mercaptan substituent on the C-terminal proline significantly improves the reactivity of prolyl thioesters via the formation of a bicyclic thiolactone intermediate. The kinetic data indicate that the reaction rate is comparable to that of the reported data of alanyl thioesters, and the mechanistic studies suggest that the ligation of two peptide segments proceeds through an NCL-like pathway instead of a direct aminolysis, which ensures the chemoselectivity and compatibility of various amino acid side chains. This 4-mercaptoprolyl thioester-based protocol also allows an efficient one-pot ligation–desulfurization procedure. The utility of this method has been further demonstrated in the synthesis of a proline-rich region of Wilms tumor protein 1.

This review compares the effects of peripheral dexamethasone and dexmedetomidine on postoperative analgesia. We included six randomized controlled trials (354 patients) through a systematic ...literature search. We found that analgesia duration was comparable between dexamethasone and dexmedetomidine (58.59 min, 95% CI (confidence interval), − 66.13, 183.31 min) with extreme heterogeneity. Secondary outcome was also compared and no significant difference was observed in sensory block onset and duration and motor block duration and also for postoperative nausea and vomiting. It is noteworthy that dexamethasone reduced analgesic consumption (fentanyl) by 29.12 mcg compared with dexmedetomidine. We performed subgroup analyses and found no significant difference between the following: (1) lidocaine vs ropivacaine (

= 0.28), (2) nerve block vs nerve block + general anesthesia (

= 0.47), and (3) upper limb surgery vs thoracoscopic pneumonectomy (

We applied trial sequential analysis to assess the risks of type I and II errors and concluded that the meta-analysis was insufficiently powered to answer the clinical question, and further analysis is needed to establish which adjuvant is better. In conclusion, we believe that existing research indicates that dexamethasone and dexmedetomidine have equivalent analgesic effects in peripheral nerve blocks.

Engineering structures are subjected to strain and deflection, due to various loads, and/or environmental effects. It is, thus, of uttermost importance to monitor the strain condition of ...critical structures, so as to prevent catastrophic failures, but also to minimize maintenance costs. In order to overcome limitations of existing conventional strain sensors, including extensive cabling arrangement and continuous power supply, a newly dual‐interrogation‐mode radio frequency identification (RFID) strain sensor is proposed in this study to achieve a longer interrogation transmission distance for wireless strain sensing, which can automatically switch between passive modes with low power consumption and active modes with ultra‐high frequency (UHF). The proposed design scheme involves the RFID tag and RFID reader for wireless strain transmission module and the improved Wheatstone bridge as strain measurement module. The proposed RFID strain sensor features the following merits: (i) Wireless strain sensing characteristics with the integration of RFID technology. (ii) Long transmission distance with dual interrogation mode: RFID tag is generally in a passive dormant state with an extremely low operating current. And the tag circuit will enter the working state in ultra‐high frequency to collect strain data only when the RFID reader enters the interrogation area, achieving semi‐active strain sensing with low energy consumption at a long distance (up to 80 m), which is especially suitable for practical strain measurement of engineering structures. (iii) Temperature self‐compensation characteristic: The developed RFID sensor includes a temperature compensation strain gauge to offset the error caused by temperature change, which will improve the measurement accuracy. Finally, extensive experiments are conducted to characterize the measurement performance, including thermal stability, tensile, and compressive strain sensing for various engineering materials.

Fungal transcription factors (TFs) implicated in the regulation of virulence gene expression have been identified in a number of plant pathogens. In Verticillium dahliae, despite its ...agricultural importance, few regulators of transcription have been characterized. In this study, a T‐DNA insertion mutant with significantly reduced virulence towards cotton was identified. The T‐DNA was traced to VdFTF1, a gene encoding a TF containing a Fungal_trans domain. Transient expression in onion epidermal cells indicated that VdFTF1 is localized to the nucleus. The VdFTF1‐deletion strains displayed normal vegetative growth, mycelial pigmentation and conidial morphology, but exhibited significantly reduced virulence on cotton, suggesting that VdFTF1 is required exclusively for pathogenesis. Comparisons of global transcription patterns of wild‐type and VdFTF1‐deletion strains indicated that VdFTF1 affected the expression of 802 genes, 233 of which were associated with catalytic processes. These genes encoded 69 potentially secreted proteins, 43 of which contained a carbohydrate enzyme domain known to participate in pathogenesis during infection of cotton. Targeted gene deletion of one VdFTF1‐regulated gene resulted in significantly impaired vascular colonization, as measured by quantitative polymerase chain reaction, as well as aggressiveness and symptom severity in cotton. In conclusion, VdFTF1, which encodes a TF containing a Fungal_trans domain, regulates the gene expression of plant cell wall degradation enzymes in V. dahliae, which are required for full virulence on cotton.
