Respiratory Syncytial Virus (RSV) is an important cause of respiratory infection in humans. Severe cases are common in children ≤2 years old, immunocompromised individuals, and the elderly. In 2020, ...RSV infection reduced in Rio Grande do Sul (RS), southern Brazil; however, in 2021 resurgence of RSV was observed. This study analyzed epidemiological and genetic features of RSV infection cases reported in 2021 in RS. Nasopharyngeal samples collected from individuals with respiratory infection negative for SARS‐CoV‐2, Influenza A and B viruses were assessed for the presence of RSV by real time RT‐qPCR. RSV‐A and RSV‐B genomic sequencing and phylogenetic reconstructions were performed for genotyping and clade characterization. Among 21,035 respiratory samples analyzed, 2,947 were positive for RSV, 947 of which were hospitalized patients. Positive cases were detected year‐round, with the highest number in June–July (winter). Children <1 year comprised 56.28% (n = 533) of the hospitalized patients infected with RSV, whereas 14.46% (n = 137) were individuals >60 years. Of a total of 361 deaths, 14.68% (n = 53) were RSV positive, mostly patients >60 years old (73.58%, n = 39). Chronic kidney disease, cardiopathy, Down syndrome and neurological diseases were associated with RSV infection. RSV‐A was identified in 58.5% (n = 117/200) of the patients, and RSV‐B in 41.5% (n = 83/200). Of 95 RSV genomes recovered from SARI cases, 66 were RSV‐A GA.2.3.5 genotype, while 29 were RSV‐B GB.5.0.5a genotype. This study provides epidemiological and molecular data on RSV cases in RS during the COVID‐19 pandemic and highlights that investigation of different respiratory viruses is essential for decision‐making and disease prevention and control measures.

We describe the main analytical approaches focused on both overall and specific causes of death for Brazil and Brazilian states.
There was an overall improvement in life expectancy at birth from 1990 to 2015, but with important heterogeneity among states. Reduced mortality due to diarrhea, lower respiratory infections, and other infectious diseases contributed the most for increasing life expectancy in most states from the North and Northeast regions. Reduced mortality due to cardiovascular diseases was the highest contributor in the South, Southeast, and Center West regions. However, among men, intentional injuries reduced life expectancy in 17 out of 27 states. Although age-standardized rates due to ischemic heart disease (IHD) and cerebrovascular disease declined over time, these remained the leading CODs in the country and states. In contrast, leading causes of premature mortality changed substantially - e.g., diarrheal diseases moved from 1st to 13th and then the 36th position in 1990, 2005, and 2015, respectively, while violence moved from 7th to 1st and to 2nd. Overall, the total age-standardized years of life lost (YLL) rate was reduced from 1990 to 2015, bringing the burden of premature deaths closer to expected rates given the country's Socio-demographic Index (SDI). In 1990, IHD, stroke, diarrhea, neonatal preterm birth complications, road injury, and violence had ratios higher than the expected, while in 2015 only violence was higher, overall and in all states, according to the SDI.
A widespread reduction of mortality levels occurred in Brazil from 1990 to 2015, particularly among children under 5 years old. Major shifts in mortality rates took place among communicable, maternal, neonatal, and nutritional disorders. The mortality profile has shifted to older ages with increases in non-communicable diseases as well as premature deaths due to violence. Policymakers should address health interventions accordingly.

Nematodes and drought are major constraints in tropical agriculture and often occur simultaneously. Plant responses to these stresses are complex and require crosstalk between biotic and abiotic ...signaling pathways. In this study, we explored the transcriptome data of wild Arachis species subjected to drought (A-metaDEG) and the root-knot nematode Meloidogyne arenaria (B-metaDEG) via meta-analysis, to identify core-stress responsive genes to each individual and concurrent stresses in these species. Transcriptome analysis of a nematode/drought bioassay (cross-stress) showed that the set of stress responsive DEGs to concurrent stress is distinct from those resulting from overlapping A- and B-metaDEGs, indicating a specialized and unique response to combined stresses in wild Arachis. Whilst individual biotic and abiotic stresses elicit hormone-responsive genes, most notably in the jasmonic and abscisic acid pathways, combined stresses seem to trigger mainly the ethylene hormone pathway. The overexpression of a cross-stress tolerance candidate gene identified here, an endochitinase-encoding gene (AsECHI) from Arachis stenosperma, reduced up to 30% of M. incognita infection and increased post-drought recovery in Arabidopsis plants submitted to both stresses. The elucidation of the network of cross-stress responsive genes in Arachis contributes to better understanding the complex regulation of biotic and abiotic responses in plants facilitating more adequate crop breeding for combined stress tolerance.

Dam failure events can result in a high suspended solids concentration in surface waters. To understand the impact of high suspended solids concentration on the treatment process of water treatment ...plants, this study investigated coagulation-flocculation stages using different coagulants (ferric chloride and a polymeric coagulant – CAT-FLOC 8799 Plus) and solids concentrations (2000–46,000 mg/L), and how they impacted on the mean diameter (d50), density (ρs), and settling velocity (vs) of the agglomerates formed. After the sedimentation process, the turbidity values varied between 201–26 NTU when FeCl3 was used as a coagulant, and 36–11 NTU when using the organic coagulant. The study reinforces the effect of particle diameter on sedimentation velocity, which is lower for cases involving smaller average diameters. Even so, solids removal was high regardless of the initial conditions and characteristics of the agglomerated particles, starting early in flocculators with removals >80 %. The results point to the need for frequent maintenance of these units to remove the sludge settled, which could compromise the operation of water treatment plants. In this sense, two alternatives were proposed as pre- and post-treatment. Under high total solids concentration (>20,000 mg/L), pre-treatment with thickeners becomes mandatory, and ultrafiltration comes as an additional barrier that will prevent high turbidity values in drinking water. In this study, the use of ultrafiltration membranes in partial or total replacement of sand filters showed stable performance (permeate flow: 130–136 L/m2h) with no tendency to fouling and guaranteeing a residual turbidity value <0.3 NTU. Future studies should prove the technical and operational feasibility of the strategies proposed, which until then showed promising results to guarantee the operation of water treatment plants in adverse scenarios.

The aim of this study was to evaluate the shear bond strength of resin cement and lithium disilicate ceramic after various surface treatments of the ceramic. Sixty blocks of ceramic (IPS e.max Press, ...Ivoclar Vivadent) were obtained. After cleaning, they were placed in polyvinyl chloride tubes with acrylic resin. The blocks were divided into six groups (n=10) depending on surface treatment: H/S/A - 10% Hydrofluoric Acid + Silane + Adhesive, H/S -10% Hydrofluoric Acid + Silane, H/S/UA - 10% Hydrofluoric Acid + Silane + Universal Adhesive, H/UA- 10% Hydrofluoric Acid + Universal Adhesive, MBEP/A - Monobond Etch & Prime + Adhesive, and MBEP - Monobond Etch & Prime. The light-cured resin cement (Variolink Esthetic LC, Ivoclar Vivadent) was inserted in a mold placed over the treated area of the ceramics and photocured with an LED for 20 s to produce cylinders (3 mm x 3 mm). The samples were subjected to a shear bond strength test in a universal test machine (Instron 5965) by 0.5 mm/min. ANOVA and Tukey tests showed a statistically significant difference between groups (p<0.05). The results of the shear strength test were H/S/A (9.61±2.50)A, H/S (10.22±3.28)A, H/S/UA (7.39±2.02)ABC, H/UA (4.28±1.32)C, MBEP/A (9.01±1.97)AB, and MBEP (6.18±2.75)BC. The H/S group showed cohesive failures, and the H/UA group was the only one that presented adhesive failures. The conventional treatment with hydrofluoric acid and silane showed the best bond strength. The use of a new ceramic primer associated with adhesive bonding obtained similar results to conventional surface treatment, being a satisfactory alternative to replace the use of hydrofluoric acid.

Protected and conserved areas (PCAs) are increasingly recognized as essential tools for their effectiveness in conservation and the benefits they provide. However, their challenges are still ...significant. The concepts, legislation, and governance surrounding PCAs are the results of social contexts. Due to the evolution of scientific knowledge, human rights, and diversified demands, new approaches are necessary to fulfill their functions. To better understand the context and possibilities, a study group was established to evaluate the current research, exchange experiences, guide dialogues, and identify lessons from experiences. The core of the experiences and cases considered and the reflections developed focused on the Brazilian context. This article analyzes the collected information and reflections related to several themes associated with challenges. The results reinforce the importance of PCAs but warn about the limitations of current conservation strategies to respond to social actors’ expectations, the needs of the vulnerable social groups, and evolving demands. The complexity of PCA systems is evident in view of the multiplicity of interests, potential contributions, and possibilities for participatory arrangements. There is a need to improve management and governance conceptions to promote the reconnection between society and nature. Therefore, the concept of collaborative conservation is proposed as an instrumental approach to advance towards inclusive and effective conservation strategies.
Abstract
The emergence and global dissemination of Severe Acute Respiratory Syndrome virus 2 (SARS-CoV-2) variants of concern (VOCs) have been described as the main factor driving the Coronavirus ...Disease 2019 pandemic. In Brazil, the Gamma variant dominated the epidemiological scenario during the first period of 2021. Many Brazilian regions detected the Delta variant after its first description and documented its spread. To monitor the introduction and spread of VOC Delta, we performed Polymerase Chain Reaction (PCR) genotyping and genome sequencing in ten regional sentinel units from June to October 2021 in the State of Minas Gerais (MG). We documented the introduction and spread of Delta, comprising 70 per cent of the cases 8 weeks later. Comparing the viral loads of the Gamma and Delta dominance periods, we provide additional evidence that the latter is more transmissible. The spread and dominance of Delta did not culminate in the increase in cases and deaths, suggesting that the vaccination may have restrained the epidemic growth. Analysis of 224 novel Delta genomes revealed that Rio de Janeiro state was the primary source for disseminating this variant in the state of MG. We present the establishment of Delta, providing evidence of its enhanced transmissibility and showing that this variant shift did not aggravate the epidemiological scenario in a high immunity setting.

Additionally, a large resistome was observed among all analyzed hosts, with emphasis on the presence of carbapenemase-encoding genes not previously reported in Latin America.
Humans and food production animals have been reported to be important reservoirs of antimicrobial resistance (AMR) genes (ARGs). The frequency of these multidrug-resistant (MDR) bacteria tends to be higher in low- and middle-income countries (LMICs), due mainly to a lack of public health policies. Although studies on AMR in humans or animals have been carried out in Brazil, this is the first multicenter study that simultaneously collected rectal swabs from humans and food-producing animals for metagenomics. Our results indicate high microbial diversity among all analyzed hosts, and several ARGs for different antimicrobial classes were also found. Platelets drive endothelial cell activation in many diseases. However, if this occurs in Plasmodium vivax malaria is unclear. As platelets have been reported to be activated and to play a role in ...inflammatory response during malaria, we hypothesized that this would correlate with endothelial alterations during acute illness. We performed platelet flow cytometry of PAC-1 and P-selectin. We measured platelet markers (CXCL4, CD40L, P-selectin, Thrombopoietin, IL-11) and endothelial activation markers (ICAM-1, von Willebrand Factor and E-selectin) in plasma with a multiplex-based assay. The values of each mediator were used to generate heatmaps, K-means clustering and Principal Component analysis. In addition, we determined pair-wise Pearson's correlation coefficients to generate correlation networks. Platelet counts were reduced, and mean platelet volume increased in malaria patients. The activation of circulating platelets in flow cytometry did not differ between patients and controls. CD40L levels (Median IQ: 517 406-651 vs. 1029 732-1267 pg/mL, P = 0.0001) were significantly higher in patients, while P-selectin and CXCL4 showed a nonsignificant trend towards higher levels in patients. The network correlation approach demonstrated the correlation between markers of platelet and endothelial activation, and the heatmaps revealed a distinct pattern of activation in two subsets of P. vivax patients when compared to controls. Although absolute platelet activation was not strong in uncomplicated vivax malaria, markers of platelet activity and production were correlated with higher endothelial cell activation, especially in a specific subset of patients.
