Schistosomiasis is a zoonotic parasitic disease. Schistosoma japonicum eggs deposited in the liver tissue induce egg granuloma formation and liver fibrosis, seriously threatening human health. ...Natural killer (NK) cells kill activated hepatic stellate cells (HSCs) or induce HSC apoptosis and inhibit the progression of liver fibrosis. However, the function of NK cells in liver fibrosis caused by S. japonicum infection is significantly inhibited. The mechanism of this inhibition remains unclear. Twenty mice were percutaneously infected with S. japonicum cercariae. Before infection and 2, 4, 6, and 8 weeks after infection, five mice were euthanized and dissected at each time point. Hepatic NK cells were isolated and transcriptome sequenced. The sequencing results showed that Tigit expression was high at 4-6 weeks post infection. This phenomenon was verified by reverse transcription quantitative PCR (RT-qPCR) and flow cytometry. NK cells derived from Tigit-/- and wild-type (WT) mice were co-cultured with HSCs. It was found that Tigit-/- NK cells induced apoptosis in a higher proportion of HSCs than WT NK cells. Schistosomiasis infection models of Tigit-/- and WT mice were established. The proportion and killing activity of hepatic NK cells were significantly higher in Tigit-/- mice than in WT mice. The degree of liver fibrosis in Tigit-/- mice was significantly lower than that in WT mice. NK cells were isolated from Tigit-/- and WT mice and injected via the tail vein into WT mice infected with S. japonicum. The degree of liver fibrosis in mice that received NK cell infusion reduced significantly, but there was no significant difference between mice that received NK cells from Tigit-/- and WT mice, respectively. Our findings indicate that Tigit knockout enhanced the function of NK cells and reduced the degree of liver fibrosis in schistosomiasis, thus providing a novel strategy for treating hepatic fibrosis induced by schistosomiasis.

According to the microglial hypothesis of schizophrenia, the hyperactivation of microglia and the release of proinflammatory cytokines lead to neuronal loss, which is highly related to the onset of ...schizophrenia. Recent studies have demonstrated that fractalkine (CX3CL1) and its receptor CX3CR1 modulate the function of microglia. Thus, the present study aimed to determine whether microglial CX3CR1 plays a role in schizophrenia-related behaviors. A classical animal model of schizophrenia, social isolation (from postnatal days 21 to 56), was used to induce schizophrenia-related behaviors in C57BL/6J and CX3CR1-/- mice, and the expression of the microglial CX3CR1 protein was examined in several brain areas of the C57BL/6J mice by Western blot analysis. The results revealed that social isolation caused deficits in the prepulse inhibition (PPI) in the C57BL/6J mice but not in the CX3CR1-/- mice and increased locomotor activity in both the C57BL/6J mice and the CX3CR1-/- mice. Moreover, the CX3CR1 protein level was increased in the medial prefrontal cortex, nucleus accumbens, and hippocampus of the C57BL/6J mice. These findings suggested that the function of microglia regulated by CX3CR1 might participate in schizophrenia-related behaviors.
Gap junctions (GJs) are multiple cellular intercellular connections that allow ions to pass directly into the cytoplasm of neighboring cells. Electrical coupling mediated by GJs plays a role in the ...generation of highly synchronous electrical activity. Accumulative investigations show that GJs in the brain are involved in the generation, synchronization and maintenance of seizure events. At the same time, GJ blockers exert potent curative potential on epilepsy in vivo or in vitro. This review aims to shed light on the role of GJs in epileptogenesis. Targeting GJs is likely to be served as a novel therapeutic approach on epileptic patients.

Coiled tubing (CT) technology is widely used in oil and gas industry. In the operation process, low cycle fatigue failure is easy to occur. Therefore it is necessary to predict the fatigue life of CT. In this paper, a new theoretical model of low cycle fatigue life for CT is established, the feasibility of the theoretical model is verified by experimental result and Finite Element (FE) value. This method effectively solves the problem that the result of the previous model is not more accurate at some conditions. Using the theoretical model, the effect of CT steel grade, wall thickness, outer diameter (OD), reel radius, guider radius and internal pressure on the fatigue life are studied by using the theoretical model. The result shows that the reel radius, CT OD and internal pressure are the sensitive parameters that affect the fatigue life of CT.

Automatic object detection by satellite remote sensing images is of great significance for resource exploration and natural disaster assessment. To solve existing problems in remote sensing image ...detection, this article proposes an improved YOLOX model for satellite remote sensing image automatic detection. This model is named RS-YOLOX. To strengthen the feature learning ability of the network, we used Efficient Channel Attention (ECA) in the backbone network of YOLOX and combined the Adaptively Spatial Feature Fusion (ASFF) with the neck network of YOLOX. To balance the numbers of positive and negative samples in training, we used the Varifocal Loss function. Finally, to obtain a high-performance remote sensing object detector, we combined the trained model with an open-source framework called Slicing Aided Hyper Inference (SAHI). This work evaluated models on three aerial remote sensing datasets (DOTA-v1.5, TGRS-HRRSD, and RSOD). Our comparative experiments demonstrate that our model has the highest accuracy in detecting objects in remote sensing image datasets.
Dihydroartemisinin (DHA), a semi-synthetic derivative of artemisinin, is known to play important roles in inhibiting proliferation rate, inducing apoptosis, as well as hindering the metastasis and ...invasion of glioma cells, but the underlying mechanisms are still unclear so far. In this study, methyl thiazolyl tetrazolium (MTT), colony-forming, wound healing, invasion, and apoptosis assays were performed to investigate the effect of DHA on malignant glioma cells. Results showed that DHA induced apoptosis of malignant glioma cells through Protein Kinase B (AKT) axis, induced death of malignant glioma cells by downregulating miR-21, and inhibited the invasion of malignant glioma cells corresponding with up-regulation of the reversion-inducing-cysteine-rich protein with kazal motifs (RECK). These results revealed that AKT axis, miR-21, and RECK play pivotal roles in DHA killing malignant glioma cells, suggesting that DHA is a potential agent for treating glioma.

Degrees of freedom and mutual coupling are two important factors to determine the ability of direction of arrival (DOA) estimation. Although coprime planar array (CPPA) has less mutual coupling than ...hourglass array (HA) and ladder array (LA), its positive and negative covariance corresponding lags are discontinuous, resulting in low degrees of freedom. In this study, we propose a generalized and symmetric coprime planar array (GSCA) by employing the symmetric property of the difference co‐array. By moving one subarray and introducing a supplementary subarray, we construct a connected virtual uniform rectangular array (URA). Through further optimizing the sensor number, the redundant lags are reduced and the array structure is more general. We show that the degrees of freedom are much higher than those of CPPA, HA and LA. Besides, the mutual coupling is close to CPPA, but less than HA and LA. The superiority of GSCA is verified by simulations.

: Two independent researchers comprehensively searched PubMed, Embase, the Cochrane Library, and Web of Science for eligible studies published from their inception to 31 March 2022. Comparisons of risk, overall survival (OS), and disease-free survival (DFS) for patients with BTC were selected as the endpoints of interest and pooled by random or fixed-effects models.
: Eleven studies with a total of 24,788,738 participants were eligible for this analysis. The overall pooled effects showed no significant differences in biliary tract cancer risk (hazard ratio (HR) = 0.82, 95% confidence interval (CI): 0.50-1.35,
= 0.436), OS (HR = 0.88, 95% CI: 0.74-1.04,
= 0.135), or DFS (HR = 1.03, 95% CI: 0.79-1.34,
= 0.829) between metformin users and non-users. When restricting participants to those with diabetes, a similar negative result was found, demonstrating that metformin use was not significantly associated with a lower risk of developing BTC compared with a lack of metformin use (HR = 0.65, 95% CI: 0.39-1.07,
= 0.089); notably, the included studies exhibited significant heterogeneity in the selection of participants and the definition of metformin users.
: Metformin may not be able to reduce the risk of BTC and improve prognosis in certain populations. Based on the limited quantity and quality of the included studies, the present results should be interpreted within their limitations, and further studies are warranted to determine the optimal timing, dose, duration, and scenario of metformin administration.
(‑)‑Epigallocatechin‑3‑gallate (EGCG), a major constituent of green tea, is a potential anticancer agent, but the molecular mechanisms of its effects are not well‑understood. The present study was ...conducted to examine the mechanism of EGCG in lung cancer cells. Alterations in long non‑coding RNAs (lncRNAs) and mRNAs were investigated in lung cancer cells treated with EGCG by lncRNA microarray analysis. Furthermore, the functions and signaling pathways regulated by EGCG were predicted by bioinformatics analysis. A total of 960 lncRNAs and 1,434 mRNAs were significantly altered following EGCG treatment. These lncRNAs were distributed across nearly all human chromosomes and the mRNAs were involved in the cell cycle and the mitotic cell cycle process. Through a combination of microarray and bioinformatics analysis, 20 mRNAs predicted to serve a key role in the EGCG regulation were identified, and certain regulatory networks involving EGCG‑regulated lncRNAs were predicted. In conclusion, EGCG affects the expression of various lncRNAs and mRNAs in the cells, therefore affecting cell functions. The results of the present study provide an insight into the mechanism of EGCG, which may be useful for therapeutic development.
