Ewing's sarcoma/PNET, a tumor of the bone and soft tissue, is one of the most common causes of tumor death among youths. This tumor does not have specific phenotypes, but does have characteristic ...chromosomal translocations. Furthermore, the expression ofEWS/FLI-1orEWS/ERGchimeric genes was found to be generated through a t(11;22)(q24;q12) or a t(21;22)(q22;q12) translocation. In this study, we identified a new chimera gene between the transactivation domain ofEWSandE1A-F,encoding the adenovirusE1Aenhancer-binding protein. Since E1A-F is known to activate matrix metalloproteinase genes, the chimera gene may possibly be involved in tumor progression and could be a novel tumor marker for Ewing's sarcoma/PNET.

A recombinant CHO cell line (GT19) secreting a high level of human growth hormone (hGH) was constructed with amplification of the introduced hGH gene. The cells grew well in the alpha MEM medium ...supplemented with 5% dialyzed fetal calf serum (dFCS), but not with less than 1% dFCS. Therefore we examined various medium components and obtained an improved medium which supported cell growth at low serum concentrations. The production of hGH by the cells was also enhanced in this medium.
The underlying pathogenesis of stroke is mediated by a variety of environmental risk factors as well as genetic ones. Thus, we have to evaluate the environmental factors precisely to identify the ...stroke-related gene polymorphisms. The Hisayama study, an epidemiological study of cardiovascular diseases, was established in 1961 in Hisayama, Japan. In 2002, a screening survey for the genetic study was performed in Hisayama. The Fukuoka Stroke Registry (FSR) is a hospital-based registration of stroke patients. Stroke specialists from eight medical centers in southern Japan have participated in FSR. In the present study, control and case subjects were recruited from the Hisayama study and FSR, respectively. We performed a genome-wide case-control study and found that a nonsynonymous SNP in PRKCH encoding a member of protein kinase C (PKCη) was significantly associated with brain infarction. As a candidate gene analysis, we investigated the role of NAD (P) H oxidase C242T polymorphism in the development of brain infarction. The C242T polymorphism was not associated with lacunar and atherothrombotic infarction; however, the presence of T-allele may have a protective role in the occurrence of atrial fibrillation and cardioembolic brain infarction. These studies may provide important information for the development of the therapeutic strategies against stroke.
A cell line designated SCCH‐196 was established from an extraskeletal small round cell sarcoma developed in a 16‐year‐old Japanese girl. The cells grew as a monolayer, and have been continuously ...propagated by serial subcultures during the past 26 months. Cells from the primary tumor and those from the SCCH‐196 cell line at passage 10 both showed the same karyotype, 51, XX, + 8, + 20, + 21, t(11;22)(q24;q12), + i(1q), + i(1q). Histologically the primary tumor was difficult to classify as either Ewing's sarcoma (ES) or peripheral neuroepithelioma (NE). Neuron‐specific enolase‐positive cells in the primary tumor and the occurrence in the upper extremity were in favor of NE, while positive reaction of SCCH‐196 cells to an ES‐speciflc monoclonal antibody 5C11 suggested a diagnosis of ES. The SCCH‐196 cell line may be useful for basic studies on differentiation of neuroectodermal tumors, and for future cloning of still unidentified genes which may be located at the breakpoints of the 11;22 translocation.

We have developed an electromagnetic generator to bury in subcutaneous area or abdominal cavity of the birds. As we can't use a solar battery, it is extremely difficult to supply a power for ...subcutaneous implantation such as biosensors under the skin due to the darkness environment. We are aiming to test the antigen-antibody reaction to confirm an avian influenza. One solution is a very small generator with the electromagnetic induction coil. We attached the developed coil to chickens and pheasants and recorded the electric potential generated as the chicken walked and the pheasant flew. The electric potential generated with physical simulator is equal to or exceeds the 7 V peak-to-peak at maximum by 560/min of flapping of wings. Even if we account for the junction voltage of the diode (200 mV), efficient charging of the double-layer capacitor is possible with the voltage doubler rectifier. If we increase the voltage, other problems arise, including the high-voltage insulation of the double-layer capacitor. For this reason, we believe the power generated to be sufficient for subcutaneous area of birds. The efficiency, magnetic 2 mm in length and coil 15mm in length, if axial direction is rectified, the magnetic flux density given to the coil could calculated to 7.1 % and generated power average 0.47mW. The improvements in size and wire insulation are expected in the future.

We devised a bird-borne transceiver unit for S-band packet radio communications based on the CC2500 transceiver, a device manufactured by Texas Instruments (TI). Our assessments determined the ...optimal parameters needed to achieve successful bird-to-center communication over a distance of 18 km and bird-to-bird communication over a distance of 200 m. These parameters included optimal modulation methods, transmission rates, and antennas. We equipped the transceiver unit with a modified dipole antenna (collinear antenna), which we tested in a 10 m anechoic chamber. Our experimental assessments and circuit design review identified the following parameters: 2FSK modulation method; 14.28 kHz frequency shift; 101.56 kHz IF reception bandwidth; and Manchester encoding (+). Our assessments showed bird-to-bird communications could be achieved over a distance of 200 m assuming MSK, FEC (+), and 500 kbps. Following tests by an official body, we obtained 28 sets of a type approval license for 2.4 GHz. In cooperation with the Yamashina Institute for Ornithology, we attempted to tag and release six or more swans. This unit gives us the ability to obtain vital data on swans. We expect this data to provide significant benefits, including clues on improving screening for infected individuals.
Chromosomes and histology were successfully studied in 33 childhood renal tumors. Thirty-one tumors were classified as one of four subtypes of Wilms' tumor. Of 24 typical Wilms' tumors, 12 had ...hyperdiploidy with nonrandom trisomies, mostly including +6 and/or +12. Three typical Wilms' tumors with an 11p13 deletion or a pericentric inversion with a break in 11p13 were not associated with aniridia. Two other typical Wilms' tumors with the 11p13 deletion and one fetal rhabdomyomatous nephroblastoma with an 11p13 translocation were associated with aniridia. Two cystic partially differentiated nephroblastomas showed hyperdiploidy with +12. Of four clear cell sarcomas of the kidney, three had normal diploidy and the other had a 2;22 translocation. Two congenital mesoblastic nephromas had hyperdiploid karyotype with trisomy 11, which was never seen in the 31 Wilms' tumors. Our findings and a review of data on 102 reported Wilms' tumors revealed 11p13 abnormalities in 24 tumors, 11p15 abnormalities in five tumors, and partial deletions of 1p, 7p, 11q, 12q, 16q, or 17p or monosomy of No. 21 or No. 22 each in four or more tumors. These findings suggest that increased copy number of genes on the nonrandom trisomic chromosomes might contribute to the genesis of many Wilm's tumors and that deletion of various tumor suppressor genes other than a Wilms' tumor gene, WT1 in 11p13, might also play a critical role in the development of some tumors.
We generated the homozygous transgenic mice with expression of the active form of TGF-beta1 by the glucagon promoter (homozygous NOD-TGF-beta1). The homozygous NOD-TGF-beta1 showed severe diabetes in ...84.6%, impaired glucose tolerance, and low serum insulin levels. The final size of endocrine and whole pancreas decreased, respectively, to 6 and 34%, compared to wild-type mice. The homozygous N(2) backcross to C57BL/6 (B6-TGF-beta1) showed no diabetes, but impaired glucose tolerance and low serum insulin levels. In homozygous NOD-TGF-beta1, the expression of p15(INK4b) was induced by 3.4-fold in pancreatic islets than that in wild-type mice. Based on these, we conclude first that excessive paracrine TGF-beta1 signaling in islets results in endocrine and exocrine pancreatic hypoplasia, second that TGF-beta1decrease the final size of endocrine and exocrine pancreas presumably through regulating cell cycle via p15(INK4b) at least in endocrine pancreas, and third that hypoplastic action of TGF-beta1 of pancreatic islets is independent of the genetic background.

Immunological characterization of tumour infiltrating lymphocytes (TIL) by immunohistological techniques was carried out in 20 cases of stage I seminoma. Routine pathological examination of these ...surgical specimens showed typical seminoma in 20 cases. Eighteen cases showed obvious TIL and immunohistological staining on frozen specimens was performed in 12. TIL in seminomas were predominantly T-cells but B-cells were also identified. T-cells were distributed diffusely with predominance of the CD8+ phenotype judged semiquantitatively. In contrast to the distribution of T-cells, B-cells tended to accumulate and occasionally formed lymphoid follicles. In such follicles the phenotypic pattern of B-cell antigens was comparable with secondary lymphoid follicles in lymphoid organs. There is an immunologically complex response to seminoma by the host with a predominant infiltration of cytotoxic/suppressor T-cells and functional maturation of B-cells.
