Long nanopore reads are advantageous in de novo genome assembly. However, nanopore reads usually have broad error distribution and high-error-rate subsequences. Existing error correction tools cannot ...correct nanopore reads efficiently and effectively. Most methods trim high-error-rate subsequences during error correction, which reduces both the length of the reads and contiguity of the final assembly. Here, we develop an error correction, and de novo assembly tool designed to overcome complex errors in nanopore reads. We propose an adaptive read selection and two-step progressive method to quickly correct nanopore reads to high accuracy. We introduce a two-stage assembler to utilize the full length of nanopore reads. Our tool achieves superior performance in both error correction and de novo assembling nanopore reads. It requires only 8122 hours to assemble a 35X coverage human genome and achieves a 2.47-fold improvement in NG50. Furthermore, our assembly of the human WERI cell line shows an NG50 of 22 Mbp. The high-quality assembly of nanopore reads can significantly reduce false positives in structure variation detection.
Perovskite light‐emitting diodes (PeLEDs) show great application potential in high‐quality flat‐panel displays and solid‐state lighting due to their steadily improved efficiency, tunable colors, ...narrow emission peak, and easy solution‐processing capability. However, because of high optical confinement and nonradiative charge recombination during electron–photon conversion, the highest reported efficiency of PeLEDs remains far behind that of their conventional counterparts, such as inorganic LEDs, organic LEDs, and quantum‐dot LEDs. Here a facile route is demonstrated by adopting bioinspired moth‐eye nanostructures at the front electrode/perovskite interface to enhance the outcoupling efficiency of waveguided light in PeLEDs. As a result, the maximum external quantum efficiency and current efficiency of the modified cesium lead bromide (CsPbBr3) green‐emitting PeLEDs are improved to 20.3% and 61.9 cd A−1, while retaining spectral and angular independence. Further reducing light loss in the substrate mode using a half‐ball lens, efficiencies of 28.2% and 88.7 cd A−1 are achieved, which represent the highest values reported to date for PeLEDs. These results represent a substantial step toward achieving practical applications of PeLEDs.

A record external quantum efficiency of 28.2% is realized for the device based on cesium lead bromide (CsPbBr3), while retaining the same spectral response for broad viewing angles.

Economic dispatch problem (EDP) is an important class of optimization problems in the smart grid, which aims at minimizing the total cost when generating certain amount of power. In this work, a ...novel consensus based algorithm is proposed to solve EDP in a distributed fashion. The quadratic convex cost functions are assumed in the problem formulation, and the strongly connected communication topology is sufficient for the information exchange. Unlike centralized approaches, the proposed algorithm enables generators to collectively learn the mismatch between demand and total amount of power generation. The estimated mismatch is then used as a feedback mechanism to adjust current power generation by each generator. With a tactical initial setup, eventually, all generators can automatically minimize the total cost in a collective sense.
The common cytokine receptor γ chain, γc, is a component of the receptors for interleukin-2 (IL-2), IL-4, IL-7, IL-9, IL-15, and IL-21. Mutation of the gene encoding γc results in X-linked severe ...combined immunodeficiency in humans, and γc family cytokines collectively regulate development, proliferation, survival, and differentiation of immune cells. Here, we review the basic biology of these cytokines, highlighting mechanisms of signaling and gene regulation that have provided insights for immunodeficiency, autoimmunity, allergic diseases, and cancer. Moreover, we discuss how studies of this family stimulated the development of JAK3 inhibitors and present an overview of current strategies targeting these pathways in the clinic, including novel antibodies, antagonists, and partial agonists. The diverse roles of these cytokines on a range of immune cells have important therapeutic implications.

DNA methylation is an important epigenetic mark involved in diverse biological processes. In plants, DNA methylation can be established through the RNA-directed DNA methylation pathway, an RNA ...interference pathway for transcriptional gene silencing （TGS）, which requires 24-nt small interfering RNAs. In mammals, de novo DNA methylation occurs primarily at two developmental stages： during early embryogenesis and during gametogenesis. While it is not clear whether establishment of DNA methylation patterns in mammals involves RNA interference in general, de novo DNA methylation and suppression of transposons in germ cells require 24-32-nt piwi-interacting small RNAs. DNA methylation status is dynamically regulated by DNA methylation and demethylation reactions. In plants, active DNA demethylation relies on the repressor of silencing 1 family of bifunctional DNA glycosylases, which remove the 5-methylcytosine base and then cleave the DNA backbone at the abasic site, initiating a base excision repair （BER） pathway. In animals, multiple mechanisms of active DNA demethylation have been proposed, including a deaminase- and DNA glycosylase-initiated BER pathway. New information concerning the effects of various histone modifications on the establishment and maintenance of DNA methylation has broadened our understanding of the regulation of DNA methylation. The function of DNA methylation in plants and animals is also discussed in this review.

Salt‐inducible kinase 2 (SIK2) is an important regulator in various intracellular signaling pathways related to apoptosis, tumorigenesis and metastasis. However, the involvement of SIK2 in gastric tumorigenesis and the functional linkage with gastric cancer (GC) progression remain to be defined. Here, we report that SIK2 was significantly downregulated in human GC tissues, and reduced SIK2 expression was associated with poor prognosis of patients. Overexpression of SIK2 suppressed the migration and invasion of GC cells, whereas knockdown of SIK2 enhanced cell migratory and invasive capability as well as metastatic potential. These changes in the malignant phenotype resulted from the ability of SIK2 to suppress epithelial–mesenchymal transition via inhibition of AKT/GSK3β/β‐catenin signaling. The inhibitory effect of SIK2 on AKT/GSK3β/β‐catenin signaling was mediated primarily through inactivation of AKT, due to its enhanced dephosphorylation by the upregulated protein phosphatases PHLPP2 and PP2A. The upregulation of PHLPP2 and PP2A was attributable to SIK2 phosphorylation and activation of mTORC1, which inhibited autophagic degradation of these two phosphatases. These results suggest that SIK2 acts as a tumor suppressor in GC and may serve as a novel prognostic biomarker and therapeutic target for this tumor.

Strong spin–orbit coupling, resulting in the formation of spin‐momentum‐locked surface states, endows topological insulators with superior spin‐to‐charge conversion characteristics, though the ...dynamics that govern it have remained elusive. Here, an all‐optical method is presented, which enables unprecedented tracking of the ultrafast dynamics of spin‐to‐charge conversion in a prototypical topological insulator Bi2Se3/ferromagnetic Co heterostructure, down to the sub‐picosecond timescale. Compared to pure Bi2Se3 or Co, a giant terahertz emission is observed in the heterostructure that originates from spin‐to‐charge conversion, in which the topological surface states play a crucial role. A 0.12 ps timescale is identified that sets a technological speed limit of spin‐to‐charge conversion processes in topological insulators. In addition, it is shown that the spin‐to‐charge conversion efficiency is temperature independent in Bi2Se3 as expected from the nature of the surface states, paving the way for designing next‐generation high‐speed optospintronic devices based on topological insulators at room temperature.

This paper presents a study on alkali activated cement (AAC) mortar produced with waste soda-lime-silica glass. The waste glass was used simultaneously as a precursor and fine aggregates in the alkali activated fly ash-slag mortar. The influences of waste glass in cullet and powder forms on workability, compressive and flexural strengths, fire resistance of the AAC mortar were investigated. The experimental results showed that the workability was gradually increased as the replacement level of natural sand by glass cullet was increased, and it was significantly improved with decreasing aggregates-to-binder ratios. The mechanical properties data indicated that the compressive strength was reduced as the glass cullet content increased. However, for the flexural strength, the optimum percentage of glass cullet replacement was 50%. Due to the low reactivity, a reduction in strength was observed when the glass powder was used to replace the fly ash and slag. Nevertheless, in terms of fire resistance, the incorporation of glass cullet could improve the resistance of the AAC to high temperature exposures (800 °C). In particular, the AAC mortar prepared with the glass powder as a precursor exhibited remarkable resistance to high temperature. The use of waste glass in AAC material was feasible from the mechanical properties and fire resistance points of view.

Lightweight cementitious composite has been used in construction for thousand years. However, before the Second World War, lightweight concrete was mainly limited to non-loadbearing blocks. With the increasingly demanding requirements of modern construction, especially in harsh environments, there is an urgent need to develop high strength lightweight concrete (HSLWC) capable of loading bearing purposes. The adoptions of supplementary cementitious materials, high-quality lightweight materials and fibers contribute to developing HSLWC, which enable the production of structural components. Numerous studies have been conducted on HSLWC with a compressive strength exceeding 40 MPa and a density below 2000 kg/m3. This paper aims to provide an in-depth review of HSLWC, covering its development strategies, functional and durability properties, and structural applications. Particular emphasis is given to the emerging technology of ultra high-performance lightweight concrete, characterized by low density, ultra high strength and superior durability. Compared to normal weight concrete, HSLWC offers better thermal insulation and fire resistance. The use of HSLWC in structures can increase load capacity and span, reduce seismic inertial forces, and improve structural efficiency. Numerous research and projects have demonstrated the feasibility and advantages of using HSLWC in various construction fields, including high-rise buildings, bridges and offshore structures. The excellent in-service performance of HSLWC in marine structures highlights its durability. To expand the applications of HSLWC in civil engineering, the paper proposes current opportunities and challenges that should be addressed to attract more attentions and foster further research in this area.
