The purpose of this study was to explore the needs and challenges reported by caregivers and teachers of children with autism spectrum disorders (ASD) in China. The study group consisted of a random ...sample of 108 caregivers and 98 teachers of children with ASD. Analysis of epidemiological data produced results consistent with those reported in the Western literature. Many Chinese children with ASD in this study lived with their extended families with support from older generations and the majority of the families had only one child. Financial factors were particularly important for both families and teachers. Limited professional training and social support were reported as barriers to a quality education for these children. Caregivers considered deteriorated family relationships as a predominant source of stress, whereas low employment salary and benefits appeared to be the main source of stress that diminished the teachers' commitment to educating children with special needs. Teachers with higher levels of education reported higher levels of dissatisfaction with employment salary, benefits, and home environment. In addition, the findings highlighted social-cultural influences on parenting stress and coping mechanisms. (Contains 6 tables.)
Aim: To investigate the effects of sodium danshensu on vessel function in isolated rat aortic ring.
Methods: Thoracic aortae from normal rats were isolated and equilibrated in organ bath with ...Krebs-Henseleit buffer and ring tension was recorded. Effects of sodium danshensu on basal tonus of the vessel and its effects on vessel contraction and relaxation with or without endothelium were observed.
Results: In thoracic arteries under basal tonus, sodium danshensu (0.3-3 g/L) produced a dose-dependent transient contraction. In phenylephrine-precontracted thoracic arteries with or without endothelium, low concentration (0.1-0.3 g/L) of sodium danshensu produced a weak contraction, while high concentrations (1-3 g/L) produced a pronounced vasodilator after a transient vasocontraction. Pre-incubation with sodium danshensu could inhibit vessel contraction induced by phenylephrine and potassium chloride in a concentration-dependent way. Sodium danshensu inhibited phenylephrine- and CaCl2-induced vasoconstriction in Ca^2+-free medium. Pre-incubation with tetraethylammonium, a non-selective K^+ channel blocker, and apamin, a small-conductance calcium-activated K^+ channel blocker partially antagonized the relaxation response induced by sodium danshensu. However, iberiotoxin (big-conductance calcium-sensitive K^+ channel blocker), barium chloride (inward rectifier K^+ channel blocker), and glibencalmide (ATP-sensitive K^+ channel blocker) had no influence on the vasodialtion effect of sodium danshensu.
Conclusion: Sodium danshensu showed a biphasic effects on vessel tension. While low dosage of sodium danshensu produced small contraction possibly through transient enhancement of Ca^2+ influx, high dosage produced significant vasodilation mainly through promoting the opening of non-selective K^+ channels and small-conductance calcium-sensitive K^+ channels in the vascular smooth muscle cells.

Background LCAR-B38M is a chimeric antigen receptor T cell product with two binding domains targeting B cell maturation antigen. Our previous reports showed a remarkable efficacy of LCAR-B38M in ...patients with relapsed/refractory multiple myeloma (RRMM) at a median follow-up of 2 years. Here, we report long-term safety and efficacy data from a median follow-up of 4 years. Methods LEGEND-2 was a phase 1, single-arm, open-label study conducted in four registered sites in China. Seventy-four participants with RRMM received LCAR-B38M treatment. Lymphodepletion was performed using cyclophosphamide or cyclophosphamide plus fludarabine. LCAR-B38M, at a median dose of 0.513 x 10.sup.6 cells/kg, was intravenously administered either in three split infusions or in a single infusion. The primary objective was the safety of LCAR-B38M, and the secondary objective was efficacy. Results As of May 25, 2021, the median follow-up was 47.8 months. All patients experienced greater than or equal to 1 adverse events (AEs). Grade greater than or equal to 3 AEs were observed in 45/74 (60.8%) patients. Cytokine release syndrome (CRS) occurred in 68/74 (91.9%) cases; 7 (9.5%) had grade greater than or equal to 3 CRS. One patient experienced grade 1 central nervous system toxicity. The overall response rate was 87.8%. Fifty-four out of 74 (73.0%) patients achieved complete response. The median progression-free survival was 18.0 months, and the median overall survival for all patients was not reached. The median duration of response was 23.3 months. Four patients experienced viral infection more than 6 months post-infusion, and four patients developed second primary non-hematological malignancies at a median time of 11.5 months post-CAR-T cell transfer. Conclusions The 4-year follow-up data of LCAR-B38M therapy demonstrated a favorable long-term safety profile and a durable response in patients with RRMM. Particles in a storage ring oscillate in the longitudinal and transverse dimensions. Therefore, the signal that beam has generated can be analyzed in the frequency domain to extract many beam ...parameters, such as tune, momentum spread, emittance and their evolution with time and so on. In this paper, the transverse spectrum in HIRFL-CSR was simulated and analyzed under different conditions, including electron cooling, hollow electron beam, solenoid effect, the tune shift caused by power supply ripple and the misalignment between ion and electron beams. The result of the simulation shows that the longitudinal magnetic field of the electron cooling device should be compensated by a "compensation solenoid", and the power supply ripple must be controlled, otherwise the accumulation would be affected and the beam would be lost.
The DNA fragment encoding matureMycobacterium tuberculosis major secretory protein Ag85B was inserted into thePichia pastoris secretory expression vector pHBM905A, under the control of theAOX1 ...promoter. The recombinant plasmid pHBM905A-85B linearized bySal I was introduced intoPichia pastoris strain GS115 by PEG1000 transformation method. After phenotype screening and PCR identification, the resulting GS115-pHBM905A-85B strain was cultivated and induced with methanol. The recombinant Ag85B protein in secreted form was attained with molecular weight of 35×103 approximately detected by SDS-PAGE and Western blot. ELISA experiment proved that the protein had good antigen specificity. Secretory expression of recombinantM. tuberculosis Ag85B inP. pastoris will open a door to mass production of the protein in heterologous host and allow ready evaluation of its immunological function.

Long-distance entanglement distribution is essential for both foundational tests of quantum physics and scalable quantum networks. Owing to channel loss, however, the previously achieved distance was ...limited to ~100 kilometers. Here we demonstrate satellite-based distribution of entangled photon pairs to two locations separated by 1203 kilometers on Earth, through two satellite-to-ground downlinks with a summed length varying from 1600 to 2400 kilometers. We observed a survival of two-photon entanglement and a violation of Bell inequality by 2.37 ± 0.09 under strict Einstein locality conditions. The obtained effective link efficiency is orders of magnitude higher than that of the direct bidirectional transmission of the two photons through telecommunication fibers.

Background and Aims
NAFLD is considered as the hepatic manifestation of the metabolic syndrome, which includes insulin resistance, obesity and hyperlipidemia. NASH is a progressive stage of NAFLD ...with severe hepatic steatosis, hepatocyte death, inflammation, and fibrosis. Currently, no pharmacological interventions specifically tailored for NASH are approved. Ovarian tumor domain, ubiquitin aldehyde binding 1 (OTUB1), the founding member of deubiquitinases, regulates many metabolism‐associated signaling pathways. However, the role of OTUB1 in NASH is unclarified.
Methods and Results
We demonstrated that mice with Otub1 deficiency exhibited aggravated high‐fat diet–induced and high‐fat high‐cholesterol (HFHC) diet–induced hyperinsulinemia and liver steatosis. Notably, hepatocyte‐specific overexpression of Otub1 markedly alleviated HFHC diet–induced hepatic steatosis, inflammatory responses, and liver fibrosis. Mechanistically, we identified apoptosis signal–regulating kinase 1 (ASK1) as a key candidate target of OTUB1 through RNA‐sequencing analysis and immunoblot analysis. Through immunoprecipitation–mass spectrometry analysis, we further found that OTUB1 directly bound to tumor necrosis factor receptor–associated factor 6 (TRAF6) and suppressed its lysine 63–linked polyubiquitination, thus inhibiting the activation of ASK1 and its downstream pathway.
Conclusions
OTUB1 is a key suppressor of NASH that inhibits polyubiquitinations of TRAF6 and attenuated TRAF6‐mediated ASK1 activation. Targeting the OTUB1‐TRAF6‐ASK1 axis may be a promising therapeutic strategy for NASH.

In the past, long-distance free-space quantum communication experiments could only be implemented at night. During the daytime, the bright background sunlight prohibits quantum communication in ...transmission under conditions of high channel loss over long distances. Here, by choosing a working wavelength of 1,550 nm and developing free-space single-mode fibre-coupling technology and ultralow-noise upconversion single-photon detectors, we have overcome the noise due to sunlight and demonstrate free-space quantum key distribution over 53 km during the day. The total channel loss is ∼48 dB, which is greater than the 40 dB channel loss between the satellite and ground and between low-Earth-orbit satellites. Our system thus demonstrates the feasibility of satellite-based quantum communication in daylight. Moreover, given that our working wavelength is located in the optical telecom band, our system is naturally compatible with ground fibre networks and thus represents an essential step towards a satellite-constellation-based global quantum network.
