Seven new formamido-diterpenes, cavernenes A–D (1–4), kalihinenes E and F (5–6), and kalihipyran C (7), together with five known compounds (8–12), were isolated from the South China Sea sponge ...Acanthella cavernosa. Structures were established using IR, HRESIMS, 1D and 2D NMR, and single X-ray diffraction techniques. The isolated compounds were assessed for their cytotoxicity against a small panel of human cancer cell lines (HCT-116, A549, HeLa, QGY-7701, and MDA-MB-231) with IC50 values in the range of 6–18 μM. In addition, compound 9 showed weak antifungal activity against Trichophyton rubrum and Microsporum gypseum with MIC values of 8 and 32 μg/mL, respectively, compound 10 displayed weak antifungal activity against fungi Candida albicans, Cryptococcus neoformans, T. rubrum, and M. gypseum with MIC values of 8, 8, 4, and 8 μg/mL, respectively.
Objective Environmental health risk has become the focus of current global research due to the fact that environmental pollution causes harm to human health. The research on health risk of heavy ...metals in cultivated soil at the provincial level based on interdisciplinary perspectives is of great significance for expanding the understanding of environmental health risk, formulating the control strategies of health risk, and protecting public health. Methods Based on the theories of human-environment relationship, the essential connotation of environmental health risk, and spatial statistical analysis methods in geography, we constructed an analytical framework for health risk of heavy metals in the soil of cultivated land. The characteristic factors, the spatial agglomeration characteristics, and the influencing mechanism of health risk were analyzed. Furthermore, the key points for health risk control were identified. Results The results show that: (1) The health risk levels of heavy metals Cr, Pb, Cd, As, and Hg in the cultivated soil of Jiangsu Province are within acceptable range. But Cr, Pb, and As were the main risk characteristic factors and need to be controlled. (2) Heavy metals in cultivated soil in some counties of Northern Jiangsu Province had a greater impact on the relative health risk level than the rest parts of the province. The degree of influence of different characteristic risk factors for the region was As > Cr > Pb, and the spatial distribution generally presented a pattern of northeast > southwest. (3) As, Cr, Pb, and the comprehensive health risks showed a spatial agglomeration trend, and the spatial structure of agglomeration exhibited a high-high and low-low pattern. (4) The q values of the four detection factors areal average fertilizer application, residents’ education level, road density, and areal average GDP were higher than 0.3, and any two detection factors showed either a nonlinear enhancement or a dual-factor enhancement effect. Conclusion The health risk intensity of heavy metals in cultivated soil puts forward a fresh thought on environment and health researchs, and the research results could provide the practical basis for formulating control strategies on heavy metals in cultivated soil in Jiangsu Province.
AIM: To investigate clarithromycin resistance positions 2142, 2143 and 2144 of the 23 Sr RNA gene in Helicobacter pylori(H. pylori) by nested-allele specific primer-polymerase chain ...reaction(nested-ASP-PCR).METHODS: The gastric tissue and saliva samples from 99 patients with positive results of the rapid urease test(RUT) were collected. The nested-ASP-PCR method was carried out with the external primers and inner allele-specific primers corresponding to the reference strain and clinical strains. Thirty gastric tissue and saliva samples were tested to determine the sensitivity of nested-ASP-PCR and ASP-PCR methods. Then, clarithromycin resistance was detected for 99 clinical samples by using different methods, including nestedASP-PCR, bacterial culture and disk diffusion. RESULTS: The nested-ASP-PCR method was successfully established to test the resistance mutation points 2142, 2143 and 2144 of the 23 SrR NA gene of H. pylori. Among 30 samples of gastric tissue and saliva, the H. pylori detection rate of nested-ASP-PCR was 90% and 83.33%, while the detection rate of ASP-PCR was just 63% and 56.67%. Especially in the saliva samples, nested-ASP-PCR showed much higher sensitivity in H. pylori detection and resistance mutation rates thanASP-PCR. In the 99 RUT-positive gastric tissue and saliva samples, the H. pylori-positive detection rate by nested-ASP-PCR was 87(87.88%) and 67(67.68%), in which there were 30 wild-type and 57 mutated strains in gastric tissue and 22 wild-type and 45 mutated strains in saliva. Genotype analysis showed that three-points mixed mutations were quite common, but different resistant strains were present in gastric mucosa and saliva. Compared to the high sensitivity shown by nested-ASP-PCR, the positive detection of bacterial culture with gastric tissue samples was 50 cases, in which only 26 drug-resistant strains were found through analyzing minimum inhibitory zone of clarithromycin. CONCLUSION: The nested-ASP-PCR assay showed higher detection sensitivity than ASP-PCR and drug sensitivity testing, which could be performed to evaluate clarithromycin resistance of H. pylori.

Alanine aminotransferase (ALT), aspartate aminotransferase (AST), γ-glutamyl- transferase (GGT), and alkaline phosphatase (ALP) are important catabolic enzymes, which play an important role in liver ...function of animals. The activities of these enzymes were monitored in milk and blood plasma and association between them was determined through regression analysis. Samples of milk and blood were collected from 35 housed multiparous Holstein cows 3-9 weeks postpartum. The results showed that there were significantly positive correlations for these four enzymes between the enzyme activities in milk and blood plasma (ALT, rs=0.852, Ρ<0.001; AST, rs=0.341, Ρ<0.001; GGT, rs=0.628, Ρ<0.001; ALP, rs=0.707, Ρ<0.001). Compared to the blood plasma, the activities of ALT (Ρ<0.05) and AST (Ρ<0.001) in milk appeared significantly lower, while the activities of GGT and ALP were significantly higher (Ρ<0.001). With the regression models, it seems that detection of ALT, AST, GGT and ALP activities in milk may be an alternative way to monitor the liver function of cows. Therefore, this study is an initial step for studying the liver function through detection of these enzymes in milk.
