We report the discovery of 28 quasars and 7 luminous galaxies at 5.7 ≤ z ≤ 7.0. This is the tenth in a series of papers from the Subaru High-z Exploration of Low-Luminosity Quasars (SHELLQs) project, ...which exploits the deep multiband imaging data produced by the Hyper Suprime-Cam (HSC) Subaru Strategic Program survey. The total number of spectroscopically identified objects in SHELLQs has now grown to 93 high-z quasars, 31 high-z luminous galaxies, 16 O iii emitters at z ∼ 0.8, and 65 Galactic cool dwarfs (low-mass stars and brown dwarfs). These objects were found over 900 deg2, surveyed by HSC between 2014 March and 2018 January. The full quasar sample includes 18 objects with very strong and narrow Ly emission, whose stacked spectrum is clearly different from that of other quasars or galaxies. While the stacked spectrum shows N v λ1240 emission and resembles that of lower-z narrow-line quasars, the small Ly width may suggest a significant contribution from the host galaxies. Thus, these objects may be composites of quasars and star-forming galaxies.

. Bottom-up synthesis of high-quality graphene is often restricted to ultrasmall amounts if performed by chemical vapour deposition or advanced synthetic organic methods, or it provides a defect-ridden structure if carried out in bulk solution
. Here we show that flash Joule heating of inexpensive carbon sources-such as coal, petroleum coke, biochar, carbon black, discarded food, rubber tyres and mixed plastic waste-can afford gram-scale quantities of graphene in less than one second. The product, named flash graphene (FG) after the process used to produce it, shows turbostratic arrangement (that is, little order) between the stacked graphene layers. FG synthesis uses no furnace and no solvents or reactive gases. Yields depend on the carbon content of the source; when using a high-carbon source, such as carbon black, anthracitic coal or calcined coke, yields can range from 80 to 90 per cent with carbon purity greater than 99 per cent. No purification steps are necessary. Raman spectroscopy analysis shows a low-intensity or absent D band for FG, indicating that FG has among the lowest defect concentrations reported so far for graphene, and confirms the turbostratic stacking of FG, which is clearly distinguished from turbostratic graphite. The disordered orientation of FG layers facilitates its rapid exfoliation upon mixing during composite formation. The electric energy cost for FG synthesis is only about 7.2 kilojoules per gram, which could render FG suitable for use in bulk composites of plastic, metals, plywood, concrete and other building materials.

The "2022 AHA/ACC/HFSA Guideline for the Management of Heart Failure" replaces the "2013 ACCF/AHA Guideline for the Management of Heart Failure" and the "2017 ACC/AHA/HFSA Focused Update of the 2013 ...ACCF/AHA Guideline for the Management of Heart Failure." The 2022 guideline is intended to provide patient-centric recommendations for clinicians to prevent, diagnose, and manage patients with heart failure.
A comprehensive literature search was conducted from May 2020 to December 2020, encompassing studies, reviews, and other evidence conducted on human subjects that were published in English from MEDLINE (PubMed), EMBASE, the Cochrane Collaboration, the Agency for Healthcare Research and Quality, and other relevant databases. Additional relevant clinical trials and research studies, published through September 2021, were also considered. This guideline was harmonized with other American Heart Association/American College of Cardiology guidelines published through December 2021. Structure: Heart failure remains a leading cause of morbidity and mortality globally. The 2022 heart failure guideline provides recommendations based on contemporary evidence for the treatment of these patients. The recommendations present an evidence-based approach to managing patients with heart failure, with the intent to improve quality of care and align with patients' interests. Many recommendations from the earlier heart failure guidelines have been updated with new evidence, and new recommendations have been created when supported by published data. Value statements are provided for certain treatments with high-quality published economic analyses.
We sequenced the genomes of 5,085 severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) strains causing two coronavirus disease 2019 (COVID-19) disease waves in metropolitan Houston, TX, an ...ethnically diverse region with 7 million residents. The genomes were from viruses recovered in the earliest recognized phase of the pandemic in Houston and from viruses recovered in an ongoing massive second wave of infections. The virus was originally introduced into Houston many times independently. Virtually all strains in the second wave have a Gly614 amino acid replacement in the spike protein, a polymorphism that has been linked to increased transmission and infectivity. Patients infected with the Gly614 variant strains had significantly higher virus loads in the nasopharynx on initial diagnosis. We found little evidence of a significant relationship between virus genotype and altered virulence, stressing the linkage between disease severity, underlying medical conditions, and host genetics. Some regions of the spike protein-the primary target of global vaccine efforts-are replete with amino acid replacements, perhaps indicating the action of selection. We exploited the genomic data to generate defined single amino acid replacements in the receptor binding domain of spike protein that, importantly, produced decreased recognition by the neutralizing monoclonal antibody CR3022. Our report represents the first analysis of the molecular architecture of SARS-CoV-2 in two infection waves in a major metropolitan region. The findings will help us to understand the origin, composition, and trajectory of future infection waves and the potential effect of the host immune response and therapeutic maneuvers on SARS-CoV-2 evolution.
There is concern about second and subsequent waves of COVID-19 caused by the SARS-CoV-2 coronavirus occurring in communities globally that had an initial disease wave. Metropolitan Houston, TX, with a population of 7 million, is experiencing a massive second disease wave that began in late May 2020. To understand SARS-CoV-2 molecular population genomic architecture and evolution and the relationship between virus genotypes and patient features, we sequenced the genomes of 5,085 SARS-CoV-2 strains from these two waves. Our report provides the first molecular characterization of SARS-CoV-2 strains causing two distinct COVID-19 disease waves.
We report the discovery of 41 new high-z quasars and luminous galaxies that were spectroscopically identified at 5.7 ≤ z ≤ 6.9. This is the fourth in a series of papers from the Subaru High-z ...Exploration of Low-Luminosity Quasars (SHELLQs) project, based on the deep multi-band imaging data collected by the Hyper Suprime-Cam (HSC) Subaru Strategic Program survey. We selected the photometric candidates using a Bayesian probabilistic algorithm and then carried out follow-up spectroscopy with the Gran Telescopio Canarias and the Subaru Telescope. Combined with the sample presented in the previous papers, we have now spectroscopically identified 137 extremely red HSC sources over about 650 deg2, which includes 64 high-z quasars, 24 high-z luminous galaxies, 6 O iii emitters at z ∼ 0.8, and 43 Galactic cool dwarfs (low-mass stars and brown dwarfs). The new quasars span in luminosity range from M1450 ∼ −26 to −22 mag, and continue to populate luminosities a few magnitudes lower than have been probed by previous wide-field surveys. In a companion paper, we derive the quasar luminosity function at z ∼ 6 over an unprecedentedly wide range of M1450 ∼ −28 to −21 mag, exploiting the SHELLQs and other survey outcomes.
HER2 mutations are infrequent genomic events in biliary tract cancers (BTCs). Neratinib, an irreversible, pan-HER, oral tyrosine kinase inhibitor, interferes with constitutive receptor kinase ...activation and has activity in HER2-mutant tumours. SUMMIT is an open-label, single-arm, multi-cohort, phase 2, 'basket' trial of neratinib in patients with solid tumours harbouring oncogenic HER2 somatic mutations (ClinicalTrials.gov: NCT01953926). The primary objective of the BTC cohort, which is now complete, is first objective response rate (ORR) to neratinib 240 mg orally daily. Secondary objectives include confirmed ORR, clinical benefit rate, progression-free survival, duration of response, overall survival, safety and tolerability. Genomic analyses were exploratory. Among 25 treatment-refractory patients (11 cholangiocarcinoma, 10 gallbladder, 4 ampullary cancers), the ORR is 16% (95% CI 4.5-36.1%). The most common HER2 mutations are S310F (n = 11; 48%) and V777L (n = 4; 17%). Outcomes appear worse for ampullary tumours or those with co-occurring oncogenic TP53 and CDKN2A alterations. Loss of amplified HER2 S310F and acquisition of multiple previously undetected oncogenic co-mutations are identified at progression in one responder. Diarrhoea is the most common adverse event, with any-grade diarrhoea in 14 patients (56%). Although neratinib demonstrates antitumour activity in patients with refractory BTC harbouring HER2 mutations, the primary endpoint was not met and combinations may be explored.
As climate change continues to modify temperature and rainfall patterns, risks from pests and diseases may vary as shifting temperature and moisture conditions affect the life history, activity, and ...distribution of invertebrates and diseases. The potential consequences of changing climate on pest management strategies must be understood for control measures to adapt to new environmental conditions. The redlegged earth mite (RLEM; Halotydeus destructor Tucker) is a major economic pest that attacks pastures and grain crops across southern Australia and is typically controlled by pesticides. TIMERITE® is a management strategy that relies on estimating the optimal timing (the TIMERITE® date) for effective chemical control of RLEM populations in spring. In this study, we assessed the efficacy of control at the TIMERITE® date from 1990 to 2020 across southern Australia using a simulation approach that incorporates historical climatic data and field experimental data on life history, seasonal abundance, and population level pesticide responses. We demonstrate that moisture and temperature conditions affect the life history of RLEM and that changes in the past three decades have gradually diminished the efficacy of the TIMERITE® strategy. Furthermore, we show that by incorporating improved climatic data into predictions and shifting the timing of control to earlier in the year, control outcomes can be improved and are more stable across changing climates. This research emphasises the importance of accounting for dynamic environmental responses when developing and implementing pest management strategies to ensure their long-term effectiveness. Suggested modifications to estimating the TIMERITE® date will help farmers maintain RLEM control outcomes amidst increasingly variable climatic conditions.

Human cancer is caused by the accumulation of mutations in oncogenes and tumor suppressor genes. To catalog the genetic changes that occur during tumorigenesis, we isolated DNA from 11 breast and 11 ...colorectal tumors and determined the sequences of the genes in the Reference Sequence database in these samples. Based on analysis of exons representing 20,857 transcripts from 18,191 genes, we conclude that the genomic landscapes of breast and colorectal cancers are composed of a handful of commonly mutated gene "mountains" and a much larger number of gene "hills" that are mutated at low frequency. We describe statistical and bioinformatic tools that may help identify mutations with a role in tumorigenesis. These results have implications for understanding the nature and heterogeneity of human cancers and for using personal genomics for tumor diagnosis and therapy.
