The Tsuruoka Metabolomics Cohort Study (TMCS) is an ongoing population-based cohort study being conducted in the rural area of Yamagata Prefecture, Japan. This study aimed to enhance the precision ...prevention of multi-factorial, complex diseases, including non-communicable and aging-associated diseases, by improving risk stratification and prediction measures. At baseline, 11,002 participants aged 35–74 years were recruited in Tsuruoka City, Yamagata Prefecture, Japan, between 2012 and 2015, with an ongoing follow-up survey. Participants underwent various measurements, examinations, tests, and questionnaires on their health, lifestyle, and social factors. This study used an integrative approach with deep molecular profiling to identify potential biomarkers linked to phenotypes that underpin disease pathophysiology and provide better mechanistic insights into social health determinants. The TMCS incorporates multi-omics data, including genetic and metabolomic analyses of 10,933 participants and comprehensive data collection ranging from physical, psychological, behavioral, and social to biological data. The metabolome is used as a phenotypic probe because it is sensitive to changes in physiological and external conditions. The TMCS focuses on collecting outcomes for cardiovascular disease, cancer incidence and mortality, disability, functional decline due to aging and disease sequelae, and the variation in health status within the body represented by omics analysis that lies between exposure and disease. It contains several sub-studies on aging, heated tobacco products, and women's health. This study is notable for its robust design, high participation rate (89%), and long-term repeated surveys. Moreover, it contributes to precision prevention in Japan and East Asia as a well-established multi-omics platform.

We attempted to reproduce the total electron content (TEC) variation in the Earth's atmosphere from the temporal variation of the solar flare spectrum of the X9.3 flare on September 6, 2017. The ...flare spectrum from the Flare Irradiance Spectral Model (FISM), and the flare spectrum from the 1D hydrodynamic model, which considers the physics of plasma in the flare loop, are used in the GAIA model, which is a simulation model of the Earth's whole atmosphere and ionosphere, to calculate the TEC difference. We then compared these results with the observed TEC. When we used the FISM flare spectrum, the difference in TEC from the background was in a good agreement with the observation. However, when the flare spectrum of the 1D-hydrodynamic model was used, the result varied depending on the presence or absence of the background. This difference depending on the models is considered to represent which extreme ultraviolet (EUV) radiation is primarily responsible for increasing TEC. From the flare spectrum obtained from these models and the calculation result of TEC fluctuation using GAIA, it is considered that the enhancement in EUV emission by approximately 15

35 nm mainly contributes in increasing TEC rather than that of X-ray emission, which is thought to be mainly responsible for sudden ionospheric disturbance. The X-rays and extreme ultraviolet (EUV) emitted during solar flares can rapidly change the physical composition of Earth’s ionosphere, causing space weather phenomena. It is important to develop an ...accurate understanding of solar flare emission spectra to understand how it affects the ionosphere. We reproduced the entire solar flare emission spectrum using an empirical model and physics-based model, and input it into the Earth’s atmospheric model, GAIA to calculate the total electron content (TEC) enhancement due to solar flare emission. We compared the statistics of nine solar flare events and calculated the TEC enhancements with the corresponding observed data. The model used in this study was able to estimate the TEC enhancement due to solar flare emission with a correlation coefficient greater than 0.9. The results of this study indicate that the TEC enhancement due to solar flare emission is determined by soft X-ray and EUV emission with wavelengths shorter than 35 nm. The TEC enhancement is found to be largely due to the change in the soft X-ray emission and EUV line emissions with wavelengths, such as Fe XVII 10.08 nm, Fe XIX 10.85 nm and He II 30.38 nm.

The Kidney Disease Improving Global Outcomes guidelines recommend nephrology referral for patients with chronic kidney disease (CKD) stages 4 to 5, significant proteinuria and persistent microscopic ...haematuria. However, the recommendations are opinion-based and which patients with CKD benefit more from nephrology referral has not been elucidated.
In this retrospective cohort study, patients referred to our nephrology outpatient clinic from April 2017 to March 2019 were included. We excluded patients considered to have an acute decline in kidney function (annual decline in estimated glomerular filtration rate eGFR >10 mL/min/1.73 m
). The slopes of eGFR before and after nephrology referral were estimated and compared by linear mixed effects models. Interaction between time and referral status (before or after referral) was assessed and effect modifications by the presence of diabetes, proteinuria (defined by urine dipstick protein 2+ or more), urine occult blood, hypoalbuminemia (defined by albumin levels less than 3.5 g/dL) and anaemia (defined by haemoglobin levels less than 11.0 g/dL) were evaluated.
The eGFR slope significantly improved from -2.05 (-2.39 to -1.72) to -0.96 (-1.36 to -0.56) mL/min/1.73 m
/year after nephrology referral (p < .001). The improvement in eGFR slope was more prominent among those with diabetes mellitus, anaemia, and hypoalbuminemia (all p-values for three-way interaction <.001 after adjustment for covariates). Further adjustments for time-dependent haemoglobin levels, the use of erythropoiesis-stimulating agents, iron supplementation, anti-hypertensives and anti-diabetic medications did not change the significance of the interactions.
Nephrology referral slows CKD progression, especially among those with hypoalbuminemia, diabetes or anaemia. Patients with hypoalbuminemia, diabetes or anaemia might benefit more from specialized care and lifestyle modifications by nephrologists. The inclusion of anaemia and hypoalbuminemia in nephrology referral criteria should be considered.

We have examined the quality of the National Research Institute for Earth Science and Disaster Prevention (NIED) seismic moment tensor (MT) catalogue obtained using a regional broadband seismic ...network (FREESIA). First, we examined using synthetic waveforms the robustness of the solutions with regard to data noise as well as to errors in the velocity structure and focal location. Then, to estimate the reliability, robustness and validity of the catalogue, we compared it with the Harvard centroid moment tensor (CMT) catalogue as well as the Japan Meteorological Agency (JMA) focal mechanism catalogue. We found out that the NIED catalogue is consistent with Harvard and JMA catalogues within the uncertainty of 0.1 in moment magnitude, 10 km in depth, and 15° in direction of the stress axes. The NIED MT catalogue succeeded in reducing to 3.5 the lower limit of moment magnitude above which the moment tensor could be reliably estimated. Finally, we estimated the stress tensors in several different regions by using the NIED MT catalogue. This enables us to elucidate the stress/deformation field in and around the Japanese islands to understand the mode of deformation and applied stress. Moreover, we identified a region of abnormal stress in a swarm area from stress tensor estimates.

A 27-year-old nulligravida woman without a history of dermatosis was hospitalized for threatened preterm labor at 29 weeks' gestation; therefore, continuous infusion of ritodrine hydrochloride was ...started. At 31 weeks' gestation, erythematous plaques appeared and spread over the body surface; therefore, a topical steroid preparation was applied. At 32 weeks' gestation, the eruptions developed into irregular annular areas of erythema with multiple pustules accompanied by severe itching, and oral prednisolone treatment was started. Bacterial cultures of the pustules were negative, and a crural cutaneous biopsy revealed Kogoj's spongiform pustules. Based on the clinicopathological findings, the most likely diagnosis was impetigo herpetiformis, which causes cutaneous symptoms closely resembling pustular psoriasis in pregnant females without a history of psoriasis. To rule out ritodrine-induced pustular eruptions, the ritodrine infusion was stopped and treatment with an MgSO4 preparation was started at 33 weeks' 3 days' gestation; however, the uterine contractions could not be suppressed. Because of the patient's highly edematous, severely painful feet, a cesarean section was performed the same day. Within several days of delivery, the eruptions began to resolve, and no recurrence was observed after treatment with oral prednisolone was stopped 31 days after delivery. On the basis of a positive patch test for ritodrine, we diagnosed pustular drug eruptions caused by ritodrine hydrochloride. Although ritodrine-induced pathognomonic cutaneous eruptions are rare, we would like to emphasize that ritodrine can cause drug-induced pustular eruptions distinctly resembling life-threatening impetigo herpetiformis.

We focus on the fact that human body kinematic structure leads to the linearity between a binocular visual space and a joint space, and propose a novel criterion for evaluating the appropriateness of ...endoscopic images. Then, we aim to establish a new autonomous endoscope positioning method based on that criterion. In this study, we first assumed that the stereo endoscope robot was regarded as virtual binocular eyes of the surgeon and the surgical instruments were regarded as virtual arms connecting the surgeon's real arms. We then made a hypothesis that the surgeon or the camera assistant operated the endoscope such that the binocular visual space coordinates of the tip of the surgical instrument and the joint angles of the surgeon's arm had a linear relationship.

SKD1 belongs to the AAA-ATPase family and is one of the mammalian class E Vps (vacuolar protein sorting) proteins. Previously we have reported that the overexpression of an ATPase activity-deficient ...form of SKD1 (suppressor of potassium transport growth defect), SKD1(E235Q), leads the perturbation of membrane transport through endosomes and lysosomes, however, the molecular mechanism behind the action of SKD1 is poorly understood. We have identified two SKD1-binding proteins, SBP1 and mVps2, by yeast two-hybrid screening and we assign them as mammalian class E Vps proteins. The primary sequence of SBP1 indicates 22.5% identity with that of Vta1p from Saccharomyces cerevisiae, which was recently identified as a novel class E Vps protein binding to Vps4p. In fact, SBP1 binds directly to SKD1 through its C-terminal region (198-309). Endogenous SBP1 is exclusively localized to cytosol, however it is redirected to an aberrant endosomal structure, the E235Q compartment, in the cells expressing SKD1(E235Q). The ATPase activity of SKD1 regulates both the membrane association of, and assembly of, a large hetero-oligomer protein complex, containing SBP1, which is potentially involved in membrane transport through endosomes and lysosomes. The N-terminal half (1-157) of human SBP1 is identical to lyst-interacting protein 5 and intriguingly, SKD1 ATPase activity significantly influences the membrane association of lyst protein. The SKD1-SBP1 complex, together with lyst protein, may function in endosomal membrane transport. A primary sequence of mVps2, a mouse homologue of human CHMP2A/BC-2, indicates 44.4% identity with Vps2p/Did4p/Chm2p from Saccharomyces cerevisiae. mVps2 also interacts with SKD1 and is localized to the E235Q compartment. Intriguingly, the N-terminal coiled-coil region of mVps2 is required for the formation of the E235Q compartment but not for binding to SKD1. We propose that both SBP1 and mVps2 regulate SKD1 function in mammalian cells.
