Amyloid-induced aggregation and precipitation of soluble proteins were investigated in vitro using the amyloid fibrils of the β(25−35) peptide, a cytotoxic fragment of the Alzheimer's β-peptide at ...positions 25−35. The aggregation rate of firefly luciferase was found to be modulated by both a chaperone molecule DnaK and the β(25−35) amyloid, but their effects were opposite in direction. The amyloid fibril drastically facilitated the luciferase aggregation, which may define a kind of anti-chaperone activity. The effect of the β(25−35) amyloid to promote protein aggregation and precipitation was further demonstrated for a wide variety of target proteins. The amount of coprecipitation was well correlated with the predicted isoelectric point of the target proteins, indicating that the interaction between the β(25−35) amyloid and the target was driven by an electrostatic force between them. This view was confirmed by the experiments using an electrically neutral mutant peptide, β(25−35)KA. It was also found that clustering of the β(25−35) peptide to form amyloid and the conformation of the target protein are additional factors that determine the strength of the amyloid−protein interaction. Spectroscopic and electron microscopic methods have revealed that the proteins coprecipitated with the β(25−35) amyloid formed amorphous aggregates deposited together with the amyloid fibrils. The conformation of protein molecules left in the residual soluble fraction was also damaged in the amyloid-containing solution. As a summary, this study has proposed a scheme for events related to the nonspecific amyloid−protein interaction, which may play substantial roles in in vivo conditions.

We present a method using high-angle annular dark field scanning transmission electron microscopy (HAADF-STEM) to determine the chemical composition of bi-metallic nanoparticles. This method, which ...can be applied in a semi-automated way, allows large scale analysis with a statistical number of particles (several hundreds) in a short time. Once a calibration curve has been obtained, e.g., using energy-dispersive X-ray spectroscopy (EDX) measurements on a few particles, the HAADF integrated intensity of each particle can indeed be directly related to its chemical composition. After a theoretical description, this approach is applied to the case of iron–palladium nanoparticles (expected to be nearly stoichiometric) with a mean size of 8.3 nm. It will be shown that an accurate chemical composition histogram is obtained, i.e., the Fe content has been determined to be 49.0 at.% with a dispersion of 10.4 %. HAADF-STEM analysis represents a powerful alternative to fastidious single particle EDX measurements, for the compositional dispersion in alloy nanoparticles.

The JSNS2 (J-PARC Sterile Neutrino Search at J-PARC Spallation Neutron Source) experiment aims to search for neutrino oscillations over a 24 m short baseline at J-PARC . The JSNS2 inner detector is ...filled with 17 tons of gadolinium-loaded liquid scintillator (LS) and both the intermediate γ-catcher and the optically separated outer veto are filled with un-loaded LS . Optical photons from scintillation are observed by 120 Photomultiplier Tubes (PMTs). A total of 130 PMTs for the JSNS2 experiment were either donated by other experiments or purchased from Hamamatsu. Donated PMTs were purchased around 10 years ago, therefore JSNS2 did pre-calibration of the PMTs including the purchased PMTs. 123 PMTs demonstrated acceptable performance for the JSNS2 experiment, and 120 PMTs were installed in the detector.

from the Hokuriku-Plus AF Registry. The primary outcome was incident WKF, defined as the %eGFR change from the baseline ≥ 30% during the follow-up period. We evaluated the association between baseline variables and incident WKF using univariate and multivariate Cox proportional hazard models. We also evaluated the non-linear association between the identified factors and incident WKF. During a median follow-up period of 3.0 years (interquartile range: 2.7–3.3), incident WKF was observed in 108 patients (32.6 per 1000 person-years). Compared to the patients without incident WKF, the patients with incident WKF were older and had a higher prevalence of heart failure (HF), diabetes mellitus (DM), and vascular disease at baseline. Those who experienced incident WKF also had higher diastolic blood pressure, lower hemoglobin, lower eGFR, higher B-type natriuretic peptide (BNP) and used warfarin more frequently. Upon multivariate analysis, age ≥ 75 years, HF, DM, and anemia were independently associated with incident WKF. Additionally, age and hemoglobin were linearly associated with the risk of incident WKF, whereas a J- or U-shaped association was observed for HbA1c and BNP. Age ≥ 75 years, HF, DM, and anemia were associated with the development of WKF in Japanese patients with NVAF. In patients with these risk factors, a careful monitoring of the kidney function and appropriate interventions may be important when possible.

Electromagnetic sensor to detect the unbalanced flow in CC strand by using the magnetic transformation just below CC mold has been proposed, noticing that the narrow face of slab has been cooled at ...beneath of mold and surface temperature rapidly decreases to Curie temperature (Tc). Laboratory experiments have been performed to verify the principle of the proposed sensor, which is composed of primary and secondary solenoid coil. The obtained experimental results showed that the relationship between the surface temperature of slab and induced electric voltage could be explained by Curie-Weiss law, when AC magnetic field has been imposed on the slab surface. Moreover, plant tests have been conducted by installing the sensor just below the narrow face of mold. It can be found that the proposed sensor have potential to measure surface temperature under severe conditions and detect the unbalanced flow in CC strand.
We developed an on-line experimental system for conducting hybrid experiments in real time. It combines a computer, which conducts vibration simulation and generates a control signal, and a hydraulic ...actuator, which conducts a vibration experiment driven by the control signal. This system compensates for actuator delay and thus enables experiments to be carried out in real time. We evaluated the stability of the experiments with respect to the mass of the structure under excitation, and we developed a new method for compensating actuator delay in order to increase the stability condition. In this method, the compensated control signal is generated from the simulation results by using not only displacement but also velocity and acceleration. This method provides a stability criterion (allowable ratio of mass of the structure under excitation to that of a numerical model) about three times larger than that from the current method.
