Despite the availabilty of imaging-based and mass-spectrometry-based methods for spatial proteomics, a key challenge remains connecting images with single-cell-resolution protein abundance ...measurements. Here, we introduce Deep Visual Proteomics (DVP), which combines artificial-intelligence-driven image analysis of cellular phenotypes with automated single-cell or single-nucleus laser microdissection and ultra-high-sensitivity mass spectrometry. DVP links protein abundance to complex cellular or subcellular phenotypes while preserving spatial context. By individually excising nuclei from cell culture, we classified distinct cell states with proteomic profiles defined by known and uncharacterized proteins. In an archived primary melanoma tissue, DVP identified spatially resolved proteome changes as normal melanocytes transition to fully invasive melanoma, revealing pathways that change in a spatial manner as cancer progresses, such as mRNA splicing dysregulation in metastatic vertical growth that coincides with reduced interferon signaling and antigen presentation. The ability of DVP to retain precise spatial proteomic information in the tissue context has implications for the molecular profiling of clinical samples.

Uremic cardiomyopathy is characterized by diastolic dysfunction (DD), left ventricular hypertrophy (LVH), and fibrosis. Angiotensin-II plays a major role in the development of uremic cardiomyopathy ...via nitro-oxidative and inflammatory mechanisms. In heart failure, the beta-3 adrenergic receptor (β3-AR) is up-regulated and coupled to endothelial nitric oxide synthase (eNOS)-mediated pathways, exerting antiremodeling effects. We aimed to compare the antiremodeling effects of the angiotensin-II receptor blocker losartan and the β3-AR agonist mirabegron in uremic cardiomyopathy. Chronic kidney disease (CKD) was induced by 5/6th nephrectomy in male Wistar rats. Five weeks later, rats were randomized into four groups: (1) sham-operated, (2) CKD, (3) losartan-treated (10 mg/kg/day) CKD, and (4) mirabegron-treated (10 mg/kg/day) CKD groups. At week 13, echocardiographic, histologic, laboratory, qRT-PCR, and Western blot measurements proved the development of uremic cardiomyopathy with DD, LVH, fibrosis, inflammation, and reduced eNOS levels, which were significantly ameliorated by losartan. However, mirabegron showed a tendency to decrease DD and fibrosis; but eNOS expression remained reduced. In uremic cardiomyopathy, β3-AR, sarcoplasmic reticulum ATPase (SERCA), and phospholamban levels did not change irrespective of treatments. Mirabegron reduced the angiotensin-II receptor 1 expression in uremic cardiomyopathy that might explain its mild antiremodeling effects despite the unchanged expression of the β3-AR.

Radiation-induced heart disease (RIHD) is a potential late side-effect of thoracic radiotherapy resulting in left ventricular hypertrophy (LVH) and fibrosis due to a complex pathomechanism leading to ...heart failure. Angiotensin-II receptor blockers (ARBs), including losartan, are frequently used to control heart failure of various etiologies. Preclinical evidence is lacking on the anti-remodeling effects of ARBs in RIHD, while the results of clinical studies are controversial. We aimed at investigating the effects of losartan in a rat model of RIHD. Male Sprague-Dawley rats were studied in three groups: (1) control, (2) radiotherapy (RT) only, (3) RT treated with losartan (per os 10 mg/kg/day), and were followed for 1, 3, or 15 weeks. At 15 weeks post-irradiation, losartan alleviated the echocardiographic and histological signs of LVH and fibrosis and reduced the overexpression of chymase, connective tissue growth factor, and transforming growth factor-beta in the myocardium measured by qPCR; likewise, the level of the SMAD2/3 protein determined by Western blot decreased. In both RT groups, the pro-survival phospho-AKT/AKT and the phospho-ERK1,2/ERK1,2 ratios were increased at week 15. The antiremodeling effects of losartan seem to be associated with the repression of chymase and several elements of the TGF-β/SMAD signaling pathway in our RIHD model.

In this paper, several spectral indices based on spectral reflectance data from the Terra satellite’s Moderate Resolution Imaging Spectro-radiometer (MODIS) sensor were evaluated in terms of their ...practical applicability for quantifying drought intensity and its geographical effects to support the drought monitoring applications. A total number of 358 MODIS 8-day composite images for the period between 2000 and 2014 were acquired and processed for the analysis; the frequency of drought phenomenon was increased during this time period. Vegetation indices, water indices, and combinations of both, which were called drought indices, were used. To validate the results, regression analyses were performed and Pearson’s r values were calculated. The Pálfai Drought Index (PAI) and the average annual yields of different crops were used as reference data. The Normalized Difference Vegetation Index (NDVI), the Difference Vegetation Index (DVI), the Normalized Difference Water Index (NDWI), the Difference Water Index (DWI), the Difference Drought Index (DDI), and the Normalized Difference Drought Index (NDDI) were found to be significant in quantifying drought intensity. Anomalies relative to the average of the period between 2000 and 2014 were calculated and standardized using the standard deviation, enabling the identification of above-average, drought-stricken areas. The results of this study can be used to create a cost-effective, near real-time and currently missing national drought monitoring system with high temporal resolution to detect regional climate changes, and to assess forest damage probability through changes in the chlorophyll and water content of forest vegetation.

Saline wetlands experience large temporal fluctuations in water supply during the year and are recharged only or mainly through precipitation, meaning they are vulnerable to climate-change-induced ...aridification. Most passive satellite sensors are unsuitable for continuous wetland monitoring due to cloud cover and their relatively low temporal resolution. However, active satellite sensors such as the C-band synthetic aperture radar of Sentinel-1 satellites offer free, cloud-independent data. We examined surface water cover changes from October 2014 to November 2018 in the strictly protected area (13,000 ha) of the Upper-Kiskunság Alkaline Lakes region in the Danube–Tisza Interfluve in Hungary, with the aim of helping with nature protection planning. Changes and sensitivity can be defined based on the knowledge of variability. We developed a method for water cover detection based on automatic classification, applying the so-called WEKA K-Means clustering algorithm. For satellite data processing and analysis, we used the Google Earth Engine cloud processing platform. In terms of validation, we compared our results with the multispectral Modified Normalized Difference Water Index (MNDWI) derived from Landsat 8 and Sentinel-2 top-of-atmosphere reflectance images using a threshold-based binary classifier (receiver operator characteristics) for the MNDWI data. Using two completely distinct methods operating in distinct wavelength ranges, we obtained adequately matching results, with Spearman’s correlation coefficients (ρ) ranging from 0.54 to 0.80.

Steroids are often considered valuable molecular tools for the development of anticancer agents with improved pharmacological properties. Conjugation of metal chelating moieties with a lipophilic ...sterane backbone is a viable option to obtain novel anticancer compounds. In this work, two estradiol-based hybrid molecules (PMA-E2 and DMA-E2) with an (N,N,O) binding motif and their Cu(II) complexes were developed. The lipophilicity, solubility, and acid-base properties of the novel ligands were determined by the combined use of UV-visible spectrophotometry, pH-potentiometry, and 1H NMR spectroscopy. The solution speciation and redox activity of the Cu(II) complexes were also investigated by means of UV-visible and electron paramagnetic resonance spectroscopy. Two structurally analogous ligands (PMAP and DMAP) were also included in the studies for better interpretation of the solution chemical data obtained. Three pKa values were determined for all ligands, revealing the order of the deprotonation steps: pyridinium-NH+ or NH(CH3)2+, secondary NH2+, and OH. The dimethylamine derivatives (DMA-E2, DMAP) are found in their H2L+ forms in solution at pH 7.4, whereas the fraction of the neutral HL species is significant (34–37%) in the case of the pyridine nitrogen-containing derivatives (PMA-E2, PMAP). Both estradiol derivatives were moderately cytotoxic in human breast (MCF-7) and colon adenocarcinoma (Colo-205) cells (IC50 = 30–63 μM). They form highly stable complexes with Cu(II) ions capable of oxidizing ascorbate and glutathione. These Cu(II) complexes are somewhat more cytotoxic (IC50 = 15–45 μM) than their corresponding ligands and show a better selectivity profile.
