The presence of mercury (Hg), particularly methylmercury (MeHg), is a concern for both human and ecological health as MeHg is a neurotoxin and can bioaccumulate to lethal levels in upper trophic ...level organisms. Recent research has demonstrated that coagulation with metal-based salts can effectively remove both inorganic mercury (IHg) and MeHg from solution through association with dissolved organic matter (DOM) and subsequent flocculation and precipitation. In this study, we sought to further examine interactions between Hg and DOM and the resulting organo-metallic precipitate (floc) to assess if (1) newly added IHg could be removed to the same extent as ambient IHg or whether the association between IHg and DOM requires time, and (2) once formed, if the floc has the capacity to remove additional Hg from solution. Agricultural drainage water samples containing ambient concentrations of both DOM and IHg were spiked with a traceable amount of isotopically enriched IHg and dosed with ferric sulfate after 0, 1, 5, and 30 days. Both ambient and newly added IHg were removed within hours, with 69–79 % removed. To a separate sample set, isotopically enriched IHg was added to solution after floc had formed. Under those conditions, 81–95 % of newly added Hg was removed even at Hg concentrations 1000-fold higher than ambient levels. Results of this study indicate coagulation with ferric sulfate effectively removes both ambient and newly added IHg entering a system and suggests rapid association between IHg and DOM. This work also provides new information regarding the ability of floc to remove additional Hg from solution even after it has formed.
Although oxygen supplementation is beneficial to support life in the clinic, excessive oxygen therapy also has been linked to damage to organs such as the lung or the eye. However, there is a lack of ...understanding of whether high oxygen therapy directly affects the kidney, leading to acute kidney injury, and what molecular mechanisms may be involved in this process. In this review, we revise our current understanding of the mechanisms by which hyperoxia leads to organ damage and highlight possible areas of investigation for the scientific community interested in novel mechanisms of kidney disease. Overall, we found a significant need for both animal and clinical studies evaluating the role of hyperoxia in inducing kidney damage. Thus, we urge the research community to further investigate oxygen therapy and its impact on kidney health with the goal of optimizing oxygen therapy guidelines and improving patient care.

The use of deep learning to auto-contour organs at risk (OARs) in gynecologic radiation treatment is well established. Yet, there is limited data investigating the prospective use of auto-contouring ...in clinical practice. In this study, we assess the accuracy and efficiency of auto-contouring OARs for computed tomography–based brachytherapy treatment planning of gynecologic malignancies.
An inhouse contouring tool automatically delineated 5 OARs in gynecologic radiation treatment planning: the bladder, small bowel, sigmoid, rectum, and urethra. Accuracy of each auto-contour was evaluated using a 5-point Likert scale: a score of 5 indicated the contour could be used without edits, while a score of 1 indicated the contour was unusable. During scoring, automated contours were edited and subsequently used for treatment planning. Dice similarity coefficient, mean surface distance, 95% Hausdorff distance, Hausdorff distance, and dosimetric changes between original and edited contours were calculated. Contour approval time and total planning time of a prospective auto-contoured (AC) cohort were compared with times from a retrospective manually contoured (MC) cohort.
Thirty AC cases from January 2022 to July 2022 and 31 MC cases from July 2021 to January 2022 were included. The mean (±SD) Likert score for each OAR was the following: bladder 4.77 (±0.58), small bowel 3.96 (±0.91), sigmoid colon 3.92 (±0.81), rectum 4.6 (±0.71), and urethra 4.27 (±0.78). No ACs required major edits. All OARs had a mean Dice similarity coefficient > 0.86, mean surface distance < 0.48 mm, 95% Hausdorff distance < 3.2 mm, and Hausdorff distance < 10.32 mm between original and edited contours. There was no significant difference in dose-volume histogram metrics (D2.0 cc/D0.1 cc) between original and edited contours (P values > .05). The average time to plan approval in the AC cohort was 19% less than the MC cohort. (AC vs MC, 117.0 + 18.0 minutes vs 144.9 ± 64.5 minutes, P = .045).
Automated contouring is useful and accurate in clinical practice. Auto-contouring OARs streamlines radiation treatment workflows and decreases time required to design and approve gynecologic brachytherapy plans.

Stable isotopes present in water (δ2H, δ18O) have been used extensively to evaluate hydrological processes on the basis of parameters such as evaporation, precipitation, mixing, and residence time. ...In estuarine aquatic habitats, residence time (τ) is a major driver of biogeochemical processes, affecting trophic subsidies and conditions in fish-spawning habitats. But τ is highly variable in estuaries, owing to constant changes in river inflows, tides, wind, and water height, all of which combine to affect τ in unpredictable ways. It recently became feasible to measure δ2H and δ18O continuously, at a high sampling frequency (1 Hz), using diffusion sample introduction into a cavity ring-down spectrometer. To better understand the relationship of τ to biogeochemical processes in a dynamic estuarine system, we continuously measured δ2H and δ18O, nitrate and water quality parameters, on board a small, high-speed boat (5 to >10 m s–1) fitted with a hull-mounted underwater intake. We then calculated τ as is classically done using the isotopic signals of evaporation. The result was high-resolution (∼10 m) maps of residence time, nitrate, and other parameters that showed strong spatial gradients corresponding to geomorphic attributes of the different channels in the area. The mean measured value of τ was 30.5 d, with a range of 0–50 d. We used the measured spatial gradients in both τ and nitrate to calculate whole-ecosystem uptake rates, and the values ranged from 0.006 to 0.039 d–1. The capability to measure residence time over single tidal cycles in estuaries will be useful for evaluating and further understanding drivers of phytoplankton abundance, resolving differences attributable to mixing and water sources, explicitly calculating biogeochemical rates, and exploring the complex linkages among time-dependent biogeochemical processes in hydrodynamically complex environments such as estuaries.

Introduction/Purpose:
Midfoot osteoarthritis affects up to 12% of the adult population greater than 50 years old. Arthrodesis is the traditional surgical procedure ...performed when nonoperative management fails but is a technically demanding procedure that requires prolonged postoperative immobilization and is not uncommonly complicated by nonunion or deep infection. The deep peroneal nerve (DPN) plays a significant role in afferent nociceptive midfoot joint pain perception, most consistently in the second and third tarsometatarsal joints and the naviculocuneiform joint. Early outcomes following DPN neurectomy have demonstrated promising results as a safe and effective treatment that allows early mobilization and weight bearing. The purpose of our study was to evaluate the mid-term patient satisfaction and pain relief after DPN neurectomy for treatment of chronic midfoot pain due to osteoarthritis.
Methods:
In this retrospective IRB-approved questionnaire-based study, we evaluated 49 out of 95 patients identified (56 feet) with an average follow-up of 34.6 (range, 9 to 51) months, who underwent DPN neurectomy (Figure 1) at our institution between September 2017 and February 2021. There were 39 women and 10 men, 42 unilateral (23 right, 19 left) and 7 bilateral procedures, with an average age of 67.9 (range, 35 to 88) years at the time of surgery. A questionnaire which included questions regarding post-surgical dorsal midfoot pain relief, surgical result satisfaction, and current functional limitations was administered via telephone. Demographic information, patient responses, and complications were recorded.
Results:
There was an 81.2% patient satisfaction with the result of the surgery in relieving their dorsal midfoot pain. Of the 49 patients, 84.9% would repeat the surgery under the same circumstances, 84.1% would recommend the surgery to a friend, 10.2% reported they wish they had undergone arthrodesis, 8.2% reported no pain relief in the first 6 months, and 55.8% reported current activity limitations because of dorsal midfoot pain. Six feet (10.7%) underwent a second procedure with an average postoperative time of 20.5 (range, 1 to 36) months. Complications include one patient with deep wound infection, one DPN neurectomy revision and burial into bone, one DPN neuroma excision, one midfoot exostosis excision, and two conversions to arthrodesis. Theoretic complications of neuroarthropathy, midfoot collapse, and development of lesser toe deformities were not observed.
Conclusion:
DPN neurectomy is a safe and effective surgical option for the management of chronic dorsal midfoot pain due to midfoot osteoarthritis after failed nonoperative management. At midterm follow-up, patients who underwent dorsal midfoot denervation reported acceptable satisfaction rates, similar to those reported for arthrodesis, with associated improved functional outcomes and few complications.
The incidence of prosthetic hip dislocation continues to increase because of the overall increase in volume of total hip replacement surgery. Closed reduction is often the preferred treatment, ...particularly in the first few months after surgery. No matter the closed reduction technique, linear traction is a requirement, thus posing a physically demanding stress opening both surgeon and patient to potential injury. We describe a fracture table closed reduction technique along with outcomes and safety data for a sample of patients. In all 10 reduction procedures, reduction was achieved quickly and without fracture or anesthetic complication. The use of a fracture table for reduction of prosthetic hip dislocation is a viable option, particularly when the surgeon may not have the physical requirements and/or qualified assistance necessary for reduction in the emergency department.

Inbreeding depression may be common in nature, reflecting either the failure of inbreeding avoidance strategies or inbreeding tolerance when avoidance is costly. The combined assessment of inbreeding ...risk, avoidance and depression is therefore fundamental to evaluate the inbreeding strategy of a population, that is how individuals respond to the risk of inbreeding. Here, we use the demographic and genetic monitoring of 10 generations of wild grey mouse lemurs (Microcebus murinus), small primates from Madagascar with overlapping generations, to examine their inbreeding strategy. Grey mouse lemurs have retained ancestral mammalian traits, including solitary lifestyle, polygynandry and male‐biased dispersal, and may therefore offer a representative example of the inbreeding strategy of solitary mammals. The occurrence of close kin among candidate mates was frequent in young females (~37%, most often the father) and uncommon in young males (~6%) due to male‐biased dispersal. However, close kin consistently represented a tiny fraction of candidate mates (< 1%) across age and sex categories. Mating biases favouring partners with intermediate relatedness were detectable in yearling females and adult males, possibly partly caused by avoidance of daughter–father matings. Finally, inbreeding depression, assessed as the effect of heterozygosity on survival, was undetectable using a capture–mark–recapture study. Overall, these results indicate that sex‐biased dispersal is a primary inbreeding avoidance mechanism at the population level, and mating biases represent an additional strategy that may mitigate residual inbreeding costs at the individual level. Combined, these mechanisms explain the rarity of inbreeding and the lack of detectable inbreeding depression in this large, genetically diverse population.

JUMONJI (JMJ) is a nuclear factor that is critical for normal cardiovascular development, evidenced by the analysis of jmj homozygous mutant mice. However, the molecular function of JMJ remains to be ...elucidated. In the present study, we investigated whether JMJ is a transcriptional modulator. Reporter gene assays using the GAL4-DNA binding domain fused to JMJ and a reporter gene consisting of the GAL4 binding sites upstream of a luciferase reporter gene indicated that JMJ functions as a powerful transcriptional repressor. The DNA binding motif of JMJ was determined using CASTing experiments by incubating a random oligonucleotide library with the GST-JMJ fusion protein coupled to agarose beads. Among the selected binding oligonucleotides, the high affinity DNA binding sequences were identified by gel retardation assays. JMJ repressed expression of the reporter genes containing the high affinity JMJ binding sequences, indicating that JMJ is a DNA-binding transcriptional repressor. The domains for transcriptional repression, DNA binding, and nuclear localization signal were mapped by mutational analyses using reporter gene assays, gel retardation assays, and immunostaining experiments, respectively. The present data demonstrate for the first time that JMJ functions as a DNA-binding transcriptional repressor. Therefore, JMJ may play a critical role in transcription factor cascade to regulate expression of heart-specific genes and normal cardiac development.
