The processing of exogenous Ags is an essential step for the generation of immunogenic peptides that will be presented to T cells. This processing relies on the efficient intracellular targeting of ...Ags, because it depends on the content of the compartments in which Ags are delivered in APCs. Opsonization of Ags by the complement component C3 strongly enhances their presentation by B cells and increases their immunogenicity in vivo. To investigate the role of C3 in the targeting of Ags, we compared the intracellular traffic of proteins internalized by complement receptor (CR) and B cell receptor (BCR) in B lymphocytes. Whereas both receptors are able to induce efficient Ag presentation, their intracellular pathways are different. CR ligand is delivered to compartments containing MHC class II molecules (MHC-II) but devoid of transferrin receptor and Lamp-2, whereas BCR rapidly targets its ligand toward Lamp-2-positive, late endosomal MHC-II-enriched compartments through intracellular vesicles containing transferrin receptor. CR and BCR are delivered to distinct endocytic pathways, and the kinetic evolution of the protein content of these pathways is very different. Both types of compartments contain MHC-II, but CR-targeted compartments receive less neosynthesized MHC-II than do BCR-targeted compartments. The targeting induced by CR toward compartments that are distinct from BCR-targeted compartments probably participates in C3 modulation of Ag presentation.

The successful deployment of the Borexino solar neutrino detector required assorted physical and chemical operations to produce exceptional pure fluids and fill multiple detector zones. The ...composition and flow rates of high purity gases and liquids had to be precisely controlled to maintain liquid levels and pressures. The system was required to meet exceptional requirements for cleanliness and leak-tightness. A large scale modular system connecting fluid receiving, purification and fluid delivery processes was developed for Borexino. At the core is a flow control system that delivers scintillator components to plants for purification, and then fills the Borexino detector volumes with ultrahigh purity buffer or ultrahigh purity scintillator. The liquid handling system maintains precise control over the liquid levels and differential pressures between the different volumes of the detectors that are separated by flexible nylon vessels. The preparation, commissioning and operation of the system for filling the Borexino detector with scintillator is described.

In the present study, we describe a method to specifically isolate intracellular compartments containing endocytosed antigen. We have demonstrated that isolated compartments represent a small ...proportion of the intracellular material, highly enriched in antigen. Antigen-containing vesicles are specifically sorted from other intracellular compartments, such as endoplasmic reticulum or Golgi apparatus, and from the plasma membrane. They remain functional in vitro since they can be acidified, and the antigen inside has been found to be partially proteolysed. In macrophages, kinetic analysis has revealed that the antigen is first found in compartments of endosomal density, carrying Rab 5 and Rab 7, then in late compartments of lysosomal density, which are rich in proteases. The global protein content of the compartments was mapped by two-dimensional electrophoresis. In B lymphocytes, this method has allowed the isolation of endocytic compartments emerging from receptor-mediated endocytosis of the antigen. After 2 h of chase, the antigen reached vesicles containing large amounts of MHC-class II molecules, invariant chain and human leucocyte antigen-DM, where peptide loading can occur.
Les femmes obèses en âge de procréer se tournent de plus en plus vers la chirurgie bariatrique. Peu d’études ont évalué l’impact des carences en micronutriments et leur supplémentation sur la ...grossesse et les nouveau-nés. Nous nous sommes concentrés sur le fer, la vitamine A et le zinc. Les objectifs étaient de définir les facteurs préconceptionnels associés à des carences en début de grossesse, de corréler les données biologiques maternelles au cours de la grossesse avec celles des nouveau-nés et de comparer les échantillons de sang au cordon entre les nouveau-nés de mères opérées et ceux des témoins.
Toutes les femmes opérées d’une chirurgie bariatrique et ayant accouché au CHU d’Angers entre 2008 et 2017 ont été incluses. Au cours de 3 visites (V1 entre 12-18 semaines de grossesse GW, V2 entre 19–25 GW, V3 entre 26–33 GW), nous avons recueilli les valeurs d’hémoglobine sérique, le fer (Fe), la ferritine, la vitamine A (Vit A) et le zinc (Zn). Les échantillons de sang au cordon de leurs nouveau-nés ont été comparés à ceux nés de mères saines.
Cent-soixante et onze femmes ont été incluses, âgées en moyenne de 28,8 ans au moment de la chirurgie. Quatre-vingt-cinq pour cent ont été traité par bypass gastrique. Les IMC moyens au moment de la chirurgie et de la grossesse étaient respectivement de 45,7 et 29,4kg/m2. Parmi, 58,5 % des femmes ont présenté une perte d’excès de poids de plus de 60 %, observée chez 144 des femmes, et cela était significativement associée à une carence en Zn à la première visite (V1) (p=0,04), de même qu’un IMC>50kg/m2 à la chirurgie (p=0,03). Des carences en Zn, en Fe et en Vit A ont été observées chez respectivement 44,2 %, 8,1 % et 8 % des femmes à V3, et chez 8,1 %, 7,7 % et 5 % des nouveau-nés. La supplémentation en Zn n’était pas significativement associée à une modification du risque de carence maternelle (p=0,18). Le seul facteur associé à une carence maternelle en Zn était une carence préalablement diagnostiquée, indépendamment du traitement proposé (p<0,001). Enfin, ni les carences maternelles, ni les supplémentations en Zn n’étaient associées à une modification du risque de carence du nouveau né (respectivement p=0,3 et 0,6).
L’IMC initial et la perte de poids importante peuvent être des facteurs de risque de carence chez les femmes enceintes opérées d’une chirurgie bariatrique. Les carences au sang de cordon étaient excessivement rares, expliquant l’absence de facteurs de risques de carences infantiles mis en évidence. Il serait nécessaire d’étudier le risque de développer des carences futures chez les nouveau-nés, et de chercher l’impact tant des carences maternelles que des traitements substitutifs proposés lors de la grossesse sur celles-ci.

Thrombin-induced aggregation and serotonin release were markedly enhanced in platelets from spontaneously hypertensive rats (SHR) when compared with those from normotensive Wistar-Kyoto rats (WKY). ...Since phosphoinositides are involved in calcium-mediated platelet responses, the metabolism of these lipids was investigated in SHR and WKY by using 32P-labeled quiescent platelets. In unstimulated cells, both the rate and extent of 32P incorporation into individual inositol-containing phospholipids and phosphatidic acid were identical in SHR and WKY. This finding suggests that the pool size and basal turnover of phosphoinositides did not differ between the two strains. In contrast, early thrombin-induced phosphoinositide metabolism, when monitored as changes in 32Pphosphatidic acid, was significantly higher in SHR than in WKY. For example, a 20-second exposure to thrombin, 0.3 U/ml, induced the formation of 1.6 times more 32Pphosphatidic acid in SHR than in WKY. These results provide evidence for a leftward shift of the dose-response and time-course curves of thrombin-induced 32Pphosphatidic acid formation in SHR. Moreover, the extent of the difference between SHR and WKY was independent of the extracellular calcium concentration. Following thrombin stimulation, 32Pphosphatidic acid formation likely reflects the initial agonist-receptor interaction; therefore, these results suggest that phospholipase C activity is enhanced in platelets of SHR and that the hypersensitivity of phospholipase C in SHR may play a role in the overall alteration of cell calcium handling and, hence, in the platelet responses of SHR.
La néphrite interstitielle aiguë (NIA) est une cause fréquente d’insuffisance rénale aiguë, dont l’origine médicamenteuse est souvent rapportée. De nombreuses classes médicamenteuses sont ...incriminées, par un mécanisme immuno-allergique non dose-dépendant.
Nous rapportons l’observation d’un patient de 63ans diabétique de type 2 hospitalisé pour une pancréatite et une insuffisance rénale aiguë à diurèse conservée. L’imagerie n’objectivait ni dilatation des voies urinaires, ni coulée de nécrose rétro-péritonéale. Le patient ne présentait aucun signe extrarénal de la lignée immuno-allergique ou dysimmunitaire, pas d’éosinophilurie. La fonction rénale était normale lors de l’introduction de la vildagliptine, soit 21jours avant le début des symptômes. Le traitement a été suspendu et plusieurs séances d’hémodialyse ont été nécessaires. L’histologie rénale retrouvait un aspect de néphropathie tubulo-interstielle aiguë avec ébauche de granulomes. L’imputabilité de la vildagliptine a été retenue sur des critères chronologiques (délai évocateur, évolution à l’arrêt du médicament), histologiques et après l’exclusion des diagnostics différentiels. Une corticothérapie par voie orale a été instaurée pour une durée totale de 3mois. La fonction rénale s’est rapidement améliorée. À 6mois, il persistait une insuffisance rénale chronique séquellaire avec une créatinine à 106μmol/L. Une déclaration de pharmacovigilance a été réalisée.
Les anti diabétiques oraux inhibiteurs de la Di-Peptidyl Peptidase 4 (DPP-4) sont une classe médicamenteuse émergente dans la prise en charge du diabète de type 2 mal contrôlé par la metformine seule. Des réactions d’hypersensibilité graves ont été décrites sous sitagliptine. À notre connaissance, notre cas est le premier rapporté de NIA immuno-allergique sous vildagliptine. Ces effets indésirables graves méritent d’être connus par le néphrologue.

α measurements. Post-shot wall outgassing also increased such that the short term in-vessel retention was only enhanced by about 20%. This increased retention level was confirmed by subsequent analysis of collector probe samples positioned in the SOL near the wall and was lower than one H or D atom per Li atom. Evidence of oxygen gettering by lithium was also found.
