Procyandin A2 (PCA2) is a polyphenolic compound which is isolated from grape seeds. It has been reported that PCA2 exhibits antioxidative and anti-inflammatory effects, but its molecular mechanism is ...still poorly understood. This study tests the hypothesis that PCA2 suppresses lipopolysaccharide (LPS)-induced inflammation and oxidative stress through targeting the nuclear factor-kappaB (NF-kappaB), mitogen-activated protein kinase (MAPK), and NF-E2-related factor 2 (Nrf2) pathways in RAW264.7 cells. PCA2 (20, 40, 80 muM) exhibited no significant cytotoxicity in RAW264.7 cells and showed an inhibitory effect on an LPS-induced nitrite level. Pro-inflammatory cytokines like tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6), prostaglandin E2 (PGE2), nitric oxide (NO), and reactive oxygen species (ROS) were suppressed by PCA2 with a concentration range of 0-80 muM. The mRNA levels of TNF-alpha and IL-6 were inhibited by PCA2 (80 muM). The hallmark-protein expression of the NF-kappaB (p-IKKalpha/beta, p-IkappaBalpha, and p-p65) and MAPK (p-p38, p-JNK, and p-ERK) pathways were decreased by PCA2 in LPS-stimulated RAW264.7 cells. In addition, immunofluorescence results indicated that PCA2 (80 muM) promoted the translocation of NF-kappaB/p65 from the cytoplasm into the nucleus. PCA2 upregulated the expressions of Nrf2 and HO-1 and downregulated the expression of Keap-1. Simultaneously, PCA2 (80 muM) reversed LPS-induced Nrf2 translocation from the nucleus into the cytoplasm. Collectively, PCA2 protect cells against the damage from inflammation and oxidative injury, which suggest a potential therapeutic strategy for inflammatory and oxidative stress through targeting NF-kappaB, MAPK, and Nrf2 pathways in RAW264.7 cells.

In the early February, 2020, we called up an experts’ committee with more than 30 Chinese experts from 11 national medical academic organizations to formulate the first edition of consensus statement ...on diagnosis, treatment and prevention of coronavirus disease 2019 (COVID-19) in children, which has been published in this journal. With accumulated experiences in the diagnosis and treatment of COVID-19 in children, we have updated the consensus statement and released the second edition recently. The current version in English is a condensed version of the second edition of consensus statement on diagnosis, treatment and prevention of COVID-19 in children. In the current version, diagnosis and treatement criteria have been optimized, and early identification of severe and critical cases is highlighted. The early warning indicators for severe pediatric cases have been summarized which is utmost important for clinical practice. This version of experts consensus will be valuable for better prevention, diagnosis and treatment of COVID-19 in children worldwide.

Modern functional adhesives have attracted considerable attention due to their reversible adhesion capacities and stimuli-responsive adhesion behavior. However, for modern functional adhesives, ...polymeric structures were highly necessary to realize adhesion behaviors. Supramolecular adhesives from low-molecular-weight monomers were rarely recognized. Compared with polymeric adhesive materials, it remains challenging for supramolecualr adhesive materials to realize tough adhesion on wet surfaces or even under water. In this study, a new supramolecular adhesive consisting of low-molecular-weight monomers was successfully designed and prepared. Strong and long-term adhesion performance was realized on various surfaces, with a maximum adhesion strength of 4.174 MPa. This supramolecular adhesive exhibits tough and stable adhesion properties in high-moisture and underwater environments (including seawater). Long-term underwater adhesion tests display the potential application of low-molecular-weight adhesive as a marine adhesive.

Revealing the relationship between plants and fungi is very important in understanding biodiversity maintenance, community stability, and ecosystem functioning. However, differences in the community ...and network structures of phyllosphere epiphytic and endophytic fungi are currently poorly documented. In this study, we examined epiphytic and endophytic fungal communities associated with the leaves of six mangrove species using Illumina MiSeq sequencing of internal transcribed spacer 2 (ITS2) sequences.
A total of 635 operational taxonomic units (OTUs) of endophytic and epiphytic fungi were obtained at a 97% sequence similarity level; they were dominated by Dothideomycetes and Tremellomycetes. Plant identity had a significant effect on the OTU richness of endophytic fungi, but not on epiphytic fungi. The community composition of epiphytic and endophytic fungi was significantly different, and plant identity had a greater effect on endophytic fungi than on epiphytic fungi. Network analysis showed that both epiphytic and endophytic network structures were characterized by significantly highly specialized and modular but lowly connected and anti-nested properties. Furthermore, the endophytic network had higher levels of specialization and modularity but lower connectance and stronger anti-nestedness than the epiphytic network.
This study reveals that the phyllosphere epiphytic and endophytic fungal communities differ, and plant identity has a greater effect on the endophytic fungi than on epiphytic fungi. These findings demonstrate the role of host plant identity in driving phyllosphere epiphytic and endophytic community structure.

Huanglian Jiedu Decoction (HLJDD) is a classical herbal formula with potential efficacy in the treatment of sepsis. However, the main components and potential mechanisms of HLJDD remain unclear. This ...study aims to initially clarify the potential mechanism of HLJDD in the treatment of sepsis based on network pharmacology and molecular docking techniques.
The principal components and corresponding protein targets of HLJDD were searched on TCMSP, BATMAN-TCM and ETCM and the compound-target network was constructed by Cytoscape3.8.2. Sepsis targets were searched on OMIM and DisGeNET databases. The intersection of compound target and disease target was obtained and the coincidence target was imported into STRING database to construct a PPI network. We further performed GO and KEGG enrichment analysis on the targets. Finally, molecular docking study was approved for the core target and the active compound.
There are 257 nodes and 792 edges in the component target network. The compounds with a higher degree value are quercetin, kaempferol, and wogonin. The protein with a higher degree in the PPI network is JUN, RELA, TNF. GO and KEGG analysis showed that HLJDD treatment of sepsis mainly involves positive regulation of transcription from RNA polymerase II promoter, negative regulation of apoptosis process, response to hypoxia and other biological processes. The signaling pathways mainly include PI3K-AKT, MAPK, TNF signaling pathway. The molecular docking results showed that quercetin, kaempferol and wogonin have higher affinity with JUN, RELA and TNF.
This study reveals the active ingredients and potential molecular mechanism of HLJDD in the treatment of sepsis, and provides a reference for subsequent basic research.
•Network pharmacology was utilized to determine the mechanisms and targets of HLJDD.•Molecular docking elucidated three key anti-sepsis components in HLJDD.•The combining of molecular docking and network pharmacology provides a way for the study of traditional Chinese medicine.

Abstract
We propose a new, efficient multiscale method to decompose a map (or signal in general) into component maps that contain structures of different sizes. In the widely used wave transform, ...artifacts containing negative values arise around regions with sharp transitions due to the application of band-limited filters. In our approach, the decomposition is achieved by solving a modified, nonlinear version of the diffusion equation. This is inspired by the anisotropic diffusion methods, which establish the link between image filtering and partial differential equations. In our case, the artifact issue is addressed where the positivity of the decomposed images is guaranteed. Our new method is particularly suitable for signals which contain localized, nonlinear features, as typical of astronomical observations. It can be used to study the multiscale structures of astronomical maps quantitatively and should be useful in observation-related tasks such as background removal. We thus propose a new measure called the “scale spectrum,” which describes how the image values distribute among different components in the scale space, to describe maps. The purpose of this paper is to explore the impact of the COVID-19 epidemic on global trade and its transformation of trade relations between the United States and China. As the two largest economies, the trade relationship between the United States and China plays an important role in this process. This paper analyzes the causes of deglobalization, which are mainly caused by the fracture of industrial chain, trade protection and industrial upgrading. At the same time, the paper focuses on the factors that have caused the deglobalization trend in the post-COVID-19 era, such as changing trade agreements, poor cross-border investment environment, and the challenges and uncertainties brought by e-commerce. Finally, it concentrates on the changes of the US and China bilateral trade in the post-COVID-19 era. Affected by the epidemic, the bilateral relationship between these two countries has become more conflictual and focused on technological competition. Periodically driven systems have displayed a variety of fascinating phenomena without analogies in static systems, which enrich the classification of quantum phases of matter and stimulate a wide ...range of research interests. Here, we employ discrete-time quantum walks to investigate a nontrivial topological effect unique to a two-dimensional periodically driven system: chiral edge states can exist at the interface of Floquet insulators whose Chern numbers vanish. Thanks to a resource-saving and flexible fiber-loop architecture, we realize inhomogeneous two-dimensional quantum walks up to 25 steps, over an effective 51×51 lattice with tunable local parameters. Spin-polarized chiral edge states are observed at the boundary of two distinct quantum walk domains. Our results contribute to establishing a well-controlled platform for exploring nontrivial topological phases.

Relativistic spin-polarized positron beams are indispensable for future electron-positron colliders to test modern high-energy physics theory with high precision. However, present techniques require ...very large scale facilities for those experiments. We put forward a novel efficient method for generating ultrarelativistic polarized positron beams employing currently available laser fields. For this purpose, the generation of polarized positrons via multiphoton Breit-Wheeler pair production and the associated spin dynamics in single-shot interaction of an ultraintense laser pulse with an ultrarelativistic electron beam is investigated in the quantum radiation-dominated regime. The pair production spin asymmetry in strong fields, significantly exceeding the asymmetry of the radiative polarization, produces locally highly polarized particles, which are split by a specifically tailored small ellipticity of the laser field into two oppositely polarized beams along the minor axis of laser polarization. In spite of radiative de-polarization, a dense positron beam with up to about 90% polarization can be generated in tens of femtoseconds. The method may eventually usher high-energy physics studies into smaller-scale laser laboratories.
