It is well known that the calcium ion is essential for maintaining life activities in living organisms, and it is of great significance to detect the intracellular calcium concentration. For the ...detection of calcium ions, we developed a new type of fluorescent carbon dots (CDs), whose surface was modified by ethylenebis(oxyethylenenitrilo)tetraacetic acid (EGTA) through a secondary hydrothermal method. This is a simple and convenient chemical preparation method because all reactions are carried out in the same autoclave, and the final product is directly the EGTA-modified CDs. The CDs exhibit bright blue fluorescence, and as the calcium concentration increases, the fluorescence intensity drops sharply. The fluorescence quenching correlates with the concentration of calcium ions and has a good linearity in the range of 15–300 μM with a detection limit of 0.38 μM. The experimental results confirmed that the detection of calcium ions by CDs is a static fluorescence quenching process. Also, cytotoxicity test and cellular imaging experiments have shown that the CDs are nontoxic and biocompatible.

To report long‐term results of a randomized controlled trial that compared cisplatin/fluorouracil/docetaxel (TPF) induction chemotherapy (IC) plus concurrent chemoradiotherapy (CCRT) with CCRT alone ...in locoregionally advanced nasopharyngeal carcinoma (NPC). Patients with stage III–IVB (except T3–4 N0) NPC were randomly assigned to receive IC plus CCRT (n = 241) or CCRT alone (n = 239). IC included three cycles of docetaxel (60 mg/m2 d1), cisplatin (60 mg/m2 d1), and fluorouracil (600 mg/m2/d civ d1–5) every 3 weeks. Patients from both groups received intensity‐modulated radiotherapy concurrently with three cycles of 100 mg/m2 cisplatin every 3 weeks. After a median follow‐up of 71.5 months, the IC plus CCRT group showed significantly better 5‐year failure‐free survival (FFS, 77.4% vs. 66.4%, p = 0.019), overall survival (OS, 85.6% vs. 77.7%, p = 0.042), distant failure‐free survival (88% vs. 79.8%, p = 0.030), and locoregional failure‐free survival (90.7% vs. 83.8%, p = 0.044) compared to the CCRT alone group. Post hoc subgroup analyses revealed that beneficial effects on FFS were primarily observed in patients with N1, stage IVA, pretreatment lactate dehydrogenase ≥170 U/l, or pretreatment plasma Epstein–Barr virus DNA ≥6000 copies/mL. Two nomograms were further developed to predict the potential FFS and OS benefit of TPF IC. The incidence of grade 3 or 4 late toxicities was 8.8% (21/239) in the IC plus CCRT group and 9.2% (22/238) in the CCRT alone group. Long‐term follow‐up confirmed that TPF IC plus CCRT significantly improved survival in locoregionally advanced NPC with no marked increase in late toxicities and could be an option of treatment for these patients.

Here the authors find that combining the conventional chemoradiotherapy with a triple induction chemotherapy (cisplatin/fluorouracil/docetaxel) prolonged survival of patients with locoregionally advanced cancer, even after more than 70 months of follow‐up. The combination treatment increased acute, but not late, toxicities, and the authors propose that it could present a new treatment option for this patient group.

There is a pressing need to develop nanoplatforms that integrate multimodal therapeutics to improve treatment responses and prolong the survival of patients with unresectable hepatocellular carcinoma ...(HCC). Mesoporous silica-coated gold nanomaterials have emerged as a novel multifunctional platform combining tunable surface plasmon resonance and mesoporous properties that exhibit multimodality properties in cancer theranostics. However, their reduced radiation-absorption efficiency and limited surface area hinder their further radiochemotherapeutic applications. To address these issues, we designed Janus-structured gold-mesoporous silica nanoparticles using a modified sol–gel method. This multifunctional theranostic nanoplatform was subsequently modified via the conjugation of folic acid for enhanced HCC targeting and internalization. The loaded anticancer agent doxorubicin can be released from the mesopores in a pH-responsive manner, facilitating selective and safe chemotherapy. Additionally, the combination of chemotherapy and radiotherapy induced synergistic anticancer effects in vitro and exhibited remarkable inhibition of tumor growth in vivo along with significantly reduced systematic toxicity. Additionally, the Janus NPs acted as targeted computed tomography (CT)-imaging agents for HCC diagnosis. Given their better performance in chemoradiotherapy and CT imaging as compared with that of their core–shell counterparts, this new nanoplatform designed with dual functionalities provides a promising strategy for unresectable HCC theranostics.

A dearomative 4 + 2 cycloaddition of 3‐nitroindoles ortho‐amino Morita−Baylis−Hillman carbonates was established under mild conditions. This method provides an efficient and practical approach for ...delivering tetrahydro‐5H‐indolo2,3‐bquinolines containing three contiguous stereocenters, two tertiary and one quaternary, in high yield (up to 95%) with excellent diastereoselectivity (all cases >25:1 dr). The potential synthetic applications of this strategy were also highlighted by the scale‐up experiment and further synthetic transformation. Moreover, the structure and relative configuration of the cycloadduct was unequivocally confirmed by single‐crystal X‐ray diffraction.

Demographic change of human populations is one of the central questions for delving into the past of human beings. To identify major population expansions related to male lineages, we sequenced 78 ...East Asian Y chromosomes at 3.9 Mbp of the non-recombining region, discovered >4,000 new SNPs, and identified many new clades. The relative divergence dates can be estimated much more precisely using a molecular clock. We found that all the Paleolithic divergences were binary; however, three strong star-like Neolithic expansions at ∼6 kya (thousand years ago) (assuming a constant substitution rate of 1×10(-9)/bp/year) indicates that ∼40% of modern Chinese are patrilineal descendants of only three super-grandfathers at that time. This observation suggests that the main patrilineal expansion in China occurred in the Neolithic Era and might be related to the development of agriculture.

It is well-known that the ratio of reduced l-glutathione (GSH) to oxidized l-glutathione (GSSG) is a vital biomarker for monitoring overall cellular health, thus detecting the intracellular ...concentration of glutathione is of great significance. Recently, an increasing number of reports have published various methods for GSH detection, but studies on the detection of GSSG are still rare. Here, we report a kind of new yellow fluorescent carbon dots (CDs) for the detection of GSSG through a fluorescence “off-on” process. Because the surface is rich in amino groups, the CDs show a positive potential. When the concentration of GSSG was continuously increased, the CDs’ fluorescence dropped sharply, while the fluorescence gradually recovered after the addition of sodium sulfide. The phenomenon of fluorescence quenching is linear with the concentration of the quencher (GSSG)(0–200 μM), and 0.18 μM is calculated as the detection limit. More interestingly, as a fluorescent probe, the CDs can be further used for fluorescence imaging in living cells and zebrafish.

Magnetic nanoparticles (NPs) have emerged as a promising tool for suicide gene therapy. However, the separate delivery of the suicide gene and prodrug in current systems limits their clinical ...translation. Therefore, improving magnetically mediated suicide gene therapy by exploring higher performance magnetic NP-based hybrid nanoplatforms is an important challenge. In the current study, shape-controlled magnetic mesoporous silica nanoparticles (M-MSNs) were prepared, and their performance in magnetic resonance imaging (MRI)-guided, magnetically targeted and hyperthermia-enhanced suicide gene therapy of hepatocellular carcinoma (HCC) was investigated. Compared with sphere-like MSNs, rod-like MSNs exhibited higher loading capacity, faster prodrug release behavior, stronger magnetically enhanced gene delivery and better magnetic hyperthermia properties. Utilizing the improved magnetic properties of the M-MSNs allowed us to demonstrate highly effective dual magnetically enhanced suicide gene therapy in vivo with decreased systematic toxicity and with the ability to monitor therapeutic outcome by MRI. Because of their magnetic targeting abilities, magnetic hyperthermia performance and MRI properties, these M-MSNs might prove to be a potentially superior candidate for suicide gene therapy of HCC.

Iron, nitrogen-co-doped carbon quantum dots (Fe,N-CDs) were prepared

a simple one-step hydrothermal method. The quantum yield of fluorescence reached about 27.6% and the blue-emissive Fe,N-CDs ...had a mean size of 3.76 nm. The as-prepared carbon quantum dots showed good solubility, a high quantum yield, good biocompatibility, low cytotoxicity, and high photostability. Interestingly, the as-prepared Fe,N-CDs exhibited good selectivity and sensitivity toward both hematin and ferric ions, and the limit of detection for hematin and ferric ions was calculated to be about 0.024 μM and 0.64 μM, respectively. At the same time, Fe,N-CDs were used for imaging HeLa cells and showed that most Fe,N-CDs were detained in the lysosome. Well-intergrown hydrophilic CAU-10-H membranes were successfully prepared by microwave-assisted secondary growth. The as-synthesized CAU-10-H membranes exhibited high and reproducible performance for ...ethanol dehydration. For a 90 wt% ethanol feed, the CAU-10-H membrane had a total flux of 493 g m-2 h-1 and water/ethanol separation factor of 148 at 65 °C. Along with the increasing temperature or water feed concentration, the total flux increased while the water/ethanol separation factor decreased. The ideal water/ethanol selectivities of the CAU-10-H membranes were found to be significantly lower than its membrane selectivities, demonstrating that the preferential adsorption of water on hydrophilic CAU-10-H membrane to hinder ethanol transport played a prominent part in the ethanol-water separation. The CAU-10-H membranes are capable of being used in acidic conditions, and therefore showing great prospects for industrial applications.
•Hydrophilic CAU-10-H is stable in water at 80 °C around 190 h.•A dense CAU-10-H membrane was synthesized by microwave-assisted secondary growth.•CAU-10-H membranes showed high pervaporation performance for ethanol dehydration.•The performance of CAU-10-H membranes remained unchanged under acidic conditions.
