Explanations for the barrier effect of the Indo-Pacific Maritime Continent (MC) on the MJO should satisfy two criteria. First, they should include specific features of the MC, namely, its intricate ...land–sea distributions and elevated terrains. Second, they should include mechanisms for both the barrier effect and its overcoming by some MJO events. Guided by these two criteria, a precipitation-tracking method is applied to identify MJO events that propagate across the MC (MJO-C) and those that are blocked by the MC (MJO-B). About a half of MJO events that form over the Indian Ocean propagate through the MC. Most of them (.75%) become weakened over the MC. The barrier effect cannot be explained in terms of the strength, horizontal scale, or spatial distribution of MJO convection when it approaches the MC from the west. A distinction between MJO-B and MJO-C is their precipitation over the sea versus land in the MC region. MJO-C events rain much more over the sea than over land, whereas rainfall over the sea never becomes dominant for MJO-B. This suggests that inhibiting convective development over the sea could be a possible mechanism for the barrier effect of the MC. Preceding conditions for MJO-C include stronger low-level zonal moisture flux convergence and higher SST in the MC region. Possible connections between these large-scale conditions and the land versus sea distributions of MJO rainfall through the diurnal cycle are discussed.

ObjectiveDiabetes mellitus (DM) is associated with an increased fracture risk; however, the impact of DM and subsequent fracture at different sites and the associations according to patient ...characteristics remain unknown.DesignMeta-analysisData sourcesThe PubMed, EMBASE and Cochrane Library databases were searched from inception to March 2018.Eligibility criteriaWe included prospective and retrospective cohort studies on the associations of DM and subsequent fracture risk at different sites.Data extraction and synthesisTwo authors independently extracted data and assessed the study quality. Relative risks (RRs) with 95% CIs were calculated using a random-effects model, and the heterogeneity across the included studies was evaluated using I2 and Q statistics.ResultsOverall, DM was associated with an increased risk of total (RR: 1.32; 95% CI 1.17 to 1.48; p<0.001), hip (RR: 1.77; 95% CI 1.56 to 2.02; p<0.001), upper arm (RR: 1.47; 95% CI 1.02 to 2.10; p=0.037) and ankle fractures (RR: 1.24; 95% CI 1.10 to 1.40; p<0.001), whereas DM had no significant impact on the incidence of distal forearm (RR: 1.02; 95% CI 0.88 to 1.19; p=0.809) and vertebral fractures (RR: 1.56; 95% CI 0.78 to 3.12; p=0.209). RR ratios suggested that compared with patients with type 2 DM (T2DM), patients with type 1 DM (T1DM) had greater risk of total (RR: 1.24; 95% CI 1.08 to 1.41; p=0.002), hip (RR: 3.43; 95% CI 2.27 to 5.17; p<0.001) and ankle fractures (RR: 1.71; 95% CI 1.06 to 2.78; p=0.029). Although no other significant differences were observed between subgroups, the association of DM with upper arm or ankle, vertebrae and total fracture differed according to sex, study design and country, respectively.ConclusionsPatients with DM had greater risks of total, hip, upper arm and ankle fractures, with T1DM having a more harmful effect than T2DM.
Plants of Artemisia annua produce artemisinin, a sesquiterpene lactone widely used in malaria treatment. Amorpha-4,11-diene synthase （ADS）, a sesquiterpene synthase, and CYP71AV1, a P450 ...monooxygenase, are two key enzymes of the artemisinin biosynthesis pathway. Accumulation of artemisinin can be induced by the phytohormone jasmonate （JA）. Here, we report the characterization of two JA-responsive AP2 family transcription factors-AaERF1 and AaERF2-from A. annua L. Both genes were highly expressed in inflorescences and strongly induced by JA. Yeast one- hybrid and electrophoretic mobility shift assay （EMSA） showed that they were able to bind to the CRTDREHVCBF2 （CBF2） and RAVlAAT （RAA） motifs present in both ADS and CYP71AV1 promoters. Transient expression of either AaERF1 or AaERF2 in tobacco induced the promoter activities of ADS or CYP71AV1, and the transgenic A. annua plants overexpressing either transcription factor showed elevated transcript levels of both ADS and CYP71AV1, resulting in increased accumulation of artemisinin and artemisinic acid. By contrast, the contents of these two metabolites were reduced in the RNAi transgenic lines in which expression of AaERF1 or AaERF2 was suppressed. These results demonstrate that AaERF1 and AaERF2 are two positive regulators of artemisinin biosynthesis and are of great value in genetic engineering of arte- misinin production.

RATIONALE:Necrosis is one of the main forms of cardiomyocyte death in heart disease. Recent studies have demonstrated that certain types of necrosis are regulated and programmed dependent on the ...activation of receptor-interacting serine/threonine-protein kinase (RIPK) 1 and 3 which may be negatively regulated by Fas-associated protein with death domain (FADD). In addition, microRNAs and long noncoding RNAs have been shown to play important roles in various biological processes recently.
OBJECTIVE:The purpose of this study was to test the hypothesis that microRNA-103/107 and H19 can participate in the regulation of RIPK1- and RIPK3-dependent necrosis in fetal cardiomyocyte-derived H9c2 cells and myocardial infarction through targeting FADD.
METHODS AND RESULTS:Our results show that FADD participates in H2O2-induced necrosis by influencing the formation of RIPK1 and RIPK3 complexes in H9c2 cells. We further demonstrate that miR-103/107 target FADD directly. Knockdown of miR-103/107 antagonizes necrosis in the cellular model and also myocardial infarction in a mouse ischemia/reperfusion model. The miR-103/107-FADD pathway does not participate in tumor necrosis factor-α–induced necrosis. In exploring the molecular mechanism by which miR-103/107 are regulated, we show that long noncoding RNA H19 directly binds to miR-103/107 and regulates FADD expression and necrosis.
CONCLUSIONS:Our results reveal a novel myocardial necrosis regulation model, which is composed of H19, miR-103/107, and FADD. Modulation of their levels may provide a new approach for preventing myocardial necrosis.
Summary
Almost all plants form trichomes, which protect them against insect herbivores by forming a physical barrier and releasing chemical repellents. Glandular trichomes produce a variety of ...specialized defensive metabolites, including volatile terpenes. Previous studies have shown that the defence hormone jasmonic acid (JA) affects trichome development and induces terpene synthases (TPSs) but the underlying molecular mechanisms remain unclear. Here, we characterized a loss‐of‐function allele of the HD‐ZIP IV transcription factor woolly (wo) and analysed its role in mediating JA signalling in tomato. We showed that knockout of wo led to extensive trichome defects, including structural and functional changes in type VI glandular trichomes, and a dramatic reduction in terpene levels. We further found that wo directly binds to TPS gene promoters to recruit SlMYC1, a JA signalling modulator, and that together these transcription factors promote terpene biosynthesis in tomato trichomes. The wo/SlMYC1 regulatory module is inhibited by SlJAZ2 through a competitive binding mechanism, resulting in a fine‐tuned JA response in tomato trichomes. Enhanced expression of SlMYC1 substantially increased terpene levels and improved tomato resistance to spider mites. Interestingly, we also found that SlMYC1 plays an additional role in glandular cell division and expansion in type VI trichomes, independent of JA. Together, our results reveal a novel, JA‐mediated regulatory mechanism that promotes insect resistance in tomato.

While there is substantial evidence that emotion regulation plays a role in the maintenance of substance and behavior addiction, its role in addiction to social networking sites (SNS) remains ...unclear. Drawing on attachment theory, we explore whether emotion regulation mediates the relationship between insecure attachment and SNS addiction among 463 college students. The participants completed the short version of the Experience in Close Relationships Scale, Difficulties in Emotion Regulation Scale, and Chinese Social Media Addiction Scale. The results indicated that attachment anxiety positively predicted SNS addiction and that emotion regulation mediated this link. These findings suggest that individuals' affective regulation capability should be a target of future interventions and treatments.
Security has always been a popular and critical topic. With the rapid development of information technology, it is always attracting people＇s attention. However, since security has a long history, it ...covers a wide range of topics which change a lot, from classic cryptography to recently popular mobile security. There is a need to investigate security-related topics and trends, which can be a guide for security researchers, security educators and security practitioners. To address the above-mentioned need, in this paper, we conduct a large-scale study on security-related questions on Stack Overflow. Stack Overflow is a popular on-line question and answer site for software developers to communicate, collaborate, and share information with one another. There are many different topics among the numerous questions posted on Stack Overflow and security-related questions occupy a large proportion and have an important and significant position. We first use two heuristics to extract from the dataset the questions that are related to security based on the tags of the posts. And then we use an advanced topic model, Latent Diriehlet Allocation （LDA） tuned using Genetic Algorithm （GA）, to cluster different security-related questions based on their texts. After obtaining the different topics of security-related questions, we use their metadata to make various analyses. We summarize all the topics into five main categories, and investigate the popularity and difficulty of different topics as well. Based on the results of our study, we conclude several implications for researchers, educators and practitioners.

Differential expression of homoeologous genes in polyploids is thought to be important to plant adaptation and novel phenotypes. PRE1 expression is specific to cotton fiber cells, upregulated during their rapid elongation stage and A-homoeologous biased in allotetraploid cultivars. Transgenic studies demonstrated that PRE1 is a positive regulator of fiber elongation.
We determined that the natural variation of the canonical TATA-box, a regulatory element commonly found in many eukaryotic core promoters, is necessary for subgenome-biased PRE1 expression, representing a mechanism underlying the selection of homoeologous genes.
Thus, variations in the promoter of the cell elongation regulator gene PRE1 have contributed to spinnable fiber formation in cotton. Overexpression of GhPRE1 in transgenic cotton yields longer fibers with improved quality parameters, indicating that this bHLH gene is useful for improving cotton fiber quality.

Immunity deteriorates with age in animals but comparatively little is known about the temporal regulation of plant resistance to herbivores. The phytohormone jasmonate (JA) is a key regulator of ...plant insect defense. Here, we show that the JA response decays progressively in Arabidopsis. We show that this decay is regulated by the miR156-targeted SQUAMOSA PROMOTER BINDING PROTEIN-LIKE9 (SPL9) group of proteins, which can interact with JA ZIM-domain (JAZ) proteins, including JAZ3. As SPL9 levels gradually increase, JAZ3 accumulates and the JA response is attenuated. We provide evidence that this pathway contributes to insect resistance in young plants. Interestingly however, despite the decay in JA response, older plants are still comparatively more resistant to both the lepidopteran generalist Helicoverpa armigera and the specialist Plutella xylostella, along with increased accumulation of glucosinolates. We propose a model whereby constitutive accumulation of defense compounds plays a role in compensating for age-related JA-response attenuation during plant maturation.
Evidence supports predictive roles of adult attachment orientations for the maintenance of social networking site (SNS) addiction, but the underlying mechanisms are mostly unknown. Based on ...attachment theory, this study explored whether online social support and the fear of missing out mediated the relationship between insecure attachment and social networking site addiction among 463 college students in China. A questionnaire was used to collect data using the Experience in Close Relationship Scale-Short Form, online social support scale, fear of missing out scale, and Chinese Social Media Addiction Scale. The results showed that online social support and fear of missing out mediated the relationship between anxious attachment and social networking site addiction in parallel paths and serially, and online social support negatively mediated the relationship between avoidant attachment and social networking site addiction. Theoretically, the present study contributes to the field by showing how insecure attachment is linked to SNS addiction. Practically, these findings could aid in future studies on SNS addiction prevention and interventions. Limitations of the present study were discussed.
