A newly emerged coronavirus (COVID-19) seriously threatens human life and health worldwide. In coping and fighting against COVID-19, the most critical step is to effectively screen and diagnose ...infected patients. Among them, chest X-ray imaging technology is a valuable imaging diagnosis method. The use of computer-aided diagnosis to screen X-ray images of COVID-19 cases can provide experts with auxiliary diagnosis suggestions, which can reduce the burden of experts to a certain extent. In this study, we first used conventional transfer learning methods, using five pre-trained deep learning models, which the Xception model showed a relatively ideal effect, and the diagnostic accuracy reached 96.75%. In order to further improve the diagnostic accuracy, we propose an efficient diagnostic method that uses a combination of deep features and machine learning classification. It implements an end-to-end diagnostic model. The proposed method was tested on two datasets and performed exceptionally well on both of them. We first evaluated the model on 1102 chest X-ray images. The experimental results show that the diagnostic accuracy of Xception + SVM is as high as 99.33%. Compared with the baseline Xception model, the diagnostic accuracy is improved by 2.58%. The sensitivity, specificity and AUC of this model reached 99.27%, 99.38% and 99.32%, respectively. To further illustrate the robustness of our method, we also tested our proposed model on another dataset. Finally also achieved good results. Compared with related research, our proposed method has higher classification accuracy and efficient diagnostic performance. Overall, the proposed method substantially advances the current radiology based methodology, it can be very helpful tool for clinical practitioners and radiologists to aid them in diagnosis and follow-up of COVID-19 cases.

HSP90 is a member of the family of heat shock proteins responsible for folding proteins into mature conformations and thus maintaining their biological function in cells. Since it is involved in all ...hallmarks of cancer, HSP90 has been considered as a promising drug target for cancer therapy. Eighteen HSP90 inhibitors have entered clinical trials, however, none has been approved by the FDA. There is still a great need for novel HSP90 inhibitors with strong anticancer activity and good safety profile. In the past several years, many new molecules were identified as HSP90 inhibitors and some of them have shown promising pharmacological profiles in preclinical evaluations. In this review, HSP90 inhibitors identified from 2014 to date are summarized and their design strategies, chemical structures, and biological activities are reviewed. The inhibitors are categorized by their different target domains and selectivity as N-terminal, C-terminal, and isoform-selective HSP90 inhibitors.
To explore the joint and independent effects of gestational weight gain (GWG) and pre-pregnancy body mass index (BMI) on pregnancy outcomes in a population of Chinese Han women and to evaluate ...pregnant women's adherence to the 2009 Institute of Medicine (IOM) gestational weight gain guidelines.
This was a multicenter, retrospective cohort study of 48,867 primiparous women from mainland China who had a full-term singleton birth between January 1, 2011 and December 30, 2011. The independent associations of pre-pregnancy BMI, GWG and categories of combined pre-pregnancy BMI and GWG with outcomes of interest were examined using an adjusted multivariate regression model. In addition, women with pre-pregnancy hypertension were excluded from the analysis of the relationship between GWG and delivery of small-for-gestational-age (SGA) infants, and women with gestational diabetes (GDM) were excluded from the analysis of the relationship between GWG and delivery of large-for-gestational-age (LGA) infants.
Only 36.8% of the women had a weight gain that was within the recommended range; 25% and 38.2% had weight gains that were below and above the recommended range, respectively. The contribution of GWG to the risk of adverse maternal and fetal outcomes was modest. Women with excessive GWG had an increased likelihood of gestational hypertension (adjusted OR 2.55; 95% CI = 1.92-2.80), postpartum hemorrhage (adjusted OR 1.30; 95% CI = 1.17-1.45), cesarean section (adjusted OR 1.31; 95% CI = 1.18-1.36) and delivery of an LGA infant (adjusted OR 2.1; 95% CI = 1.76-2.26) compared with women with normal weight gain. Conversely, the incidence of GDM (adjusted OR 1.64; 95% CI = 1.20-1.85) and SGA infants (adjusted OR 1.51; 95% CI = 1.32-1.72) was increased in the group of women with inadequate GWG. Moreover, in the obese women, excessive GWG was associated with an apparent increased risk of delivering an LGA infant. In the women who were underweight, poor weight gain was associated with an increased likelihood of delivering an SGA infant. After excluding the mothers with GDM or gestational hypertension, the ORs for delivery of LGA and SGA infants decreased for women with high GWG and increased for women with low GWG.
GWG above the recommended range is common in this population and is associated with multiple unfavorable outcomes independent of pre-pregnancy BMI. Obese women may benefit from avoiding weight gain above the range recommended by the 2009 IOM. Underweight women should avoid low GWG to prevent delivering an SGA infant. Pregnant women should therefore be monitored to comply with the IOM recommendations and should have a balanced weight gain that is within a range based on their pre-pregnancy BMI.

Flavins are a class of organic compounds with the basic structure of 7,8-dimethy-10-alkyl isoalloxazine. They are ubiquitous in nature and participate in many biochemical reactions. Due to various ...existing forms, there is a lack of systematic research on the absorption and fluorescence spectra of flavins. In this study, employing the density functional theory (DFT) and time-dependent (TD) DFT, we calculated the pH-dependent absorption and fluorescence spectra of flavin of three redox states (quinone, semiquinone, and hydroquinone) in solvents. The chemical equilibrium of three redox states of flavins and the pH effect on the absorption spectra and fluorescence spectra of flavins were carefully discussed. The conclusion helps with identifying the existing forms of flavins in solvent with different pH values.

Paeoniflorin is traditionally used to treat inflammatory disorders. In our laboratory, we have scientifically validated the anti-inflammatory effect of paeoniflorin. In this study, it has been aimed ...to evaluate in vivo anti-inflammatory effect of paeoniflorin isolated from the dried peeled root of Paeonia lactiflora Pall. It was further intended to find out the probable mechanism of anti-inflammatory effect of paeoniflorin. The anti-inflammatory effect of paeoniflorin (15, 30 and 45mg/kg) was measured employing TNBS-induced ulcerative colitis model of acute inflammation. The TNBS injection resulted significant colitis formation when compared with un-injected mice.
The anti-inflammatory effects of paeoniflorin for ulcerative colitis were assessed by body weight, colonic weight and length, macroscopic scores, and histopathological examinations. In addition, the colonic tissue levels of inflammation markers, including myeloperoxidase (MPO), IL-2, IL-6, IL-10, IL-12, IL-1β, TNF-α and IFN-γ were also determined to assess the effect of paeoniflorin. In addition, western blot demonstrated that paeoniflorin inhibited NF-kappaB signaling pathway and apoptosis in TNBS-induced ulcerative colitis tissues. In conclusion, all the findings of this study suggested that paeoniflorin has the anti-inflammatory effect in ulcerative colitis via inhibiting MAPK/NF-kappaB pathway and apoptosis in mice.

SUMMARY
Central loop transient electromagnetic (TEM) data are often interpreted by conventional 1-D or quasi-2-D inversion techniques. For example, the lateral constrained inversion (LCI) is a ...powerful technique for quick interpretation of central loop TEM data, and can produce spatially consistent resistivity images for profile data by assuming spatial correlation between adjacent model parameters. Such quasi-2-D techniques are very powerful in cases multidimensional effects are small or negligible. However, the inverse solution of conventional LCI methods strongly depends on subjective interpreter choices such as the model regularization and the imposed lateral constraints. Due to inherent non-linearity and nonuniqueness of the TEM inverse problems, this can result in biased model parameters and their estimated model uncertainties. We present a transdimensional Markov chain Monte Carlo method for the quasi-2-D inversion of TEM data using a Bayesian inference framework. We term the approach quasi-2-D, since the model is parametrized in 2-D with unstructured Voronoi cells, whereas the TEM response at each station is predicted using a 1-D forward solution to make the problem computationally affordable. During the inversion, the number of Voronoi cells as well as their positions and resistivities are variable. Accordingly, the level of model complexity is automatically determined by the framework and adapted to the spatial resolution of the data, thus avoiding the need for subjective model regularization or spatial constraints. The approach is validated using synthetic data and compared to 1-D Bayesian and conventional Gauss Newton inversion techniques. The application to dense field data from a floating TEM survey leads to a consistent subsurface image with unbiased uncertainty estimates and a plausible depth of investigation. The quantitative uncertainty information provided by the Bayesian framework is beneficial in identifying resolution.
Among all bioluminescent organisms, the firefly is the most famous, with a high luminescent efficiency of 41%, which is widely used in the fields of biotechnology, biomedicine and so on. The entire ...bioluminescence (BL) process involves a series of complicated in-vivo chemical reactions. The BL is initiated by the enzymatic oxidation of luciferin (LH2). However, the mechanism of the efficient spin-forbidden oxygenation is far from being totally understood. Via MD simulation and QM/MM calculations, this article describes the complete process of oxygenation in real protein. The oxygenation of luciferin is initiated by a single electron transfer from the trivalent anionic LH2 (L3−) to O2 to form 1L•2−…O2•−; the entire reaction is carried out along the ground-state potential energy surface to produce the dioxetanone (FDO−) via three transition states and two intermediates. The low energy barriers of the oxygenation reaction and biradical annihilation involved in the reaction explain this spin-forbidden reaction with high efficiency. This study is helpful for understanding the BL initiation of fireflies and the other oxygen-dependent bioluminescent organisms.

On May 28, 2021, the FDA granted accelerated approval to sotorasib (Lumakras, Amgen) for the treatment of adults with advanced non-small cell lung cancer (NSCLC) with a Kirsten rat sarcoma ...proto-oncogene (KRAS) G12C mutation who have received at least one prior systemic therapy. The approval was based on CodeBreaK 100 (Study 20170543), a dose-escalation and dose-expansion trial in patients with an advanced, KRAS G12C-mutated, solid tumor. The overall response rate (ORR) observed in patients with KRAS G12C-mutated NSCLC treated with sotorasib (n = 124) was 36% 95% confidence interval (CI), 28-45. The median duration of response was 10.0 months (95% CI, 6.9-not estimable). The most common adverse reactions (≥20%) were diarrhea, musculoskeletal pain, nausea, fatigue, hepatotoxicity, and cough. This is the first approval of a targeted therapy for KRAS G12C-mutated NSCLC. Because of pharmacokinetic data and ORRs of patient cohorts who took sotorasib at lower doses in the dose-escalation portion of CodeBreaK 100, a dose comparison study is being conducted as a post-marketing requirement.
