p21WAF1/CIP1 is a broad-acting cyclin-dependent kinase inhibitor. Its stability is essential for proper cell-cycle progression and cell fate decision. Ubiquitylation by the multiple E3 ubiquitin ...ligase complexes is the major regulatory mechanism of p21, which induces p21 degradation. However, it is unclear whether ubiquitylated p21 can be recycled. In this study, we report USP11 as a deubiquitylase of p21. In the nucleus, USP11 binds to p21, catalyzes the removal of polyubiquitin chains conjugated onto p21, and stabilizes p21 protein. As a result, USP11 reverses p21 polyubiquitylation and degradation mediated by SCFSKP2, CRL4CDT2, andAPC/CCDC20 in a cell-cycle–independent manner. Loss of USP11 causes the destabilization of p21 and induces the G1/S transition in unperturbed cells. Furthermore, p21 accumulation mediated by DNA damage is completely abolished in cells depleted of USP11, which results in abrogation of the G2 checkpoint and induction of apoptosis. Functionally, USP11-mediated stabilization of p21 inhibits cell proliferation and tumorigenesis in vivo. These findings reveal an important mechanism by which p21 can be stabilized by direct deubiquitylation, and they pinpoint a crucial role of the USP11-p21 axis in regulating cell-cycle progression and DNA damage responses.

Three-dimensional ordered Sn-KIT-6 mesoporous silica materials are prepared by one-step hydrothermal synthesis method. The desulfurization rate of Sn-KIT-6 achieved 99.7% under the optimal reaction conditions. SnO2 was proposed as the catalytic site for the oxidative desulfurization. The excellent catalytic performance was attributed to the high dispersion of SnO2 in KIT-6. The desulfurization rate of Sn-KIT-6 reduced to 81% after 6 recycles, indicating SnO2 may run off in the oxidative desulfurization process.

Salmonella, an important foodborne pathogen, was estimated to be responsible for 95.1 million cases and 50,771 deaths worldwide. Sixteen serovars were responsible for approximately 80% of Salmonella ...infections in humans in China, and infections caused by a few uncommon serovars have been reported in recent years, though not with S. Welikade. This study reports the first clinical case caused by S. Welikade in China and places Chinese S. Welikade isolates in the context of global isolates via genomic analysis. For comparison, S. Welikade isolates were also screened in the Chinese Local Surveillance System for Salmonella (CLSSS). The minimum inhibitory concentrations (MICs) of 28 antimicrobial agents were determined using the broth microdilution method. The isolates were sequenced on an Illumina platform to identify antimicrobial resistance genes, virulence genes, and phylogenetic relationships. The S. Welikade isolate (Sal097) was isolated from a two-year-old boy with acute gastroenteritis in 2021. Along with the other two isolates found in CLSSS, the three Chinese isolates were susceptible to all the examined antimicrobial agents, and their sequence types (STs) were ST5123 (n = 2) and ST3774 (n = 1). Single nucleotide polymorphism (SNP)-based phylogenetic analysis revealed that global S. Welikade strains can be divided into four groups, and these three Chinese isolates were assigned to B (n = 2; Sal097 and XXB1016) and C (n = 1; XXB700). In Group B, the two Chinese ST5123 isolates were closely clustered with three UK ST5123 isolates. In Group C, the Chinese isolate was closely related to the other 12 ST3774 isolates. The number of virulence genes in the S. Welikade isolates ranged from 59 to 152. The galF gene was only present in Group A, the pipB2 gene was only absent from Group A, the avrA gene was only absent from Group B, and the allB, sseK1, sspH2, STM0287, and tlde1 were found only within Group C and D isolates. There were 15 loci unique to the Sal097 isolate. This study is the first to characterize and investigate clinical S. Welikade isolates in China. Responsible for a pediatric case of gastroenteritis in 2021, the clinical isolate harbored no antimicrobial resistance and belonged to phylogenetic Group B of global S. Welikade genomes.

A green and practical electrooxidation protocol for C3 acyloxylation of 2H-indazoles with carboxylic acids in the presence of potassium carbonate has been developed by intermolecular ...cross-dehydrogenative C(sp2)–O coupling. It is sustainable without the use of transition-metal catalysts, stoichiometric oxidants, and even extra supporting electrolytes. Potassium carbonate plays a key role in the reaction, and reacts with carboxylic acids to form carboxylates. Mechanistic studies by cyclic voltammetry and control experiments reveal that the reaction proceeds via an ECE mechanism. In addition, various carboxylic acids and 2H-indazoles are suitable in this system and provide a series of novel desired products at room temperature in an undivided cell.

The monitoring of reactive oxygen species (ROS) in biological system has been occupying research hotspots recently, since the biochemical alterations caused by the overproduction of ROS are the main ...incentives of diseases and accelerated aging process. In this work, we proposed an effective and simple strategy for the construction of high-performance ROS electrochemical sensor. Noble metal alloy nanoparticles (AuPt nanoparticles) with high catalytic activity were spontaneously coated on the freestanding metallic molybdenum disulfide (MoS2) paper independent of any auxiliary conditions. Results have found that the abundant defects and electrons distributed on the metallic MoS2 paper could provide active sites for the nucleation and growth of noble metal nanoparticles. Besides, the excellent mechanical properties of the MoS2 paper promote the formation of flexible sensors. The fabricated MoS2 paper-based sensor was demonstrated to detect ROS with the advantages of wide linear range, prominent selectivity and flexibility, satisfactory detection stability, as well as simple and convenient preparation process. Furthermore, the desirable results obtained in the real sample experiments operated in plant extract pave the way for further real-time monitoring of plant physiological status to provide valuable information for guidance during plant growth.

The CRISPR/Cas system is known as one of the directions of the next generation of mainstream molecular diagnostic technology. However, most current CRISPR/Cas molecular diagnostics still rely on the ...pre-amplification of nucleic acid due to the limited sensitivity of CRISPR/Cas alone, which has no significant advantage over commercial Taqman-PCR and TwistAmp® Exo kits. Herein, we report an aM-level sensitive cascade CRISPR-Dx system (ASCas) that eliminates nucleic acid pre-amplification, thus avoiding aerosol contamination and greatly reducing the testing environment and personnel skill requirements for molecular diagnostics. Most importantly, the Cas13a nucleases with high sensitivity and trans-cleavage efficiency can rapidly cleaved RNA bubbles on the hybridized cascade probe at low concentration target RNA detection, which results in the destruction of the cascade probe and releases a large amount of trigger DNA for further signal amplification of secondary Cas12a reactions. Therefore, the ASCas system achieves amplification-free, ultra-sensitivity (1 aM), and ultra-fast (20 min) RNA detection. In addition, the ASCas system replaces the complicated screening process of primers and probes with the programmed Cas13a-crRNA design so that a suitable detection system can be constructed more quickly and straightforwardly for the mutation-prone SARS-CoV-2 virus.
•A novel cascade CRISPR-Dx system achieves aM-level sensitivity and detects SARS-CoV-2 standard within 20 min.•The cascade probe superimposes the Cas12a and Cas13a reactions, enabling detection without nucleic acid amplification.•This system reduces specialized equipment requirements and is suitable for developing countries.•The ASCas system is suitable for detecting other RNA targets and has potential applications in cancer diagnosis.

Triticum aestivum
L.) was used as the model species to evaluate the ecotoxicological effects of polluted soil in pot experiments. Results showed that the content of heavy metals and PAHs varied greatly in soil samples, among which the heavy metal pollution at S1, S2 and S3 was the most serious. The health risk assessment of PAHs indicated that non-carcinogenic and carcinogenic values for all soil samples were below the threshold levels. Statistical analysis of the correlation between contaminated soil and toxic effects in wheat found that the significance values of regression equations were all less than 0.05 for chlorophyll content, peroxidase (POD) and amylase (AMS) activity. This indicates that the chlorophyll content, POD and AMS activity in wheat leaves could be suitable biomarkers for evaluation of the combined toxicity of multiple pollutants. This study provides a reference for future research on the risk assessment of soil containing multiple pollutants from industrial chemical parks.
