The maize smut fungus Ustilago maydis is a model organism for elucidating host colonization strategies of biotrophic fungi. Here, we performed an in depth transcriptional profiling of the entire ...plant-associated development of U. maydis wild-type strains. In our analysis, we focused on fungal metabolism, nutritional strategies, secreted effectors, and regulatory networks. Secreted proteins were enriched in three distinct expression modules corresponding to stages on the plant surface, establishment of biotrophy, and induction of tumors. These modules are likely the key determinants for U. maydis virulence. With respect to nutrient utilization, we observed that expression of several nutrient transporters was tied to these virulence modules rather than being controlled by nutrient availability. We show that oligopeptide transporters likely involved in nitrogen assimilation are important virulence factors. By measuring the intramodular connectivity of transcription factors, we identified the potential drivers for the virulence modules. While known components of the b-mating type cascade emerged as inducers for the plant surface and biotrophy module, we identified a set of yet uncharacterized transcription factors as likely responsible for expression of the tumor module. We demonstrate a crucial role for leaf tumor formation and effector gene expression for one of these transcription factors.

Plant proteases are key regulators of plant cell processes such as seed development, immune responses, senescence and programmed cell death (PCD). Apoplastic papain-like cysteine proteases (PL) are ...hubs in plant-microbe interactions and play an important role during abiotic stresses. The apoplast is a crucial interface for the interaction between plant and microbes. So far, apoplastic maize PL and their function have been mostly described for aerial parts. In this study, we focused on apoplastic PLCPs in the roots of maize plants. We have analyzed the phylogeny of maize PLCPs and investigated their protein abundance after salicylic acid (SA) treatment. Using activity-based protein profiling (ABPP) we have identified a novel root-specific PLCP belonging to the RD21-like subfamily, as well as three SA activated PLCPs. The root specific PLCP CP1C shares sequence and structural similarities to known CP1-like proteases. Biochemical analysis of recombinant CP1C revealed different substrate specificities and inhibitor affinities compared to the related proteases. This study characterized a root-specific PLCP and identifies differences between the SA-dependent activation of PLCPs in roots and leaves.
Paleomagnetic records of middle Neoproterozoic (820 to 780 Ma) rocks display high amplitude directional variations that lead to large discrepancies in paleogeographic reconstructions. Hypotheses to ...explain these data include rapid true polar wander (TPW), a geomagnetic field geometry that deviates from a predominantly axial dipole field, a hyper‐reversing field (>10 reversals/Ma), and/or undiagnosed remagnetization. To test these hypotheses, we collected 1,057 oriented cores over a 85 m stratigraphic succession in the Laoshanya Formation (Yangjiaping, Hunan, China). High precision U‐Pb dating of two intercalated tuff layers constrain the age of the sediments between 809 and 804 Ma. Thermal demagnetization isolates three magnetization components residing in hematite which are not time‐progressive but conflated throughout the section. All samples possess a north and downward directed component in geographic coordinates at temperatures up to 660°C that is ascribed to a Cretaceous overprint. Two components isolated above 660°C reveal distinct directional clusters: one is interpreted as a depositional remanence, while the other appears to be the result of a mid‐Paleozoic (460 to 420 Ma) remagnetization, which is likely widespread throughout South China. The high‐temperature directions are subtly dependent on lithology; microscopic and rock magnetic analyses identify multiple generations of hematite that vary in concentration and distinguish the magnetization components. A comparison with other middle Neoproterozoic paleomagnetic studies in the region indicates that the sudden changes in paleomagnetic directions, used elsewhere to support the rapid TPW hypothesis (ca. 805 Ma), are better explained by mixtures of primary and remagnetized components, and/or vertical axis rotations.
Plain Language Summary
Paleomagnetic directions recorded in 820 to 780 million year old rocks from South China exhibit large amplitude changes that vary rapidly, which have been interpreted to indicate extraordinarily fast motion of Earth's crust and mantle, up to 90° within a 5 million year span, with respect to the spin axis of the core. This hypothetical phenomenon, called rapid true polar wander (TPW), could be responsible for dramatic global environmental change at that time. To test this theory, we collected over 1,000 samples from a well exposed section where the incongruous directions are found. Our measurements suggest that some of the rocks acquired a new magnetic signal during a pervasive remagnetization event in South China around 440 million years ago, long after original deposition of the rocks. New hematite growth has a demagnetization spectrum that partially overlaps or completely obscures the original magnetic signal, which was previously unrecognized. This implies that rapid TPW is likely an artifact of magnetic overprinting in ancient rocks from South China. Our results suggest that South China was in a relatively stable position at high latitudes 809 to 804 million years ago. We find no evidence to support exceptionally fast continental drift or an abnormal geomagnetic field geometry during that time.

Context.
Turbulence dominated by large-amplitude, nonlinear Alfvén-like fluctuations mainly propagating away from the Sun is ubiquitous in high-speed solar wind streams. Recent studies have ...demontrated that slow wind streams may also show strong Alfvénic signatures, especially in the inner heliosphere.
Aims.
The present study focuses on the characterisation of an Alfvénic slow solar wind interval observed by Solar Orbiter between 14 and 18 July 2020 at a heliocentric distance of 0.64 AU.
Methods.
Our analysis is based on plasma moments and magnetic field measurements from the Solar Wind Analyser (SWA) and Magnetometer (MAG) instruments, respectively. We compared the behaviour of different parameters to characterise the stream in terms of the Alfvénic content and magnetic properties. We also performed a spectral analysis to highlight spectral features and waves signature using power spectral density and magnetic helicity spectrograms, respectively. Moreover, we reconstruct the Solar Orbiter magnetic connectivity to the solar sources both via a ballistic and a potential field source surface (PFSS) model.
Results.
The Alfvénic slow wind stream described in this paper resembles, in many respects, a fast wind stream. Indeed, at large scales, the time series of the speed profile shows a compression region, a main portion of the stream, and a rarefaction region, characterised by different features. Moreover, before the rarefaction region, we pinpoint several structures at different scales recalling the spaghetti-like flux-tube texture of the interplanetary magnetic field. Finally, we identify the connections between Solar Orbiter in situ measurements, tracing them down to coronal streamer and pseudostreamer configurations.
Conclusions.
The characterisation of the Alfvénic slow wind stream observed by Solar Orbiter and the identification of its solar source are extremely important aspects for improving the understanding of future observations of the same solar wind regime, especially as solar activity is increasing toward a maximum, where a higher incidence of this solar wind regime is expected.

The investigation of multi-spacecraft coordinated observations during the cruise phase of BepiColombo (ESA/JAXA) are reported, with a particular emphasis on the recently launched missions, Solar ...Orbiter (ESA/NASA) and Parker Solar Probe (NASA). Despite some payload constraints, many instruments onboard BepiColombo are operating during its cruise phase simultaneously covering a wide range of heliocentric distances (0.28 AU–0.5 AU). Hence, the various spacecraft configurations and the combined
in-situ
and remote sensing measurements from the different spacecraft, offer unique opportunities for BepiColombo to be part of these unprecedented multipoint synergistic observations and for potential scientific studies in the inner heliosphere, even before its orbit insertion around Mercury in December 2025. The main goal of this report is to present the coordinated observation opportunities during the cruise phase of BepiColombo (excluding the planetary flybys). We summarize the identified science topics, the operational instruments, the method we have used to identify the windows of opportunity and discuss the planning of joint observations in the future.
An adult red-legged seriema (Cariama cristata) presented with a comminuted fracture of the tibiotarsus and fibula. Surgery was performed, and a type II external fixator, with 2 distal and 2 proximal ...pins, was used to stabilize the fracture. After a 10-day stabilization period, the bird developed a second fracture on the same bone, proximal to the first fracture site. Another surgery was performed on the seriema similar to the first one. However, in this second surgical procedure a single pin, instead of 2 perpendicular pins, was placed proximally to the fracture site. After the second surgical procedure, bone marrow stem cells (BMSCs) from the seriema's left ulna were collected. Twenty-seven days after the second surgery, the BMSCs were transplanted, into the fracture sites. Twenty-four days after the stem cells were injected into the fractures (51 days after the second surgical procedure), radiographic images revealed healing bone calluses at the fracture sites. The fracture healing was relatively long for this case (a total of 75 days). The addition of bone marrow stem cell therapy to the use of external fixation may have contributed to the healing observed radiographically 24 days after administration; therefore, bone marrow stem cell therapy, in addition to traditional surgical fracture reduction and stabilization, may be a promising therapeutic approach for avian cases with similar injuries and bone anatomy. However, as this is a single case, this therapeutic modality deserves further application and study. Moreover, we suggest modifications in the bone marrow stem cell collection and therapy, which may be useful for future studies and application involving birds.

Diagnostic accuracy of needle-based optical coherence tomography (OCT) for prostate cancer detection by visual and quantitative analysis is defined. 106 three-dimensional (3-D)-OCT data sets were ...acquired in 20 prostates after radical prostatectomy and precisely matched with pathology. OCT images were grouped per histological category. Two reviewers performed blind assessments of the OCT images. Sensitivity and specificity for malignancy detection were calculated. Quantitative analyses by automated optical attenuation coefficient calculation were performed. OCT can reliably differentiate between fat, cystic, and regular atrophy and benign glands. The overall sensitivity and specificity for malignancy detection was 79% and 88% for reviewer 1 and 88% and 81% for reviewer 2. Quantitative analysis for differentiation between stroma and malignancy showed a significant difference (4.6 mm − 1 versus 5.0 mm − 1 Mann-Whitney U-test p < 0.0001). A Kruskal-Wallis test showed a significant difference in median attenuation coefficient between stroma, inflammation, Gleason 3, and Gleason 4 (4.6, 4.1, 5.9, and 5.0 mm − 1, respectively). However, attenuation coefficient varied per patient and a related-samples Wilcoxon signed-rank test showed no significant difference per patient (p = 0.17). This study confirmed the one to one correlation of histopathology and OCT. Precise matching showed that most histological tissues categories in the prostate could be distinguished by their unique pattern in OCT images. In addition, the optical attenuation coefficient can play a role in the differentiation between stroma and malignancy; however, a per patient analysis of the optical attenuation coefficient did not show a significant difference.
Background and aims
Microsatellite instability (MSI) is a potential indicator of prognosis in patients with colorectal cancer (CRC). To date, there are a limited number of studies which investigated ...its role in advanced CRC. Our study investigated the value of high degree of MSI (MSI-H) in patients treated with 5-FU/oxaliplatin-based chemotherapy which has been done by only one further study recently.
Patients and methods
In this study, we investigated tumour tissues from 108 patients with metastatic CRC who were treated in a prospective, randomised trial comparing two oxaliplatin and 5-FU-based therapy regimens (FUFOX vs. CAPOX) involving a total of 474 patients. We determined the incidence and prognostic value of a high degree of microsatellite instability. The specimens were analysed by PCR corresponding to the National Institute of Health reference panel. In addition, immunostaining of the mismatch repair proteins MLH1, MSH2 and MSH6 was performed.
Results and findings
The incidence of MSI-H was 4%. MSI-H was correlated with a lower rate of disease control compared to non-MSI-H patients (
p
= 0.02). However, there was no correlation between MSI-H and progression-free survival or overall survival.
Interpretation and conclusion
MSI-H incidence in metastatic CRC was low. Our data suggest that MSI-H may be correlated with a poorer response to a 5-FU/oxaliplatin treatment. This finding needs confirmation in a larger cohort.

Knowledge workers experience many interruptions during their work day. Especially when they happen at inopportune moments, interruptions can incur high costs, cause time loss and frustration. Knowing ...a person's interruptibility allows optimizing the timing of interruptions and minimize disruption. Recent advances in technology provide the opportunity to collect a wide variety of data on knowledge workers to predict interruptibility. While prior work predominantly examined interruptibility based on a single data type and in short lab studies, we conducted a two-week field study with 13 professional software developers to investigate a variety of computer interaction, heart-, sleep-, and physical activity-related data. Our analysis shows that computer interaction data is more accurate in predicting interruptibility at the computer than biometric data (74.8% vs. 68.3% accuracy), and that combining both yields the best results (75.7% accuracy). We discuss our findings and their practical applicability also in light of collected qualitative data.
