A polyclonal antiserum against cysteine synthase (CSase, EC 4.2.99.8) isozymes (CSases 1, 2, and 3) was prepared for physiological study of the isozymes. From the results of immunotitration and ...double immunodiffusion analyses, the polyclonal antiserum was proved to have the same degree of reactivity toward each isozyme. The accumulation of the isozymes during germination of spinach seeds was investigated by Western blot analysis. Although CSase 1 was not detected in the cotyledons of dark-grown seedlings, it was found in the green cotyledons of seedlings that had been grown in the light. In the extract of roots, CSase 1 was not detected in any developmental stage. On the other hand, CSases 2 and 3 were detected in all developmental stages and tissues. These results indicate that the expression of CSase 1 is regulated by light, and that the regulatory mechanisms of Csase 1 expression differs from those of CSase 2 and 3 expression. Western blot analysis showed that the hydrated seeds contained a new Csase band, which was different from known CSases in terms of electrophoretic mobility
The entire distribution of lymphatics in whole mount preparations of the Japanese monkey was studied using the enzyme-histochemical technique reported by KATO et al. (1990, 1991). In this staining, ...the lymphatic endothelium was colored dark brown by its positive 5′-nucleotidase activity, while most blood vessels (especially arterioles) were colored blue due to their positive alkaline phosphatase reaction. The whole mount preparations of the pleura treated enzyme-histochemically clearly indicated the distribution, branching patterns and running courses of lymphatic vessels. They revealed numerous short blind-ending knobs which represented the initial portions of lymphatics. These knobs were seen near the surface of the parietal pleura along its entire extent. In the costal and diaphragmatic pleura, the lymphatics ran parallel to the intercostal muscle fibers, but perpendicular to the tendinous and muscular fibers of the diaphragm; they formed ladders, independent of the courses of blood vessels. In the mediastinal pleura, lymphatic vessels showed a tree-like branching accompanying blood vessels. Under the light microscope, toluidine-blue stained semithin sections revealed the initial part of lymphatics as a small irregularly outlined cavity (7-10μm in diameter) surrounded by a dense connective tissue. This lymphatic dilation was sometimes located close to a thin mesothelial layer. Such a structure suggesting a “stoma” was seen near the attachment of the muscular diaphragm to the sternum and along the borders of the ribs. Transmission electron microscopy revealed an occasional interruption in the mesothelium. This stoma continued to a submesothelial cavity whose base comprised an attenuated endothelium of an extended lymphatic vessel.
BAY o 9867 (Ciprofloxacin), a new pyridonecarboxylic acid derivative, was investigated on its serum level, urinary excretion, bile and exudate concentration, clinical effectiveness and side effects. ...Serum level and urinary excretion were investigated in 3 adult volunteers before and after meal by cross over method. 1. Following oral administration of 200mg of BAY o 9867 before meal, the serum level showed its peak of mean 1.24μg/ml after 1.5 hours, and after meal its peak of mean 1.0μg/ml after same hours. Urinary excretion rate until 12 hours was 47.6% before meal and 34.7% after meal. 2. Following oral administration of 200mg of this drug in 2 patients with biliary tract drainage, the bile concentration in one patient, reached its peak of 5.5μg/ml at 4-5 hours and in another patient 1.9μg/ml at 3-4 hours after administration. 3. Following oral administration of same dosage in 5 patients after radical mastectomy, the exudate concentration was showed after 3 hours, and reached mean 0.5μg/ml after 6 hours. 4. BAY o 9867 600mg/day was given to 63 patients with infections in the field of surgery.The results obtained were as follows; “excellent” in 13 cases, “good” in 36, “fair” in 12 and “poor” in 2, and effectiveness rate 77.8%. 5. Side effects related BAY o 9867 were observed in 2 cases, such as one case noted epigastralgia and the other diarrhea, but they were mild. As abnormal laboratory data, 1 case with eosinophilia and 1 case with increase of GOT, GPT value were noted.
