To help doctors and patients evaluate lumbar intervertebral disc degeneration (IVDD) accurately and efficiently, we propose a segmentation network and a quantitation method for IVDD from T2MRI. A ...semantic segmentation network (BianqueNet) composed of three innovative modules achieves high-precision segmentation of IVDD-related regions. A quantitative method is used to calculate the signal intensity and geometric features of IVDD. Manual measurements have excellent agreement with automatic calculations, but the latter have better repeatability and efficiency. We investigate the relationship between IVDD parameters and demographic information (age, gender, position and IVDD grade) in a large population. Considering these parameters present strong correlation with IVDD grade, we establish a quantitative criterion for IVDD. This fully automated quantitation system for IVDD may provide more precise information for clinical practice, clinical trials, and mechanism investigation. It also would increase the number of patients that can be monitored.
Phase engineering of nanomaterials (PEN) offers a promising route to rationally tune the physicochemical properties of nanomaterials and further enhance their performance in various applications. ...However, it remains a great challenge to construct well‐defined crystalline@amorphous core–shell heterostructured nanomaterials with the same chemical components. Herein, the synthesis of binary (Pd‐P) crystalline@amorphous heterostructured nanoplates using Cu3−χP nanoplates as templates, via cation exchange, is reported. The obtained nanoplate possesses a crystalline core and an amorphous shell with the same elemental components, referred to as c‐Pd‐P@a‐Pd‐P. Moreover, the obtained c‐Pd‐P@a‐Pd‐P nanoplates can serve as templates to be further alloyed with Ni, forming ternary (Pd‐Ni‐P) crystalline@amorphous heterostructured nanoplates, referred to as c‐Pd‐Ni‐P@a‐Pd‐Ni‐P. The atomic content of Ni in the c‐Pd‐Ni‐P@a‐Pd‐Ni‐P nanoplates can be tuned in the range from 9.47 to 38.61 at%. When used as a catalyst, the c‐Pd‐Ni‐P@a‐Pd‐Ni‐P nanoplates with 9.47 at% Ni exhibit excellent electrocatalytic activity toward ethanol oxidation, showing a high mass current density up to 3.05 A mgPd−1, which is 4.5 times that of the commercial Pd/C catalyst (0.68 A mgPd−1).
Binary (Pd‐P) and ternary (Pd‐Ni‐P) nanoplates, both with crystalline@amorphous core–shell nanostructures, are synthesized using Cu3−χP nanoplates as templates. The obtained c‐Pd‐Ni‐P@a‐Pd‐Ni‐P heterostructured nanoplates exhibit superior electrocatalytic performance toward the ethanol oxidation reaction in alkaline media compared to c‐Pd‐P@a‐Pd‐P heterostructured nanoplates and commercial Pd/C catalysts.

Metal–organic framework (MOF) heterostructures have shown promising applications in gas adsorption, gas separation, catalysis, and energy, arising from the synergistic effect of each component. ...However, owing to the difficulty in controlling the size, shape, nucleation, and growth of MOFs, it remains a great challenge to construct MOF heterostructures with precisely controlled orientation, morphology, dimensionality, and spatial distribution of each component. Here, we report a seeded epitaxial growth method to prepare a series of hierarchical MOF heterostructures by engineering the structures, sizes, dimensionalities, morphologies, and lattice parameters of both MOF seeds and the secondary MOFs. In these heterostructures, PCN-222 (also known as MOF-545) nanorods selectively grow along the major axis of the ellipsoid-like PCN-608 nanoparticles, on the two end facets of the hexagonal prism-like NU-1000 nanorods, and on the two basal planes of the hexagonal PCN-134 nanoplates, while Zr–BTB nanosheets selectively grow on the six edge facets of PCN-134 nanoplates. The selective epitaxial growth of MOFs opens the way to synthesize different hierarchical heterostructures with tunable architectures and dimensionalities, which could process various promising applications.

The BA.1, BA.2, and BA.3 omicron subvariants of SARS-CoV-2 showed similar but substantial resistance to vaccine-induced and infection-induced serum neutralising activity.1,2 The new BA.2.12.1, ...BA.2.13, BA.4, and BA.5 omicron subvariants containing Leu452 substitutions show more infectious potential than BA.2.3 We examined neutralising activity against the BA.1, BA.2, BA.2.11, BA.2.12.1, BA.2.13, BA.4, and BA.5 omicron subvariants in serum from people who received BBIBP-CorV (Sinopharm) primary immunisation, people who received BBIBP-CorV or ZF2001 (Anhui Zhifei Longcom) boosters, and people with omicron breakthrough infections (appendix pp 4, 7). Using an in-house pseudovirus neutralisation assay we found that two BBIBP-CorV doses induced detectable neutralising antibodies against spike protein mutation D614G in 21 (84%) individuals, but neutralising activity against omicron subvariants (BA.1, BA.2, BA.2.11, BA.2.12.1, BA.2.13, and BA.4/BA.5) was not or only minimally detectable (appendix pp 2–3, 8). Neutralising activity against omicron subvariants was observed in 24–48% of people who received a BBIBP-CorV booster and 30–53% of people who received a ZF2001 booster (appendix pp 2–3, 9). ...serum samples with neutralising antibody titres higher than the limit of detection (limit of detection was 30) against the omicron subvariants had lower neutralising activity, with a 4·6–17·1-times lower GMT than the GMT against D614G (appendix pp 2–3).

To fully decipher the immunogenicity of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Spike protein, it is essential to assess which part is highly immunogenic in a systematic way. ...We generate a linear epitope landscape of the Spike protein by analyzing the serum immunoglobulin G (IgG) response of 1,051 coronavirus disease 2019 (COVID-19) patients with a peptide microarray. We reveal two regions rich in linear epitopes, i.e., C-terminal domain (CTD) and a region close to the S2′ cleavage site and fusion peptide. Unexpectedly, we find that the receptor binding domain (RBD) lacks linear epitope. We reveal that the number of responsive peptides is highly variable among patients and correlates with disease severity. Some peptides are moderately associated with severity and clinical outcome. By immunizing mice, we obtain linear-epitope-specific antibodies; however, no significant neutralizing activity against the authentic virus is observed for these antibodies. This landscape will facilitate our understanding of SARS-CoV-2-specific humoral responses and might be useful for vaccine refinement.

Hepatitis B virus (HBV) infection is the major public health problem worldwide. In clinical practice, serological and molecular assays are the most commonly used diagnostic methods to detect HBV ...infection in clinical practices.
Here we present a rapid and sensitive recombinase aided amplification assay (RAA) to detect HBV at 39.0 °C for 30 min without DNA extraction from serum samples. The analytical sensitivity of RAA assay was 100 copies per reaction and showed no cross reaction with human immunodeficiency virus (HIV) and hepatitis C virus (HCV). The universality of RAA assay was validated by testing of 41 archived serum samples with predefined HBV genotypes (B, C and D).
A total of 130 archived suspected HBV infected serum samples were detected by commercial qPCR with DNA extraction and RAA assay without DNA extraction (heat-treatment). Compared with qPCR assay as a reference, the RAA assay obtained 95.7% sensitivity and 100% specificity and a kappa value of 0.818.
We developed a rapid, convenient, highly sensitive and specific method to detect HBV without DNA extraction in clinical samples. This RAA method of HBV detection is very suitable for clinical testing.

Microsatellite instability (MSI) has been approved as a pan-cancer biomarker for immune checkpoint blockade (ICB) therapy. However, current MSI identification methods are not available for all ...patients. We proposed an ensemble multiple instance deep learning model to predict microsatellite status based on histopathology images, and interpreted the pathomics-based model with multi-omics correlation. Methods: Two cohorts of patients were collected, including 429 from The Cancer Genome Atlas (TCGA-COAD) and 785 from an Asian colorectal cancer (CRC) cohort (Asian-CRC). We established the pathomics model, named Ensembled Patch Likelihood Aggregation (EPLA), based on two consecutive stages: patch-level prediction and WSI-level prediction. The initial model was developed and validated in TCGA-COAD, and then generalized in Asian-CRC through transfer learning. The pathological signatures extracted from the model were analyzed with genomic and transcriptomic profiles for model interpretation. Results: The EPLA model achieved an area-under-the-curve (AUC) of 0.8848 (95% CI: 0.8185-0.9512) in the TCGA-COAD test set and an AUC of 0.8504 (95% CI: 0.7591-0.9323) in the external validation set Asian-CRC after transfer learning. Notably, EPLA captured the relationship between pathological phenotype of poor differentiation and MSI (P < 0.001). Furthermore, the five pathological imaging signatures identified from the EPLA model were associated with mutation burden and DNA damage repair related genotype in the genomic profiles, and antitumor immunity activated pathway in the transcriptomic profiles. Conclusions: Our pathomics-based deep learning model can effectively predict MSI from histopathology images and is transferable to a new patient cohort. The interpretability of our model by association with pathological, genomic and transcriptomic phenotypes lays the foundation for prospective clinical trials of the application of this artificial intelligence (AI) platform in ICB therapy.
The incorporation of metal–organic frameworks (MOFs) into membrane‐shaped architectures is of great importance for practical applications. The currently synthesized MOF‐based membranes show many ...disadvantages, such as poor compatibility, low dispersity, and instability, which severely limit their utility. Herein, we present a general, facile, and robust approach for the synthesis of MOF‐based composite membranes through the in situ growth of MOF plates in the channels of anodized aluminum oxide (AAO) membranes. After being used as catalysis reactors, they exhibit high catalytic performance and stability in the Knoevenagel condensation reaction. The high catalytic performance might be attributed to the intrinsic structure of MOF‐based composite membranes, which can remove the products from the reaction zone quickly, and prevent the aggregation and loss of catalysts during reaction and recycling process.

Omicron subvariants BQ.1 and BQ.1.1 with increased resistance to neutralising antibodies can pose a challenge to immunity induced by vaccination or infection and render therapeutic monoclonal antibodies ineffective.3–6 Our results suggest that BQ.1 and BQ.1.1 extensively, but incompletely, escape omicron subvariant breakthrough infection neutralisation, including the most recent BA.5.1.2, BA.2.76, and BF.7 infections.
